Reflections on the calculation methods of the imbalance degree in the biophysical properties of meridian points

Objective To analyze and compare the features and strengths of different methods for calculating the biophysical properties of meridian points,and thus propose corresponding suggestions to fully achieve the research and application value of biophysical properties of meridian *** We searched and collected the literature on the imbalance of biophysical properties of meridian points between January 1,2005 and March 1,2020 in China National Knowledge Infrastructure(CNKI),Wanfang Academic Journal Full-text Database(Wanfang),Chongqing VIP Database(CQVIP)and PubMed database,and then analyzed,compared,and summarized the applied methods for calculating the imbalance degree of the biophysical properties of meridian *** The current methods for calculating the imbalance degree of the biophysical properties of meridian points are diverse and can be summarized as the following three:direct comparison of the measured values of the left and right namesake points,difference method,and ratio *** low uniformity of the calculation methods has limited the promotion and application of its research *** In future research on the biophysical properties of meridian points,multidisciplinary cooperation in terms of imbalance degree calculation methods,detection instruments,and health data models is necessary to achieve more widely applicable scientific conclusions and more generalized experimental results.

Objective:To investigate the effects of Tropomodulinl(Tmodl),a cytoskeleton protein that caps the pointed ends of F-actin,on the immune functions and biophysical properties of dendritic cells(DCs) and the underlying mechanisms in order to further understand the biological behaviors of ***:The expressions of Tmod1,the surface makers and the inflammatory cytokines in bone marrow-derived DCs from wild type(C57 BL/6 J) mice and Tmod1 knockout mice were ***,the immune functions and the biophysical properties of DCs were compared,including the migration abilities,endocytosis and antigen-presenting capacity in vitro and in *** osmotic fragilities,Young’s modulus and F-actin content were also compared between these two kinds of ***,we detected the changes of NF-κB and MAPKs signaling pathways to discover the molecular *** and Conclusion:Tmod1 was expressed in immature DCs(imDCs) and its expression level was significantly elevated in mature DCs(mDCs).It seems that Tmod1 has no obvious effects on imDCs’ ***,Tmod1-deficient mDCs were failed to express enough MHC-Ⅱ,co-stimulated molecules(CD80/86,CD40) and CCR7 on their cell surface and secrete enough inflammatory cytokines such as IL-12,TNF-α and IFN-γ,which leaded to their deficient antigen-presenting abilities and chemotaxis *** underlying mechanism is due to the inhibitions of the LPSstimulated NF-κB and p38 MAPK signaling pathways and the disturbed reorganization of the F-actin cytoskeleton including less F-actin content and shorter dendritic ***,as compared to mDCs in wild type mice,mDCs in Tmod1 knockout mice showed higher osmotic fragility and lower Young’s *** results indicated that Tmod1 is also involved in regulating the biophysical changes of mDCs induced by *** results can provide a new insight on the functions of Tmod1 and improve our understanding of DCs immune functions.

Dendritic cells(DCs) are professional antigen-presenting cells(APCs),which play an important role in initiating and amplifying both the innate and adaptive immune *** performing their immune functions,DCs are always suffered the stimulation of shear stress(SS) derived from blood,interstitial and lymph fluids,however,how DCs responses to physiological fluid flow-derived SS are still *** this study,the DCs were exposed to different laminar flow SS for a given time course,the morphology was observed under microscope,the endocytotic activities of im DCs were detected by FITC-dextran particles,the biomechanical characteristics including membrane fluidity and osmotic fragility of cells were measured,the expression levels of immune function related-molecules were investigated by flow cytometry and real-time *** results showed that the morphological characteristics and F-actin cytoskeleton organizations of cells were significantly remodeled by SS,the endocytotic activities of imD Cs were significantly impaired by SS(p<0.05),the membrane fluidities of im DCs and m DCs were respectively significantly increased and decrease(p<0.05),the osmotic fragilities of im DCs were increased,moreover,immune function related-molecules including CD11 c,CD40,CD80,CD83,CD86,CCR7,CD1 a and CD205 were down-regulated by SS at the transcriptional and translational levels(p<0.05 or p<0.01).These data indicated that SS could be a negative regulator for DCs-induced immune response,supporting the viewpoint of immuno-mechanobiology or *** is significant for further understanding the DCs biological behavior.

Aim:Skin firmness is one of the key parameters to define skin quality and facial *** the minimally invasive anti-aging strategies,hyaluronic acid(HA)injection is widely accepted to improve skin *** most of the available HA injectables are designed and intended for intradermal injection,a novel HA/sorbitol composition containing 2.6% high molecular weight hyaluronic acid stabilized by sorbitol was recently developed to be specifically injected into the adipose tissue to improve the quality of all skin layers,especially the skin ***:The HA/sorbitol composition was investigated versus product comparators in terms of biophysical properties,tolerance in subcutaneous tissue with in vivo implantation study,and skin firmness assessment on human skin ***:The HA/sorbitol composition was characterized by unique and differentiated biophysical properties,proper distribution and high tolerance of the gel composition into the adipose tissue,and the ability to efficiently improve skin ***:The HA/sorbitol composition represents a new attractive solution to treat facial skin aging with an injection strategy specifically targeting the adipose tissue instead of the dermis,to improve the quality of the skin.

Protocol for analyzing protein liquid–liquid phase separation

Numerous cellular functions occur in spatially and temporally confined regions. Recent studies have shown that membrane-less organelles and compartments in the cell are assembled via liquid–liquid phase separation(LLPS). In vitro LLPS assays using recombinant expressed and purified proteins are necessary for us to further understand how the assembly of phase-separated compartments is regulated in cells. However, uniform standards and protocols are lacking for these in vitro studies. Here, we describe a step-by-step protocol commonly used to investigate in vitro LLPS using purified *** protocol includes expression and purification of the studied proteins, induction of LLPS of the purified proteins, and studies of the biophysical properties of the liquid droplets formed by LLPS. These protocols can be easily followed by researchers to investigate the LLPS behaviors of proteins of interest.
