Noncanonical NF-kB family of transcription factors is involved in diverse biological processes, including dendritic cells(DCs) development, maturation and subsequent adaptive *** NF-kB disorder contributes to the pathogenesis of various autoimmunity and inflammatory ***, the underlying mechanism of noncanonical NF-kB termination during cell division and cell cycle process remains poorly *** transcriptomic and immunological approaches, we identify that protelysis fate of NIK is regulated by an E3 ligase CRL4, which was considered as a pivotal regulator of mitotic and S-phase ***4 promotes polyubiquitination and subsequent degradation of NIK during DC differentiation, but it is dispensable for TRAF3 degradation induced by ***2 deficiency facilitates NIK accumulation and RelB nucleus ***2 DC-conditional KO mice display IL-23 hyper-production and more sensitive to autoimmune *** work illustrates CRL4 is crucial for controlling NIK stability and highlight a unique mechanism that controls inflammatory diseases.
