Methylation patterns in 5' terminal regions of pluripotency-related genes in bovine in vitro fertilized and cloned embryos

In order to investigate DNA methylation profiles of five pluripotency-related genes （Oct4, Sox2, Nanog, Rexl and Fgf4） during bovine maternal to zygotic transition （MZT） in both in vitro fertilized （IVF） and nuclear transfer （NT） embryos, sodium bisulfite sequencing method was used to detect DNA methylation levels, accompanied by the statistical analysis of embryo developmental rates. The results showed that Oct4, Nanog, Rexl and Fgf4 were respectively demethylated by 25.22% （P 〈 0.01）, 3.84% （P 〉 0.05）, 31.82% （P 〈 0.01） and 10% （P 〉 0.05） while Sox2 retained unmethylafion during MZT in IVF embryos. By contrast, Oct4 and Rexl respectively underwent demethylation by 23.04% （P 〈 0.01） and 6.02% （P 〉 0.05）, and, reversely, Sox2, Nanog and Fgf4 respectively experienced remethylation by 0.84% （P 〉 0.05）, 5.39% （P 〉 0.05） and 5.46% （P 〉 0.05） during MZT in NT embryos. Interestingly, the CpG 14 site of Sox2 was specifically methylated in both 8-cell and morula NT embryos. In addition, the development of blastocysts between IVF and NT embryos showed no significant difference. DNA methylation analysis showed that only Oct4 and Sox2 underwent the correct methylation reprogramming process, which may be responsible for the development of blastocysts of NT embryos to a certain extent. In conclusion, the five genes respectively experienced demethylation to different extents and incomplete DNA methylation reprogramming during bovine MZT in both IVF and NT embryos, suggesting that they may be used as indicators for bovine embryo developmental competence.

In this work, the binding interaction between a new bismuth（HI） Schiff base complex{ [Bi（valen）]C1.2H2O} [H2valen=N,N′bis（2-hydroxy-3-methoxyphenylmethylidene）-2,6-pyridinediamine, C2 IH19N3O4]and bovine serum albumin（BSA） was studied via fluorescence quenching method. The static quenching constant KLBand some thermodynamic parameters（e.g.,△G^θ, △H^θ and △S^θ） of the interaction were estimated. In particular, thebinding site between the complex and BSA was explored by molecular docking. On the basis of the mechanism ofFOrster energy transfer, the binding distance between the bismuth（Ⅲ） Schiff base complex and BSA（Trp.213） as wellas the transfer efficiency was obtained. Furthermore, the effect of the bismuth（Ⅲ） Schiff base complex on the con-formation of BSA was investigated using synchronous fluorescence spectroscopy.

The title compound, methyl 4-(2,4-dichlorophenyl)-2,7,7-trimethyl-5-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylate(C_(20) H_(21)Cl_2NO_3, 1) was synthesized and the crystal structure was —determined by single-crystal X-ray diffraction. It crystallizes in tetragonal system, space group P421 c with a = 16.076(3), b = 16.076(3), c = 14.750(2) ?, V = 3811.8(14) ?3, Z = 8, R = 0.0377 and wR = 0.0845. The interactions between compound 1 and bovine serum albumin(BSA) were investigated using fluorescence spectroscopy. The results revealed that compound 1 can effectively quench the intrinsic fluorescence of BSA by static quenching mechanism. The thermodynamic parameters revealed that the action forces between compound 1 and BSA were mainly van der Waals forces and hydrogen bonds. According to F?rster's non-radioactive energy transfer theory, the binding distance between compound 1 and BSA had been determined. Furthermore, the synchronous fluorescence showed that compound 1 has few effects on the microenvironment and conformation of BSA in the binding process.

Male germline stem cells （mGSCs） are unique adult germ cells with self-renewal potential and spermatogenesis function in the testis. However, further studies are needed to establish a long-term cultural system of mGSCs in vitro, especially for large animals such as bovine mGSCs. In this study, we first established a stable immortalized bovine male germline stem cell line by transducing Simian virus 40 （SV40） large T antigen. The proliferation of these cells was improved significantly. These cells could express spermatogonial stem cell （SSC）-specific markers, such as PLZF, PGP9.5, VASA, LIN28A, and CD49F, both in the mRNA and protein levels. Additionally, these cells could be differentiated into three germ layer cells to enter meiosis, form colonies, and proliferate in the seminiferous tubules of busulfan-induced infertile mice. The immortalized bovine mGSCs maintain the criteria of mGSCs.

The aim of this study is to reveal the regulation mechanism of the effect of Semen vaccariae and Taraxacu mogono on the cell-cell adhersion molecule, E-cadherin and β-catenin on the proliferation role and secretion function of bovine mammary epithelial cells cultured in vitro. Firstly, the epithelial character of bovine mammary epithelial cells was authenticated using immunofluorescence, then the cell grow curve was observed and investigated after S. vaccariae and T. mogono treatment. On the effect of S. vaccariae and T. mogono, cell adhesion molecules E-cadherin, β-catenin and CycinD1 mRNA and protein were detected by qRT-PCR and Western blotting, respectively. The results showed that the cellular keratin 18 expressed positively and proliferfated vigorously after S. vaccariae and T. mogono treament. The mRNA and protein levels of E-cadherin and CycinD1 were remarkably higher （P〈0.05） in 36 h after S. vaccariae and T. mogono treatment. The cell proliferation at 36 h was increased significantly （P〈0.05）. In conclusion, S. vaccariae and T. mogono have a positive impact on the cell proliferation and an effect on the adhesion molecules E-cadherin, β-catenin and CycinD1 in the Wnt signaling pathway.

Twelve quarters of six lactating cows were inoculated with Mycoplasma leachii strain GN407 through intramammary ductal infusion; another 12 quarters were inoculated with heat-inactivated *** culture medium as negative *** procedures,including clinical assessment,etiology assessment,pathology and immunohistochemistry(IHC),were used to elucidate the pathogenicity of *** in bovine *** post-inoculation days(PIDs) 3 to 9,12 inoculated quarters developed mild to severe clinical mastitis and mammary tissue histopathological changes,including inflammatory cell infiltration and architectural destruction of mammary gland *** *** antigen was also detected by IHC in the mammary gland epithelial cells of the inoculated quarters as a weak signal on PID 6 and as a strong signal on PID *** control quarters also developed mild mastitis and histopathological changes on PID 9,and *** was also detected by *** the experimental period,the quarters of the negative control cow were clinically and pathologically normal,and the *** antigen was not *** conclusion,direct histological and immunohistochemical evidence confirmed that *** causes clinical bovine mastitis through histopathological lesions induced by invasion of the pathogen into mammary gland cells and through inflammatory cell infiltration.

Reconstructed embryos derived from intersubspecies somatic cell nuclear transfer （SCNT） have poorer developmental potential than those from intrasubspecies SCNT. Based on our previous study that Holstein dairy bovine （HD） mitochondrial DNA （mtDNA） haplotype compatibility between donor karyoplast and recipient cytoplast is crucial for SCNT embryo development, we performed intersubspecies SCNT using HD as donor karyoplast and Luxi yellow heifer （LY） as recipient cytoplast according to mtDNA haplotypes determined by polymerase chain reaction- restriction fragment length polymorphism （PCR-RFLP） analysis. The results demonstrated that intersubspecies mtDNA homotype SCNT embryos had higher pre- and post-implantation developmental competence than intrasubspecies mtDNA heterotype embryos as well as improved blastocyst reprogramming status, including normal H3K9 dimethylation pattern and promoter hypomethylation of pluripotent genes such as Oct4 and Sox2, suggesting that intersubspecies SCNT using LY oocytes maintains HD cloning efficiency and may reprogram HD nuclei to develop into a normal cloned animal ultimately. Our results indicated that karyoplast-cytoplast interactions and mtDNA haplotype compatibility may affect bovine intersubspecies SCNT efficiency. This study on bovine intersubspecies SCNT is valuable for understanding the mechanisms of mtDNA haplotype compatibility between karyoplast and cytoplast impacting the bovine SCNT efficiency, and provides an alternative and economic resource for HD cloning.

Effect of bovine serum albumin (BSA) on the temperature-dependent association behavior of poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO-PPO-PEO) block copolymers was investigated using pyrene fluorescence spectroscopy. The critical micellization temperature (CMT) of pluronics in aqueous solution was increased by the addition of BSA. A closed association model was used to obtain the standard free energies (△G0), enthalpies (△H 0), and entropies (△S 0) of micellization. The standard enthalpy and entropy of micellization for pluronic polymers in water were decreased with an increase of the BSA content. The more PPO component in the pluronic polymer, the higher the changed values of micellization enthalpy and entropy. The hydrophobic part of the pluronics, PPO, was responsible for the interaction between pluronics and BSA. Hydrophobic interaction between PPO and BSA was correlated to the alternation of the PPO-PPO interaction by the addition of BSA, which would shift the CMT toward higher temperature and alter the thermodynamic parameters of micellization for pluronics in aqueous solutions.

BGVD:An Integrated Database for bovine Sequencing Variations and Selective Signatures

Next-generation sequencing has yielded a vast amount of cattle genomic data for global characterization of population genetic diversity and identification of genomic regions under natural and artificial ***,efficient storage,querying,and visualization of such large datasets remain ***,we developed a comprehensive database,the bovine Genome Variation Database(BGVD).It provides six main functionalities:gene search,variation search,genomic signature search,Genome Browser,alignment search tools,and the genome coordinate conversion *** contains information on genomic variations comprising^60.44 M SNPs,~6.86 M indels,76,634 CNV regions,and signatures of selective sweeps in 432 samples from modern cattle *** can quickly retrieve distribution patterns of these variations for 54 cattle breeds through an interactive source of breed origin map,using a given gene symbol or genomic region for any of the three versions of the bovine reference genomes(ARS-UCD1.2,UMD3.1.1,and Btau5.0.1).Signals of selection sweep are displayed as Manhattan plots and Genome Browser *** further investigate and visualize the relationships between variants and signatures of selection,the Genome Browser integrates all variations,selection data,and resources,from NCBI,the UCSC Genome Browser,and Animal ***,all these features make the BGVD a useful archive for in-depth data mining and analyses of cattle biology and cattle breeding on a global *** is publicly available at http://***/BosVar.
