Heteromeric.and Homomeric.Geranyl Diphosphate Synthases from c.tharanthus roseus and Their Role in Monoterpene Indole Alkaloid Biosynthesis

c.tharanthus roseus is the sole sourc. of two most important monoterpene indole alkaloid （MIA） anti- c.nc.r agents： vinblastine and vinc.istine. MIAs possess a terpene and an indole moiety derived from terpenoid and shikimate pathways, respec.ively. Geranyl diphosphate （GPP）, the entry point to the formation of terpene moiety, is a produc. of the c.ndensation of isopentenyl diphosphate （IPP） and dimethylallyl diphosphate （DMAPP） by GPP synthase （GPPS）. Here, we report three genes enc.ding proteins with sequenc. similarity to large subunit （***） and small subunit （***） of heteromeric.GPPSs, and a homomeric.GPPSs. *** is a bifunc.ional enzyme produc.ng both GPP and geranyl geranyl diphosphate （GGPP）, *** is inac.ive, whereas c.GPPS is a homomeric.enzyme forming GPP. c.-expression of both subunits in Esc.eric.ia c.il resulted in heteromeric.enzyme with enhanc.d ac.ivity produc.ng only GPR While *** and c.GPPS showed higher expression in older and younger leaves, respec.ively, *** showed an inc.easing trend and dec.eased gradually. Methyl jasmonate （MelA） treatment of leaves sig- nific.ntly induc.d the expression of only ***. GFP loc.lization indic.ted that *** is plastidial whereas c.GPPS is mitoc.ondrial. Transient overexpression of *** in c. roseus leaves resulted in inc.eased vindoline, immediate monomeric.prec.rsor of vinblastine and vinc.istine. Although c. roseus has both heteromeric.and homomeric.GPPS enzymes, our results implic.te the involvement of only heteromeric.GPPS with *** regulating the GPP flux for MIA biosynthesis.
