Hepatocellular carcinoma(HCC)is one of the leading causes of cancer-related deaths,characterized by highly hypoxic tumor ***-inducible factor-1α(HIF-1α)is a major regulator involved in cellular response to changes of oxygen levels,supporting the adaptation of tumor cells to *** d(Bd)is an isolated natural quassinoid with multiple anti-cancer ***,we identified Bd could significantly inhibit the HIF-1α expression and its subsequently mediated HCC cell *** biophysical proteomics approaches,we identified inhibitor of β-catenin and T-cell factor(ICAT)as the functional target of *** targeting ICAT,Bd disrupted the interaction of bcatenin and ICAT,and promoted β-catenin degradation,which in turn induced the decrease of HIF-1α ***,Bd could inhibit HCC cells proliferation and tumor growth in vivo,and knockdown of ICAT substantially increased resistance to Bd treatment in *** data highlight the potential of Bd as a modulator of β-catenin/HIF-1α axis mediated HCC metabolism.

Background:Non-small cell lung cancer(NSCLC)is considered one of the leading causes of cancer-related *** the availability of drugs for the treatment of NSCLC,the need for the development of novel agents with high efficiency and fewer adverse effects remains *** natural compound bruceine d(Bd)is widely recognized for its notable anti-inflammatory,antiparasitic,and hypoglycemic ***,it is unclear whether Bd can be used as a novel agent for NSCLC *** and Methods:MTT and colony formation assays were used to assess the antiproliferative effect of Bd on NSCLC *** healing and transwell assays were performed to determine the effect of Bd on the migration and invasion of H1299 cells,*** blotting assay was used to detect the expression levels of ***:We demonstrated that Bd significantly inhibited the proliferation of H1299,A549,and H226 cells with respective IC50 values of 6.06±0.52,7.15±0.90,and 7.21±0.75μ*** addition,Bd suppressed colony formation of H1299 cells in a dose-dependent *** treatment with Bd,the migration and invasive capabilities of H1299 cells were significantly inhibited in a dose-and time-dependent ***,the results of Western blotting demonstrated that Bd treatment resulted in the upregulation of the protein expression of E-cadherin and downregulation of the expression of N-cadherin,twist,snail,integrinαv,integrinβ4,matrix metalloproteinase-7,andβ-catenin ***:Bd inhibits proliferation,migration,and invasion of NSCLC cells;therefore,Bd may be considered for its potential in adjuvant therapy for NSCLC.

植物中的一些次生代谢物具有除草生物活性,而这些次生代谢物也是除草剂开发的先导结构来源之一。本研究以鸦胆子（Brucea javanica（L.）Merr.）种子去油后所得废渣为研究对象,系统测定了其甲醇提取物及不同溶剂萃取物的除草活性,并通过活性跟踪对其具有除草活性的化学成分进行了分离,为明确鸦胆子中的除草活性成分及开发利用鸦胆子去油后废渣的功能奠定了研究基础。主要研究结果如下:1.采用培养皿滤纸法,测定了鸦胆子废渣的甲醇提取物、石油醚萃取物、乙酸乙酯萃取物和正丁醇萃取物对牛筋草、三叶鬼针草和刺苋种子的萌发及生长的影响。结果表明:在10 mg/m L浓度下,鸦胆子废渣的甲醇提取物能显著抑制3种杂草的种子萌发及生长,其中对三叶鬼针草的芽长和刺苋的根长的抑制作用最为显著,抑制率均为100.00%;在5 mg/m L浓度下,石油醚萃取物、乙酸乙酯萃取物和正丁醇萃取物对3种杂草的种子萌发及生长的抑制作用由强到弱依次为乙酸乙酯萃取物、正丁醇萃取物、石油醚萃取物,其中乙酸乙酯和正丁醇萃取物对牛筋草的种子萌发及生长的抑制作用最为显著,抑制率均为100.00%。2.采用盆栽试验法,测定了鸦胆子废渣的甲醇提取物、石油醚萃取物、乙酸乙酯萃取物和正丁醇萃取物对牛筋草和三叶鬼针草种子的出苗及生长的影响。结果表明:在10 mg/m L浓度下,鸦胆子废渣的甲醇提取物能抑制牛筋草和三叶鬼针草出苗,其中对牛筋草出苗的抑制作用最为显著;在5 mg/m L浓度下,石油醚萃取物、乙酸乙酯萃取物和正丁醇萃取物对2种杂草的出苗及生长的抑制作用由强到弱依次为正丁醇萃取物、乙酸乙酯萃取物、石油醚萃取物,其中正丁醇萃取物能完全地抑制牛筋草和三叶鬼针草出苗及生长。3.通过硅胶柱层析、凝胶柱层析、中压制备等方法从鸦胆子废渣的正丁醇萃取层中分离出9个化合物,通过核磁共振（NMR）、质谱（MS）图谱信号并结合文献鉴定出化合物结构,分别为bruceine d、E、F,pityriacitrin,4-（9H-?-carbolin-1-yl）-4-oxobut-2-enoic acid methyl ester,protocatechuic acid,vanillic acid,carisphthalate和2,2’-oxybis（1,4-di-tert-butylbenzene）,其中pityriacitrin、4-（9H-?-carbolin-1-yl）-4-oxobut-2-enoic acid methyl ester、carisphthalate和2,2’-oxybis（1,4-di-tert-butylbenzene）是首次从鸦胆子中分离得到。4.通过培养皿滤纸法测定了鸦胆子废渣中苦木内酯类化合物bruceine d、E及F这三个化合物对牛筋草和三叶鬼针草种子萌发及生长的影响。结果显示bruceine d、E和F均具有显著的除草活性。在浓度为250?g/m L时,bruceine d完全抑制了牛筋草的种子萌发及生长和三叶鬼针草的芽伸长,而bruceine E和F完全抑制了三叶鬼针草的芽伸长。此外,不同浓度的bruceine d对2种杂草种子萌发及生长表现出不同程度的抑制作用。bruceine d在实验浓度范围31.25～500?g/m L内,对牛筋草的根长的抑制率均达100.00%;在125～500?g/m L浓度范围内,对牛筋草的种子萌发及芽长和三叶鬼针草的芽长的抑制率均达100.00%。上述结果表明:鸦胆子废渣对杂草的种子萌发及生长具有抑制作用,且从鸦胆子废渣中分离到的bruceine d具有显著的除草活性,苦木内酯类化合物可能是鸦胆子废渣中具有除草活性的主要次生代谢产物。因而,鸦胆子废渣有作为植物源除草剂的潜力,具有进一步研究与开发的价值。
