To evaluate the preclinical safety of recombinant replicating vaccinia virus-based HIV vaccines （rVVV）in balb/c mice. The study was composed of groups 1, 2, 3, 4, wherein the groups 1 and 2 received only one priming immunization and the groups 3, 4 received prime-boost immunizations at a 6-week interval. The dose given was 50μl/skin of back/2 dosing point by intradermal injection approximately 2.2×10 6 PFU. cellular responses were evaluated in spleenocytes after restimulation on the following basis: detection of IFN-γ producing cD8 + -positive T cells, and cD4 + /cD8 + T cells ratio by FAcS assay. Humoral responses were evaluated in serum samples on the basis of specific anti-rVVV antibodies. A real-time quantitative PcR was employed for detection of biodistribution of rVVV in tissues and organs. The results showed that there were no significant toxic effects and damages to major organs in control and rVVV-treated animals. The results of FAcS analysis assay for the group 4 that received prime-boost immunization showed cellular immune responses, as indicated by high IFN-γ production by cD8 + -positive T cells. There was no significant change for the cD4 + /cD8 + T cells ratio in control and rVVV-treated animals. Immunogenicity of rVVV given with prime-boost doses was stronger than that of rVVV given with a single priming dose in balb/c mice. Following intradermal injection of rVVV in mice, all tissues were Q-PcR-negative on Day 35 after dosing. The study showed that there were no significant toxic effects and the rVVV could induce both humoral and cellular immune responses in balb/c mice.

＜正＞The impacts of astaxanthin enriched algal powder on auxiliary memory improvement were conducted by using balb/c mice daily which were pre-supplemented with different dosages of cracked green algal（Haematococcus pluvialis）powder for 30 days.The supplemented mice were first tested to find a hidden platform by swimming in Morris water maze for 8 days.Then,the mice were used to search a visible platform in Morris water maze for 5 days.After that,the mice were practiced to find a safety place,an insulated platform in a chamber for 2 days.During above animal experimental periods,similar algal meals containing astaxanthin of 0,0.26,1.3 and 6.4 mg/Kg of body weight were continuously fed to each group of the tested mice.The profiles of latency,the distances,the speeds and the direction angles to the platforms as well as the diving frequency in each group were measured and analyzed.The process of mice jumping up onto insulated platform and diving down to copper shuttered bottom with 36 voltages electrical charge were auto-video monitored too. The results of Morris maze experiment showed that middle dosage of H.pluvialis meals containing 1.3 mg astaxanthin/Kg body weight significanfly shortened the latency and distance of mice finding a hidden platform.However,the swim velocity had no obvious changes in each supplemented groups.However,the visible platform test showed a significant increase in the latency and swim distance,and a significant decrease in the swim speed for all groups of mice orally supplemented with H.pluvialis powder compared to the placebo group（P＜0.05 or P＜0.01）.The mice supplemented with algal meal hesitantly turned around the original hidden platform,in contract the mice supplement placebo easily forget the original location and accepted the visible platform as new safety place.Above results illustrated that astaxanthin enriched H.pluvialis powder has an auxiliary avail of memory improvement. Background:Experimental animals are used to study physiological phenomena,pathological mechanisms,and disease prevention.The gut microbiome is known as a potential confounding factor for inconsistent data from preclinical studies.Although many gut microbiome studies have been conducted in recent decades,few have focused on gut microbiota fluctuation among representative mouse strains.Methods:A range of frequently used mouse strains were selected from 34 isolation packages representing disease-related animal(DRA),immunity defect animal(IDA),or gene-editing animal(GEA)from the balb/c and c57BL/6J backgrounds together with normal mice,and their microbial genomic DNA were isolated from mouse feces to sequence for the exploration of gut microbiota.Results:Mouse background strain,classification,introduced source,introduced year,and reproduction type significantly affected the gut microbiota structure(pckground strain contributing the greatest influence(R^(2)=0.237).In normal groups,distinct gut microbiota types existed in different mouse strains.Sixty-four core operational taxonomic units were obtained from normal mice,and 12 belonged to Lactobacillus.Interestingly,the gut microbiota in c57BL/6J was more stable than that in balb/c mice.Furthermore,the gut microbiota in the IDA,GEA,and DRA groups significantly differed from that in normal groups(pcompared with the normal group,there was a significantly higher chao 1 and Shannon index(pchanged classes occurred with Firmicutes and Bacteroidetes.The abundances of Helicobacter,Blautia,Enterobacter,Bacillus,clostridioides,Paenibacillus,and clostridiales all significantly decreased in the IDA,GEA,and DRA groups,whereas those of Saccharimonas,Rikenella,and Odoribacter all significantly increased.

In vitro activity and in vivo efficacy of a combination therapy of diminazene and chloroquine against murine visceral leishmaniasis

The present study evaluated the in vitro activity and in vivo efficacy of diminazene combined with chloroquine as a potential drug against Leishmania donovani. Amphotericin B was used as a positive control drug. In vitro activity involved incubation of various drug concentrations with promastigotes or vero cells in culture before determination of parasite growth inhibition or cell death while in vivo evaluations involved infection of various mice groups with virulent L. donovani parasites and treatment with test drug compounds following disease establishment. Weight changes in experimental mice were also evaluated before infection and throughout the experiment. The results indicated that the diminazene-chloroquine combination was at least nine times more efficacious than individual drugs in killing promastigotes in culture. The diminazene-chloroquine combination was safer （Ld50=0.03±0.04） than Amphotericin B （Ld50=0.02±0.01）. Body weight in infected mice increased significantly （P=0.0007） from day 7 to day 37 following infection （P=0.026）. However, body weight remained comparable in all mice groups during treatment （P=0.16）. The diminazene-chloroquine combination significantly reduced splenic parasite numbers as compared to individual drug therapies （P=0.0001） although Amphotericin B was still more efficacious than any other treatment （P=0.0001）. Amongst the test compounds, the diminazene-chloroquine combination showed the lowest level of IgG antibody responses with results indicating significant negative correlation between antileishmanial antibody responses and protection against disease. These findings demonstrate the positive advantage and the potential use of a combined therapy of diminazene-chloroquine over the constituent drugs. Further evaluation is recommended to determine the most efficacious combination ratio of the two compounds.

Non-thermal Plasma Suppresses Bacterial colonization on Skin Wound and Promotes Wound Healing in mice

The present study evaluated the effect of non-thermal plasma on skin wound healing in balb/c mice.Two 6-mm wounds along the both sides of the spine were created on the back of each mouse（n=80） by using a punch biopsy.The mice were assigned randomly into two groups,with 40 animals in each group：a non-thermal plasma group in which the mice were treated with the non-thermal plasma;a control group in which the mice were left to heal naturally.Wound healing was evaluated on postoperative days（POD） 4,7,10 and 14（n=5 per group in each POD） by percentage of wound closure.The mice was euthanized on POD 1,4,7,10,14,21,28 and 35（n=1 in each POD）.The wounds were removed,routinely fixed,paraffin-embedded,sectioned and HE-stained.A modified scoring system was used to evaluate the wounds.The results showed that acute inflammation peaked on POD 4 in non-thermal plasma group,earlier than in control group in which acute inflammation reached a peak on POD 7,and the acute inflammation scores were much lower in non-thermal group than in control group on POD 7（P0.05）.The amount of granular tissue was greater on POD 4 and 7 in non-thermal group than in control group（P0.05）.The re-epithelialization score and the neovasularization score were increased significantly in non-thermal group when compared with control group on POD 7 and 10（P0.05 for all）.The count of bacterial colonies was 103 cFU/mL on POD 4 and 20 cFU/mL on POD 7,significantly lower than that in control group（109 cFU/mL on POD 4 and 1012 cFU/mL on the POD 7）（P0.05）.It was suggested that the non-thermal plasma facilitates the wound healing by suppressing bacterial colo-nization.

Neospora caninum immune mapped protein 1(NcIMP1) is a novel vaccine candidate against neosporosis

The Neospora caninum immune mapped protein 1(Nc IMP1) was identified as a membrane protein,and a previous study indicated that Nc IMP1 could be a promising vaccine candidate against neosporosis. In this study, the immune response and protection efficacy of Nc IMP1 were evaluated. The coding sequence of Nc IMP1 was inserted into the eukaryotic expression vector pc DNA3.1(+), resulting in the recombination plasmid pc DNAIMP1, which was used for the intramuscular immunization of balb/c mice. After immunization, the immune response was evaluated using a lymphoproliferative assay and cytokine and antibody measurements. Quantification of the cerebral parasite burden of mice challenged with 2106 N. caninum was performed 14 days after the last immunization. The results showed that the mice immunized with pc DNA-IMP1 developed a high level of specific antibody responses against recombinant Nc IMP1,with a mixed Ig G1/Ig G2 a response and a predominance of Ig G2 a production. The cellular immune response was associated with the production of IFN-γ, IL-2, IL-4 and IL-10 cytokines. The experiment was terminated 30 days p.i.,and the cerebral parasite burden in each mouse was assessed by quantitative PcR. The parasite burden was significantly reduced in the pc DNA-IMP1-vaccinated mice. These data suggest that IMP1 is a promising vaccine candidate against neosporosis.
