Tobacco bushy top virus（TBTV）,a member of Umbravirus,contains a positive single genomic RNA with 4152 *** genome of TBTV lacked 5’ cap and 3’ poly（A） tail and encoded4 proteins including components of replicase（p35 and p63）,and movement proteins（p26 and p27） from5’to3’.For the cap-independent translation of p35,previous report showed the existence of a bte-like element in TBTV by using firefly luciferase（Flue） reporter *** confirmed the bte-like element by using in vitro translation of full-length TBTV in wheat germ extract（WGE）.Furthermore,we used forward and reverse genetics methods to identify new elements regulating the expression of *** expression of p35 among different TBTV isolates showing severe or mild pathogenicity had 3 fold ***,the regions altering the expression of p35 were mapped 5’ terminal 500 nt and3’terminal 200 nt located downstream of bte-like element by constructing chimeric ***,EMSA assay also implied the potential RNA-RNA interaction between 3’ terminal 200 nt and 5’ terminal200 nt,which may play role in the expression of *** the 3’ terminal 200 nt,the region of 4093 to 4152 nt played the essential role in the expression of p35 by analyzing the deletion mutants of *** a word,it is suggested that at least two cis-elements located 3’ terminal region were associated with the cap-independent translation of p35 and may both form RNA-RNA interaction with 3’ terminal region.
