RHOBTB3是一个多功能蛋白,它能够参与Cul3/Rocl介导的cyclin E泛素化-蛋白酶体降解或剪切加工,促进细胞从S期向G2期的转化,从而保证了细胞周期的有序进行。另一方面,RHOBTB3的C末端异戊烯基化之后参与某些种类的囊泡运输。我们实验室还发现它在低氧信号通路中具有重要的作用。本文展示了利用酵母双杂交的方法寻找RHOBTB3相互作用蛋白。其中我们锁定了 cox11进行深入研究。cox11是一个线粒体内膜蛋白,是Complex Ⅳ的成员,它的存在对于Complex Ⅳ的组装至关重要。我们发现cox11有三种形式的蛋白存在:它的L-form主要定位在高尔基上,可以与RHOBTB3相互作用;M-form和S-form主要定位在线粒体上,无法与RHOBTB3相互作用。在葡萄糖饥饿刺激的条件下,cox11的L-form被降解或剪切加工,这种现象可以被MG132所抑制。此外,cox11还可能参与调控了 RHOBTB3缺失所引起的细胞ATP水平下降,或葡萄糖饥饿刺激条件下,线粒体形态的改变。另外,RHOBTB3也许参与了 cox11 L-form降解或剪切加工这一过程。对这些问题的深入研究有助于我们进一步了解RHOBTB3和cox11的功能,以及细胞应对葡萄糖饥饿的机制。

Toxoplasma gondii is the caustive agent of toxoplasmosis and can infect almost any warm-blooded animals, including human. Rhoptry protein 9(ROP9) is a 36 kDa soluble protein present only in the tachyzoite stage during *** life cycle and participate in the early stages of parasite invasion. Until up to now, host proteins interacting with ROP9 still have not been found. In our study, a yeast two-hybrid system was used to screen host proteins interacting with ROP9. ROP9 was inserted into the pGBKT7 vector as a bait which expressed fusion protein with the GAL4 DNA-binding domain. Bait proteins expression detection was carried on via western blotting and the autoactivation of bait plasmid was tested compared with the empty vector pGBKT7. A human brain c DNA library was used to identified host proteins interacting with ROP9 by yeast-two-hybrid system. After screening, positive hits were sequenced and analysed with BLAST analysis. We obtained 5 host protein sequences and four of which are false positives. The interaction between ROP9 and cytochrome coxidase11(cox11) needs to be further confirmed. Our study firstly report interactions between Toxoplasma gondii ROP9 and host proteins. The results will facilitate the understanding of the function of ROP9 and host signal tranduction ROP9 involved.
