The structures and behaviors of nanomaterials atnano-bio interfaces are the basis of biomedical effects of nanomaterials. It is crucial to reveal the interfacial behaviors of metal nanoparticles when they interact with cells, however, highly sensitive and in situ analytical techniques are crucial and required. To understandthe mechanism of biomedical effects from interfacial aspects, we systematically developed analytical methods replying on synchrotron radiation and quantify both interfacial structures and nanomaterials. These techniques can capture both their location and chemical changessuch as adsorption of biomolecules, chemical transformation, catalysis, and redox by synchrotron radiation techniques. For example, spectroscopic techniques are used to characterize the binding structure of hard protein corona on nanomaterials, which shows how corona mediates cytotoxicity. X-ray liquid scattering can as well reveal the binding structures of nano-lipid interfaces to study the interaction between nanomaterials and biological membrane. Moreover, X-rayhigh resolution imaging is employed to capturethe accumulation and location of intracellular metal nanomaterials companying with their chemical form. Finally,we develop methods to characterize metal nanomaterial-involved catalysis and degradation in biological systems and show the determinant for the cytotoxicity from the free radicals or the products after chemical transformation. In conclusion, these methods provide accurate information for chemical forms, structures, and element imaging with promises in nanotoxicology and nanomedicine.
