The porcine reproductive and respiratory syndrome virus(PRRSV)RNA endoribonuclease nsp11 belongs to the Xendo U superfamily,and plays a crucial role in viral RNA *** date,no structure has been reported for an arterivirus endoribonuclease ***,we report the first crystal structure of the arterivirus nsp11 from PRRSV,which exhibits a unique structure and assembles into an asymmetric dimer that is completely different from the hexameric structure of coronavirus ***,the structures of the PRRSV nsp11 and coronavirus nsp15 cocatalytic domains were perfectly superimposed,especially in the"active site loop"and"supporting loop"***,our biochemical data demonstrated that PRRSV nsp11 was dimeric form in *** of the major dimerization site determinants(Ser74 and Phe76)in the dimerization interface destabilized the stable dimers in solution and severely diminished the endoribonuclease activity,indicating that the dimer was the biologically functional *** the dimeric structure,the catalytic loop(His129-His144)and supporting loop(Val162-Thr179)are packed against one another and stabilized by monomer-monomer *** results support the hypothesis that these two loops may be flexibly disordered when the dimeric forms are ***,the mutagenesis analysis demonstrated that the endoribonuclease activity was essential for nsp11s function as an interferon-β*** findings may help elucidate the mechanism underlying PRRSV replication and may provide great potential for antiviral drug development.
