Regenerative medicine and anti-aging research have made great strides at the molecular and cellular levels in dermatology and the medical aesthetic field,targeting potential treatments with skin therapeutic and intervention pathways,which make it possible to develop effective skin regeneration and repair *** the rapid development of computational biology,bioinformatics as well as artificial intelligence(A.I.),the development of new ingredients for regenerative medicine has been greatly accelerated,and the success rate has been *** application cases have appeared in topical skin regeneration and repair *** review will briefly introduce the application of bioactive peptides in skin repair and anti-aging as emerging ingredients in cosmeceutics and emphasize how *** computational biology technology may accelerate the development of innovative peptide molecules and ultimately translate them into potential skin regenerative and anti-aging ***,two research routines have been summarized and current limitations as well as directions were discussed for border applications in future research.

Suppression of postprandial blood glucose elevation by buckwheat(Fagpopyrum esculentum) albumin hydrolysate and identification of the peptide responsible to the function

The objective of this study is to evaluate the suppressive effect of buckwheat-albumin hydrolysate on postprandial hyperglycemia and identify the peptide responsible to the function. Buckwheat-albumin hydrolysate was prepared by using digestive enzymes and was orally administered to rats together with soluble starch. The blood was taken from the tail vein up to 90 min after oral administration to measure blood-glucose and plasma-insulin levels. The peptide with α-amylase inhibitory activity was purified from the buckwheathydrolysate by gel-filtration chromatography, α-amylase affinity chromatography, and high performance liquid chromatography(HPLC). The amino-acid sequence of the peptide was identified by a protein sequencer and was compared with that in the buckwheat-genome database. Buckwheat-albumin hydrolysate significantly suppressed the elevation of blood glucose level 15 min after starch administration. The amino-acid sequence of the peptide with α-amylase inhibitory activity was YVEPDCGNLGCCYHC in the parental protein of molecular mass 17.8 k Da and theoretical pI 4.77. The amino-acid sequence, molecular weight, and pI of the parental protein in buckwheat albumin were different from those of α-amylase inhibitor in wheat albumin. This study suggests that the novel α-amylase inhibitor identified in buckwheat albumin is a potential candidate for a functional food material to suppress postprandial blood glucose elevation.

Screening of protease-producing marine yeasts for production of the bioactive peptides

Over 400 yeast strains from seawater and sediments were obtained, but only five strains named HN2 -3, N13d, N13C, Mb5 and HN3 - 2 among them could form clear zones around their colonies on the double plates with 2.0% casein. peptides in the hydrolysate produced by the proteases from strains HN2 -3 and N13d had higher angiotensin I-converting-enzyme （ACE）-inhibitory activity. The two marine yeast strains were identified to be Aureobasidium pullulans according to the results of routine yeast identification and molecular methods. After purification of the proteases from the two marine yeast strains, it was found that the optimal pH for them was both 9.0, both of them were serine alkaline protease. However, the optimal temperature for the protease from the strain HN2 -3 was 52℃ while that from strain N13d was 48℃. ACE-inhibitory activity of the peptides in the hydrolysate of shrimp protein produced by the purified protease from the strain HN2 -3 was the highest while antioxidant activity in the hydrolysate of spirulina protein produced by the purified protease from the strain N13d was the highest.

Effects of periodical application of bioactive peptides derived from cottonseed on performance,immunity,total antioxidant activity of serum and intestinal development of broilers

This experiment aimed to examine the effect of periodical application of bioactive peptides derived from cottonseed(BPC)in comparison with using sub-therapeutic doses of lincomycin and the excessive in-clusion of vitamin E on performance,immunity,total antioxidant capacity of serum and intestinal morphology of broiler chickens.A total of 240 one-d-old male broiler chicks with similar initial weight(Ross strain)were randomly assigned to 6 groups(8 chicks/pen):non-treated group(basal diet),basal diet supplemented with 2 mg/kg lincomycin,basal diet supplemented with 50 IU vitamin E,basal diet supplemented with 6 g BPC/kg in starter period,basal diet supplemented with 6 g BPC/kg in starter and grower periods and basal diet supplemented with 6 g BPC/kg throughout the whole *** highest final body weight was obtained in the group supplemented with BPC in starter and grower *** the finisher phase,broilers fed the diet containing BPC in the starter period and in the whole trial had significantly(P0.05).Broilers supplemented with BPC in starter and grower,and in the whole trial had significantly(P<0.05)higher antibody titers against sheep red blood cells(SRBC).The highest total antioxidant capacity was obtained in broilers supplemented with the excessive level of vitamin E,furthermore it tended to improve in broilers fed the diet containing BPC in the whole *** summary,the results of the study indicated that addition of BPC in broiler diets in the

Isolation and identification of novel casein-derived bioactive peptides and potential functions in fermented casein with Lactobacillus helveticus

The present study here establishes a complete and effective method for isolating,purifying and identifying extracellular and intracellular peptides,and also describes the characters and bioactivities of peptides from fermented casein with Lactobacillus *** peptides are much larger in quantity and more complex in composition than extracellular peptides,between which the correlation reveals proteolytic and metabolic *** addition,totally 241 different peptide sequences were identified by Nano LC–MS/MS from casein(212)and Lactobacillus helveticus proteins(29).These casein-derived peptides mostly originated from-casein,followed byS1-casein,-casein,andS2-casein,and came from extracell(69)and intracell(143),in which common peptides have a total of ***-four of the identified peptides were previously described as bioactive,including angiotensinconverting enzyme(ACE)-inhibitory,antioxidant,immunomodulating,antimicrobial,DPP-IV inhibitory,antiamnesic and anticancer effects and so *** peptides with the potential of some biological activities are obtained,which were described in previous studies.A total of 47 novel peptides of 5 to 26 amino acids that were not disclosed were *** new sources of natural bioactive peptides may have the very high application value as potential new peptide drugs for treatment human *** product peptide DELQDKIHPF found in both extracell and intracell was quantitatively analyzed using the MRM mode of UPLC-U3Q,23.1 and 9.76 ng/mL,*** quantitative analysis of the potential bioactive peptide may also advance the production of peptide products in the future.

Anti-diabetic and anti-hyperlipidemic effects of sea cucumber(Cucumaria frondosa) gonad hydrolysates in type Ⅱ diabetic rats

The present study investigated the anti-diabetic activity and potential mechanisms of sea cucumber gonad hydrolysates(SCGH)in a rat model of type II diabetes induced by streptozotocin(STZ)combined with high-fat diet(HFD).Results showed that SCGH significantly reduced water intake,fasting blood glucose level and glycated hemoglobin ***,the oral glucose tolerance,insulin resistance and plasma lipid level in diabetic rats were also ***,histological analysis showed that SCGH effectively protected the tissue structure of *** addition,mechanism studies showed that SCGH improved glucose metabolism via activating the IRS/Akt signaling pathway,and promoted lipid metabolism via activating the AMPK signaling *** summary,these findings suggested that SCGH have potential anti-diabetic effects by improving insulin resistance and lipid metabolism disorders.

Selenopeptide(Sec-Arg-Gly-Asp-Cys,URGDC) and cysteamine-cofunctionalized gold nanoparticles monolayer protected cluster(MPC),catalyzes the reaction of H O by GSH,exhibiting Glutathione peroxidase(GPx)-like *** nanozyme was constructed through two self-assemble steps:First,URGDC was conjugated to AuNPs,forming a multivalent catalyst(MPC 1);second,the positively charged cysteamine was subsequently employed to optimize the enzyme activity by regulating the surface chemistry of gold nanoparticles(MPC 2).The systematical study of the catalytic capability showed that the Nanozyme catalyzed reaction exhibits saturation kinetics:the reactive rate data complied with the Michaelis-Menten model of enzyme kinetics,and the GPx activity of MPC land MPC 2 is 5.05 and 7.21 U/umol,about 14 and 20 times of that of free *** kinetic analysis indicated that the catalytic mechanism of free peptide is ping-pong mechanism,while it is the ordered mechanism for nanozyme(MPC 1,MPC 2).And the catalytic constant K/K of both GSH and H O are one order of magnitude higher than that of free *** manifested that gold nanoparticle as a scaffold excels in limiting the mobility and constraining conformation of URGDC,and the multivalent selenopeptide act cooperatively in the catalysis *** surface/interface regulationof gold nanoparticles facilitates the recognition and binding of *** rational design of nanozyme greatly improves and amplifies the catalytic activity of ’active unit’ *** work provides an alternative strategy to establish GPx mimic and new insights regarding the surface chemistry of AuNPs as a nanozyme.
