A biomaterial-based therapy for lower limb ischemia using Sr/Si bioactive hydrogel that inhibits skeletal muscle necrosis and enhances angiogenesis

Muscle necrosis and angiogenesis are two major challenges in the treatment of lower-limb ischemic *** this study,a triple-functional Sr/Si-containing bioceramic/alginate composite hydrogel with simultaneous bioactivity in enhancing angiogenesis,regulating inflammation,and inhibiting muscle necrosis was designed to treat lower-limb ischemic *** particular,sodium alginate,calcium silicate and strontium carbonate were used to prepare injectable hydrogels,which was gelled within 10 *** importantly,this composite hydrogel sustainedly releases bioactive Sr^(2+)and SiO_(3)^(2-) ions within 28 *** biological activity of the bioactive ions released from the hydrogels was verified on HUVECs,SMCs,C2C12 and Raw 264.7 cells in vitro,and the therapeutic effect of the hydrogel was confirmed using C57BL/6 mouse model of femoral artery ligation in *** results showed that the composite hydrogel stimulated angiogenesis,developed new collateral capillaries,and re-established the blood *** addition,the bioactive hydrogel directly promoted the expression of muscle-regulating factors(MyoG and MyoD)to protect skeletal muscle from necrosis,inhibited M1 polarization,and promoted M2 polarization of macrophages to reduce inflammation,thereby protecting skeletal muscle cells and indirectly promoting *** results indicate that these bioceramic/alginate composite bioactive hydrogels are effective biomaterials for treating hindlimb ischemia and suggest that biomaterial-based approaches may have remarkable potential in treating ischemic diseases.

Fabricating oxygen self-supplying 3D printed bioactive hydrogel scaffold for augmented vascularized bone regeneration

Limited cells and factors,inadequate mechanical properties,and necrosis of defects center have hindered the wide clinical application of bone-tissue engineering ***,we construct a self-oxygenated 3D printed bioactive hydrogel scaffold by integrating oxygen-generating nanoparticles and hybrid double network hydrogel *** hydrogel scaffold possesses the characteristics of extracellular matrix;Meanwhile,the fabricated hybrid double network structure by polyacrylamide and CaCl2-crosslinked sodium carboxymethylcellulose endows the hydrogel favorable compressive strength and 3D ***,the O2 generated by CaO2 nanoparticles encapsulated in ZIF-8 releases steadily and sustainably because of the well-developed microporous structure of ZIF-8,which can significantly promote cell viability and proliferation in vitro,as well as angiogenesis and osteogenic differentiation with the assistance of Zn2+.More significantly,the synergy of O2 and 3D printed pore structure can prevent necrosis of defects center and facilitate cell infiltration by providing cells the nutrients and space they need,which can further induce vascular network ingrowth and accelerate bone regeneration in all areas of the defect in ***,this work provides a new avenue for preparing cell/factor-free bone-tissue engineered scaffolds that possess great potential for tissue regeneration and clinical alternative.

An intrinsically bioactive hydrogel with on-demand drug release behaviors for diabetic wound healing

Prolonged,intense inflammation and excessive oxidative stress hinder diabetic wounds from healing normally,leading to disorders downstream including the postponement of re-epithelialization and extracellular matrix(ECM)***,we report a hyaluronic acid(HA)and chitosan based hydrogel(OHA-CMC)with inherent antibacterial and hemostatic activities fabricated via Schiff base *** encapsulating nanotechnologically-modified curcumin(CNP)and epidermal growth factor(EGF)into the hydrogel,OHA-CMC/CNP/EGF exhibited extraordinary antioxidant,anti-inflammatory,and migration-promoting effects in ***,OHA-CMC/CNP/EGF presented on-demand drug release in synchrony with the phases of the wound healing ***,curcumin was rapidly and constantly released to alleviate inflammation and oxidative stress in the early phase of wound healing,while a more gradual and sustained release of EGF supported late proliferation and ECM *** a diabetic full-thickness skin defect model,OHA-CMC/CNP/EGF dramatically improved wound healing with ideal re-epithelialization,granulation tissue formation,and skin appendage regeneration,highlighting the enormous therapeutic potential this biomaterial holds as a diabetic wound dressing.
