Correlation between expression of gastrin, somatostatin and cell apoptosis regulation gene bcl-2/bax in large intestine carcinoma

AIM: To explore the correlation between expression of somatostatin (SS), gastrin (GAS) and cell apoptosis regulation gene bcl-2/bax in large intestine ***: Sixty-two large intestine cancer tissue samples were randomly and retrospectively selected from patients with large intestine carcinoma. Immunohistochemical staining for bcl-2, bax, GAS, SS was performed according to the standard streptavidin-biotin-peroxidase (S-P) *** to the semi-quantitative integral evaluation, SS and GAS were divided into three groups as follows. Scores1-3 were defined as the low expression group, 4-8 as the intermediate expression group, 9-16 as the high expression group. bax and bcl-2 protein expressions in different GAS and SS expression groups of large intestine carcinoma were ***: The positive expression rate of bax had a prominent difference between SS and GAS high, intermediate and low expression groups (P＜0.05, x2ss = 9.246; P＜0.05,x2GAS = 6.981). The positive expression rate of bax in SS high (80.0%, 8/10) and intermediate (76.5%, 13/17)expression groups was higher than that in low expression group (40.0%, 14/35) (P＜0.05, x2high vs low = 5.242; P＜0.05,x2middle vs low = 6.097). The positive expression rate of bax in GAS high expression group (27.3%, 3/8) was lower than that in low expression group (69.4%, 25/36) (P＜0.05,x2 = 4.594). However, bax expression in GAS intermediate expression group (46.7%, 7/15) was lower than that in low expression group, but not statistically significant. The positive expression rate of bcl-2 had a prominent difference between SS and GAS high, intermediate and low expression groups (P＜0.05, x2ss = 7.178; P＜0.05, x2GAS = 13.831). The positive expression rate of bcl-2 in GAS high (90.9%, 10/11)and intermediate (86.7%, 13/15) expression groups was higher than that in low expression group (44.4%, 16/36)(P＜0.05,x2high vs low = 5.600; P＜0.05, x2 middle vs low = 7.695).However, the positive expression rate of bcl-2 in SS

Apopotic gene bax expression in carotid plaque

The expression of bax in carotid atherosclerosis and its regulation is far from defined. Objectives To investigate bax expression in stable/fibrous and instable/vulnerable carotid plaque and its clinical significance. Methods 25 cases of carotid plaque specimens obtained from endarterectomy were divided into two groups, stable/fibrous 14 cases, vulnerable/instable 11 cases; aortic artery and its branches from hepatic transplantation donors 6 case as control The expression of proapoptotic bax was detected by immunohistochemistry （IHC）, in situ hybridization （ISH） and in situ TdT dUTP nick end labeling （TUNEL）. Results 5 cases of bax （＋） were detected by ICH and ISH, 4 case of TUNEL （＋） were detected by TUNEL in stable/fibrous carotid plaque , while 10 cases were bax （＋）by IHC（P 〈 0.05 ） , 11 case by ISH and 9 case by TUNEL were detected in instable/vulnerable carotid plaque （P 〈0.01 ）, respectively. The intensity of bax （＋） cells by IHC and ISH was 8.63±2.62 and 10.32 ± 3.12 in fibrous plaques,whereas 122 ±21.64 and 152 ± 23.35 in vulnerable plaques, respectively. No expression of bax was found in controlled group. Conclusion The higher expression of bax in vulnerable carotid plaque may be one mechanisms in molecular pathogenesis of carotid atherosclerosis which affect plaque stability and be the cause of higher incidence of stroke than fibrous carotid plaques, the regulation of bax expression in different stage of atherosclerosis may provide targets in gene therapy for carotid atherosclerosis.

Triptolide and management of systemic malignancies besides pancreatic carcinomas

The recent article by Zhou et al was highly interesting and thought provoking. The authors have clearly shown that triptolide administration is associated with upregulation of the bax gene, resulting in an attenuating effect on cell growth in gastrointestinal malignancies such as pancreatic carcinomas. The article by Zhou et al is all the more important because it highlights the rapidly increasing role of triplodide in the management of systemic malignancies. For instance, triptolide acts on the PI3K/Akt/NF-κB pathway, thereby enhancing apoptosis secondary to the administration of bortezomib in multiple myeloma cells. Similar synergisms are seen when triptolide is administered along with 5-fluoruracil for the management of colonic carcinomas. Similarly, triptolide causes down-regulation of the Bcl-2 gene, resulting in control of cell growth in tumors, such as glioblastoma multiformes.

Intravenous acid fibroblast growth factor protects intestinal mucosal cells against ischemia-reperfusion injury via regulating Bcl-2/bax expression

AIM: To detect the effect of acid fibroblast growth factor (aFGF) on apoptosis and gene expression of bax and bcl-2 gene in rat intestine after ischemia/reperfusion (I/R) injury, and to explore the protective mechanisms of ***: One hundred and eight Wistar rats were randomly divided into sham-operated control group (C)(n = 6), intestinal ischemia group (I) (n = 6), aFGF treatment group (A) (n = 48) and intestinal ischemia reperfusion group (R) (n = 48). In group I, the animals were killed after 45 min of superior mesenteric artery (SMA) occlusion, while in groups R and A, the rats sustained 45 min of SMA occlusion and were then treated with normal saline and aFGF, respectively, sustained 15 min, 30 min, 1, 2, 6, 12, 24, or 48 h of reperfusion, respectively. In group C, SMA was separated, but without occlusion. Apoptosis in intestinal villus was determined with terminal deoxynucleotidyl transferase mediated dUTP-biotin nickend labeling technique (TUNEL). Intestinal tissue samples were taken not only for detection of bax and bcl-2 gene expression by RT-PCR, but also for detection of bax and bcl 2 protein expression and distribution by immunohistochemical ***: The rat survival rates in aFGF treated group were higher than group R (P＜0.05) and the improvement of intestinal histological structures was observed at 2, 6, and 12 h after the reperfusion in group A compared with group R. The apoptotic rates were (41.17±3.49)%, (42.83±5.23)% and (53.33±6.92)% at 2, 6 and 12 h after reperfusion, respectively in group A, apparently less than those of group R at matched time points (50.67±6.95, 54.17±7.86, 64.33±6.47, respectively) (P＜0.05). BACKGROUND： Operation of spine does not involve in spinal cord; however, spinal cord injury occurs at post-operation induced by unclear factors. Meanwhile, after decompression of lumbar spinal canal, symptoms are severer and severer. In addition, during extirpation of cervical and lumbar intervertabral disc, spinal cord and its vessels are not damaged, but spinal cord injury is also suffered from patients with partial improvement. All statuses mentioned above are related to expressed changes of bc1-2 gene inhibiting apoptosis and bax gene accelerating apoptosis. BJECTIVE： To observe motor function of hindlimbs of ischemia/reperfusion model in rabbits at various time points after reperfusion and expressions of apoptosis correlated protein Bcl-2 and bax. DESIGN： Completely randomized grouping design and contrast study SETTING： Department of Orthopedics, the First Affiliated Hospital of Medical School, Xi＇an Jiao Tong University MATERIALS ： Forty-eight New Zealand white rabbits of both genders were randomly divided into sham operation group （n =24） and model group （n =24）, and then, rabbits in each group were observed at four time points： 8, 24, 72 and 168 hours after reperfusion, with 6 in each time point. Rabbit-anti-rabbit Bcl-2 antibody and rabbit-anti-rabbit bax antibody were provided by Boster Company. The procedures were accordant to the kit introduction. METHODS ： The experiment was carded out at Laboratory of Orthopaedics of First Affiliated Hospital of Medical School, Xi＇an Jiao Tong University from April to August 2005. （1） Delayed paralysis models of spinal cord after ischemia/reperfusion were established based on method of Zivin et al. The Variety Selection and Testing for Effect on HepG-2 Cell Line of Dioscorea membranacea Pierre （Hua Khao Yen Tai） Extract Using Molecular genetics

In this research, Dioscorea membranacea Pierre was studied by using 30 samples from Khao Ruak Sub-district, Chai Badan District, Lop Buri Province, in Thailand. In this research, some morphology was studied including shapes, leaves, stem colors, epidermal cells, stomata sizes and stipules. To study the genetic relationships, the AFLP technique and computer program were used. The Dioscorea membranacea Pierre was classified into 2 groups according to its phylogenetic type： the first group was ＂Hua Khao Yen Tai-Nuea＂（Smilax corbularia Kunth）, and the second group included 30 further samples of Hua Khan Yen Tai （Dioscorea membranacea Pierre）. The ethanolic crude extract was also applied to test the anti-proliferative activity in the liver hepatocellular carcinoma （HepG2） cell lines which illustrates the characteristics of apoptosis： cell shrinkage, membrane blebbing and nuclear condensation. The expression ofbax gene is increased more than that of the control group while Bcl-2 gene which is anti-apoptotic is decreased. Furthermore, the result of western blot analysis reveals the up-regulation of bax protein and down-regulation of Bcl-2 protein when compared with untreated cells. This might indicate that ethanolic crude extracts of Hua Khao Yen Tai could induce apoptosis and anti-proliferative on HepG2 cell lines, The results also revealed that some morphology cannot be used to predict which Dioscorea membranacea Pierre plants would be most effective.

Effect of whole methanolic extract of Galium verum on AGO cell line

Objectives:Although many studies have suggested the anticancer properties of Galium verum,there is still no accurate information regarding its side effects on normal ***,this study aimed to investigate the dual effects of the whole Galium verummethanolic extract on the normal human fibroblast cell line(AGO)cell line at different ***:The cell line was randomly divided into a control group and groups exposed to concentrations of 12.5 to 400μg/*** was performed by the maceration *** addition,the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazoliumbromide(MTT)method was applied to measure cytotoxicity and flow ***,BCL2 associated X(bax)andB-cell lymphoma 2(BCL2)genes were expressed by the real-time polymerase chain reaction to evaluate apoptosis and reactive oxygen species(ROS).Finally,data were compared between groups using a one-way analysis of ***:A significant reduction was observed in the cell viability of 90%at a concentration of 400μg/mL compared to the *** comparison,a significant increase was reported in cell viability at concentrations of 25-200μg/mL(Pbax/BCL2gene ratio(Pbax/BCL2gene expression and ROS ***,the extract exerted an increased effect on cell viability in a concentration-dependent manner on AGO and increased cell growth at concentrations less than 400μg/mL,highlighting different effects of the whole extract on the AGO cell line.
