Protection of Allergic Asthma in Mice by Black Rice Bran Bioprocessed with Shiitake Mushroom Mycelia

We evaluated the potential of orally fed new food formulations to inhibit biomarkers reported to be involved in the causes of allergic asthma in mice. Asthma, a serious non-communicable disease, affects both adults and children and can be undertreated. New functional foods could provide therapeutic approaches. Here, the anti-asthma mechanism of a new functional food and three isolated fractions produced by bioprocessing black rice bran with shiitake mushroom mycelia was evaluated in mast cells, B cells, and orally fed mice and compared with non-bioprocessed black rice bran. In vitro, the treatments inhibited RBL-2H3 cell degranulation and immunoglobulin E (IgE) production. The in vitro anti-asthma effects were confirmed in orally fed mice following asthma induction by alumina and chicken egg ovalbumin (OVA). The suppression of asthma resulted from the inhibition of inflammation- and immune-related substances, including OVA-specific IgE, thymic stromal lymphopoietin, eotaxin, leukotriene C4, prostaglandin D2, and vascular cell adhesion molecule-1 in bronchoalveolar lavage fluid and serum. The treatment also reversed the thickening of the lung airway wall. The inflammation and asthma inhibition seems to be regulated by the balance of the T-helper cells’ Th1/Th2 immune response and the inhibition of multiple biomarkers associated with the cause of asthma. Future human clinical studies with adults and children should determine the potential therapeutic value of the anti-asthma effects of the new functional foods.

Lethal and Sublethal Effects of Leaves Extracts from Native and Exotic Plants in Southern Brazil against Spodoptera frugiperda （Lepidoptera： Noctuidae） Caterpillars

The caterpillar Spodopterafrugiperda is an important pest of several crops, due to the damage it causes and the difficulty of its control. The method of controlling these pests is through chemical insecticides, which cause adverse effects to the biotic and abiotic environment. The selection of new insecticides that meet the requirements of efficacy, safety, selectivity and those are economically viable, is highlighting the study of mechanisms of plant defense. The present study aimed to verify the insecticide effect of 27 plants in three different types of plants extracts： aqueous, decoction and polypeptide on S. frugiperda. Three plants were chosen for performing sublethal effects bioassays： two plants with the highest corrected mortality （E. pulcherrima and R. simsii） and the other plant （Maytenus ilicifolia （espinheira santa）） with corrected mortality similar to control. In preliminaries bioassays, 25 plants showed corrected mortality less than 50%, according to Abbott＇s formula. R. simsii （azalea） and E. pulcherrima （billed parrot） showed 51% and 68% of corrected mortality, respectively. Only R. simsii aqueous and the three extracts of E. pulcherrima differ significantly from control （P 〈 0.05）. The aqueous extracts and decoction of R. simsii and polypeptide extract of E. pulcherrima reduced the larval period of S. frugiperda, different from control （P 〈 0.05）. The polypeptide extract of R. simsii reduced the pupal period of the target insect. The extracts obtained by decoction, aqueous and polypeptide of E. pulcherrima extracts and decoction of polypeptides and R. simsii affected the fertility and fecundity of S. frugiperda. For the other parameters, there is no significant difference when compared with the control. The results of this study suggest that the extracts obtained by decoction, aqueous and polypeptide of E. pulcherrima and R. simsii can be used to control S. frugiperda. Dissociation enhanced lanthanide fluoroimmunoassay（DELFIA）, as one of the most sensitive luminescent bioassay techniques, has been widely adopted in a variety of research and medical institutions. However, commercial DELFIA using molecular probes like lanthanide（Ln3+）-chelates as biolabels often suffers from a series of disadvantages, such as a low labelling ratio of Ln3+ per biomolecule（up to 10-30）, low photostability and high cost of Ln3+-chelates. To overcome these concerns, herein we develop a unique and facile bioassay method based on the dissolution-enhanced luminescence of inorganic lanthanide nanoparticles（NPs） [1]. Due to the highly concentrated Ln3+ ions in a single Ln3+-NP, an extremely high Ln3+ labeling ratio can be achieved, which enormously amplifies the luminescent signal and thus improves the detection sensitivity as compared to that of DELFIA（Fig. 1）. Utilizing sub-10 nm NaEuF4 NPs as dissolution-enhanced luminescent nanoprobes, we demonstrate the successful in vitro detection of carcinoembryonic antigen（CEA, an important tumor marker） in human serum samples with a detection limit down to 0.1 pg/mL（0.5 f M）, which is the lowest among those ever reported（about 3 orders of magnitude improvement relative to that of DELFIA）. The proposed dissolution-enhanced luminescent bioassay, as an alternative to commercial DELFIA by simply replacing the molecular probes with inorganic lanthanide nanoprobes in its protocol, shows great promise in versatile biomedical applications such as ultrasensitive and multiplexed in vitro detection of disease markers in clinical diagnosis [2, 3].

In vitro antibacterial and antitumor activities of some medicinal plant extracts, growing in Turkey

Objective: To investigate antibacterial and antitumor activities of 51 different extracts prepared with 3 types of solvents (water, ethanol and methanol) of 16 different plant species (Ajuga reptans (A. reptans) L., Phlomis pungens (P. pungens) Willd., Marrubium astracanicum (M. astracanicum) Jacq., Nepeta nuda (N. nuda) L., Stachys annua (S. annua) L., Genista lydia (G. lydia) Boiss., Nuphar lutea (N. lutea) L., Nymphaea alba (N. alba) L., Vinca minor (V. minor) L., Stellaria media (S. media) L., Capsella bursa-pastoris (C. bursa-pastoris) L., Galium spurium (G. spurium) L., Onosma heterophyllum (O. heterophyllum) Griseb., Reseda luteola (R. luteola) L., Viburnum lantana (V. lantana) L. and Mercurialis annua (M. annua) L.) grown in Turkey was conducted. Methods: Antibacterial activity was evaluated with 10 bacteria including Streptococcus pyogenes (S. pyogenes), Staphylococcus aureus (S. aureus), Staphylococcus epidermidis (S. epidermidis), Escheria coli (E. coli), Pseudomonas aeruginosa (P. aeruginosa), Salmonella typhimurium (S. typhimurium), Serratia marcescens (S. marcescens), Proteus vulgaris (P. vulgaris), Enterobacter cloacae (E. cloacea), and Klebsiella pneumoniae (K. pneumoniae) by using disc diffusion method. Antitumor activity was evaluated with Agrobacterium tumefaciens (A. tumefaciens)-induced potato disc tumor assay. Results: Best antibacterial activity was obtained with ethanolic extract of P. pungens against S. pyogenes . Ethanolic and methanolic extract of N. alba and ethanolic extract of G. lydia also showed strong antibacterial activities. Results indicated that alcoholic extracts especially ethanolic extracts exhibited strong antibacterial activity against both gram-positive and gram-negative bacteria. Best antitumor activity was obtained with methanolic extracts of N. alba and V. lantana (100% tumor inhibition).
