Comparison of the potential rate of population increase of brown and green color morphs of Sitobion avenae （Homoptera： Aphididae） on barley infected and uninfected with barley yellow dwarf virus

Life tables of brown and green color morphs of the English grain aphid, Sitobion avenae （Fabricius） reared on barley under laboratory conditions at 20 ＋ 1℃,65% ± 5% relative humidity and a photoperiod of 16 ： 8 h （L ： D） were compared. The plants were either：（i） infected with the barley yellow dwarf virus （BYDV）; （ii） not infectedwith virus but previously infested with aphids; or （iii） healthy barley plants, which were not previously infested with aphids. Generally, both color morphs of S. avenae performedsignificantly better when fed on BYDV-infected plants than on plants that were virus free but had either not been or had been previously infested with aphids. Furthermore,when fed on BYDV-infected plants, green S. avenae developed significantly faster and had a significantly shorter reproductive period than the brown color morph. There wereno significant differences in this respect between the two color morphs ofS. avenae when they were reared on virus-free plants that either had been or not been previously infestedwith aphids. These results indicate that barley infected with BYDV is a more favorable host plant than uninfected barley for both the color morphs ofS. avenae tested, particularly the green color morph.

The movement protein（MP） of barley yellow dwarf virus-GAV（BYDV-GAV） was previously considered to bind the viral RNA and transport viral genome,however,a new function of MP of BYDV-GAV on plant physiology was found in this *** Arabidopsis thaliana plants that constitutively expressed MP were produced and exhibited a late-flowering *** key proteins involved in a signal transduction pathway about flowering in Arabidopsis,vascular plant one-zinc protein 1（AtVOZl） and vascular plant one-zinc protein 2（AtVOZ2）,were found to interact with MP by yeast two-hybrid assay and bimolecular fluorescence complementation ***,N-terminal of the MP was confirmed to be essential for the *** result of quantitative real-time PCR（qRTPCR） revealed that MP up-regulated the expression ofAtVDZ1 and AtVOZ2 and also affected other genes in phyB signal transduction ***,our work indicated that MP of BYDV-GAV can delay flowering in Arabidopsis through interacting with AtVOZ1 and *** findings provided new insights into the molecular mechanisms by which MP influence plant growth and development.

Disruption of host mitosis plays an important role in viral pathogenesis However,the underlying molecular mechanisms are still not well ***,we report that the 17 K protein,conserved among the grass-infecting barley yellow dwarf viruses(BYDVs) and related poleroviruses,disrupts G2/M transition and inhibits cell growth in both host(barley) and nonhost(fission yeast and Arabidopsis thaliand)organisms through directly interrupting the mitotic entry switch Weel-Cdc25-Cdkl(CDKA in plant and Cdc2 in fission yeast).17 K binds to Cdc25 and CDKA/Cdc2 but not Weel,and decreases Cdc25’s phosphatase activity in ***,17 K expression up-regulates Weel *** alterations lead to abnormal accumulation of the CDKA/Cdc2 phosphorylated at critical mitosis-regulating residues,which impedes the function of CDKA/Cdc2 and results in G2/M ***,perturbing 17 K-CDKA interaction by silencing CDKA expression aggravates BYDV-induced dwarfing *** work suggests that the 17 K protein is an important contributor to BYDV-induced host *** the interaction between 17 K and Weel-Cdc25-CDKA may aid the control of cereal yellow dwarf ***,direct interruption of Weel-Cdc25-Cdkl is also being found for an increasing number of animal viral proteins,indicating commonality in the molecular mechanism underlying the disruption of mitosis by plant and animal viruses.
