Composite Sophora Colon-Soluble Capsule Ameliorates DSS-Induced Ulcerative Colitis in Mice via Gut Microbiota-Derived butyric acid and NCR+ ILC3

Objective: To investigate the effects of composite Sophora colon-soluble Capsule(CSCC) on gut microbiota-mediated short-chain fatty acids(SCFAs) production and downstream group 3 innate lymphoid cells(ILC3s) of dextran sulfate sodium(DSS)-induced ulcerative colitis(UC) mice model. Methods: The main components of CSCC were analyzed by hybrid ultra-high-performance liquid chromatography ion mobility spectromety quadrupole time-of-flight mass spectrometry(UHPLC-IM-QTOF/MS). Twenty-four male BALB/c mice were randomly divided into 4 groups(n=6) by using a computer algorithm-generated random digital, including control, DSS model, mesalazine, and CSCC groups. A DSS-induced colitis mice model was established to determine the effects of CSCC by recording colonic weight, colonic length, index of colonic weight, and histological colonic score. The variations in ILC3s were assessed by immunofluorescence and flow cytometry. The results of gut microbiota and SCFAs were acquired by 16s r DNA and gas chromatography-mass spectrometry(GC-MS) analysis. The expression levels of NCR^(+)ILC3^(-), CCR6^(+)Nkp46-(Lti) ILC3^(-), and ILCreg-specific markers were detected by enzyme-linked immunosorbent assay, and real-time quantitative polymerase chain reaction and Western blot, respectively. Results: The main components of CSCC were matrine, ammothamnine, Sophora flavescens neoalcohol J, and Sophora oxytol U. After 7 days of treatment, CSCC significantly alleviated colitis by promoting the reproduction of intestinal probiotics manifested as upregulation of the abundance of Bacteroidetes species and specifically the Bacteroidales_S24-7 genus(Pbutyric acid increased the most after CSCC treatment. Meanwhile, compared with the model group, Lti ILC3s and its biomarkers were significantly downregulated and NCR^(+) ILC3s were significantly elevated in the CSCC group(P<0.01). Further experiments revealed that ILC3s were differentiated from Lti ILC3s to NCR^(+)

Efficient Separation of butyric acid by an Aqueous Two-phase System with Calcium Chloride

One of the bottlenecks for bioproduction of butyric acid as bulk chemical is the difficulty in separating butyric acid from the fermentation broth,compared with the petroleum-based chemical synthesis *** the present work,a novel separation methodology was developed based on an aqueous two-phase system with inor-ganic *** chloride was screened out for effective separation of butyric acid from butyric acid-water-salt *** appropriate concentration range of butyric acid and salt,butyric acid was enriched in the upper phase and most of calcium ions remained in the lower ***"salting out"effect is very efficient to separate butyric acid from the simulated butyrate fermentation broth,which consists of butyric acid and acetic acid with concentration ratio of 4︰1,so that the final ratio of butyric acid/acetic acid in the upper phase is improved to 9.87. The aqueous two-phase system was used to separate butyric acid from the actual fermentation broth with satisfac-tory result.

The Near Surface Morphology of P3HT：PCBM Blend Films Studied by Near-Edge X-Ray Absorption Fine Structure

The microstructure of poly（3-hexylthiophene）（P3HT） and [6,6]-phenyl-C61 butyric acid methyl ester （PCBM） blends with various annealing temperatures are investigated in detail by the near-edge x-ray absorption fine structure. Under higher anneMing temperature （Tanneal ≥ 160℃）, P3HT shows an unusual fluidity and aggregation in the surface. After annealing, the enrichment polymer component recrystallizes and forms a single P3HT phase layer in the surface of blend films. Moreover, it gives direct evidence of the PCBM content diffusing to the near surface of blend films during annealing treatment. These findings are beneficial to improving the morphology of polymer/fullerene blend films.

In the past two decades, organic solar cells （OSCs） have at- tracted significant attention owing to their merits of achieving light-weight, flexible, low-cost devices [1,2]. Ternary OSC as an efficient way to improve the power conversion efficiencies （PCEs） of OSCs has inspired great interest from researchers to investigate ternary OSCs and result in many efficient de- vices. Ternary OSCs allow collecting high- and low-energy photons by separated semiconductors in a single active layer to minimize the thermalization loss and broaden absorption range to improve the performance of OSCs. However, most reported ternary OSCs only have active layer of about 100 nm in thickness.

The specificity of ten non-digestible carbohydrates to enhance butyrate-producing bacteria and butyrate production in vitro fermentation

Butyrate and butyrate-producing bacteria are important indicators of gut microbial metabolism in human *** non-digestible carbohydrates(NDCs),including inulin,fructooligosaccharide(FOS),oatsβ-glucans(OGS),oatsβ-glucan oligosaccharides(OGOS),Astragalus polysaccharides(APS),Astragalus oligosaccharides(AOS),xanthan gum oligosaccharides(XGOS),gellan gum oligosaccharides(GGOS),curdlan oligosaccharides(COS),and pullulan oligosaccharides(POS)were used to investigate NDC specifi city in modulating butyrate-producing bacteria and butyrate production in 48 h in vitro fermentation studies in combination with fecal inocula from 7 healthy donors and 11 patients with type 2 diabetes(T2D).We observed that the amount of these ten NDCs utilized depended on NDC structure and inter-individual gut microbial *** and GGOS fermentations signifi cantly increased butyrate-producing bacteria(especially f_Lachnospiraceae)and butyric acid ***,XGOS and GGOS fermentations showed a better ability to consistently modulate gut microbiota composition and metabolic properties between individuals of healthy donors or T2D patients when compared to inulin,FOS,APS,AOS,OGS,OGOS,COS and POS *** research indicated that xanthan gum and gellan gum oligosaccharides have strong specifi city to enhance butyrate-producing bacteria and butyrate production.

Irritable bowel syndrome with diarrhea(IBS-D)is chronic intestinal dysfunction with diarrhea and othercomplicated clinical symptoms,and it has a great impact on the daily life and mental state of *** studies have reported that ingestion of probiotics can significantly alleviate a variety of *** pupose of this study was to investigate the IBS-D-alleviating effects of a probiotic strain,lactobacillus plantarum(***)CCFM8610,with multiple health-promoting *** studywas a 12-week,randomized,double-blind,placebo-controlled,pilot clinical ***-five patientswere randomly assigned to receive the placebo,oligosaccharides,or *** CCFM8610(1×10^(10) colony-forming units(CFU)per day),with a 2-week run-in period,an 8-week intervention period,anda 2-week follow-up observation *** patients'clinical symptoms and quality of life were exam-ined by the IBS symptom severity scale(IBS-SsS)and the IlBS quality of life scale(IBS-QOL).Changes ingut microbiota composition and diversity were measured at the end of the intervention *** oraladministration of *** CCFM8610 significantly decreased the IBS-SSS and IBS-QOL scores,reduced IBS-D symptom severity,recovered gut microbiota diversity,decreased the relative abundanceof bloating-related genus Methanobrevibacter,and increased the relative abundance of butyric acid-producing genera,including Anaerostipes,Anaerotruncus,Bifidobacterium,butyricimonas,*** findings suggest that ingestion of *** CCFM8610 can significantly alleviate clinical symptoms and gut microbiota dysbiosis in IBS-D *** IBS-D-alleviating effect of ***-tarum CCFM8610 may be related to theincrease in the relative abundance of butyric acid-producing genera in the intestine.

Fecal microbiota transplantation for irritable bowel syndrome:An intervention for the 21st century

Irritable bowel syndrome(IBS)affects about 12%of the global *** IBS does not develop into a serious disease or increase mortality,it results in a considerable reduction in the quality of *** etiology of IBS is not known,but the intestinal microbiota appears to play a pivotal role in its *** is no effective treatment for IBS,and so the applied treatments clinically focus on symptom *** microbiota transplantation(FMT),an old Chinese treatment,has been applied to IBS patients in seven randomized controlled trials(RCTs).Positive effects on IBS symptoms in various degrees were obtained in four of these RCTs,while there was no effect in the remaining *** the seven RCTs there were marked differences in the selection processes for the donor and treated patients,the transplant dose,the route of administration,and the methods used to measure how the patients responded to *** present frontier discusses these differences and proposes:(1)criteria for selecting an effective donor(superdonor);(2)selection criteria for patients that are suitable for FMT;(3)the optimal FMT dose;and(4)the route of transplant *** appears to be safe,with only mild,self-limiting side effects of abdominal pain,cramping,tenderness,diarrhea,and *** it is early to speculate about the mechanisms underlying the effects of FMT,the available data suggest that changes in the intestinal bacteria accompanied by changes in fermentation patterns and fermentation products(specifically short-chain fatty acids)play an important role in improving the IBS symptoms seen after *** appears to be a promising treatment for IBS,but further studies are needed before it can be applied in everyday clinical practice.
