Acoustic trauma-induced auditory cortex enhancement and tinnitus

There is growing evidence suggests that noise-induced cochlear damage may lead to hyperexcitability in the central auditory system(CAS)which may give rise to ***,the correlation between the onset of the neurophysiological changes in the CAS and the onset of tinnitus has not been well *** investigate this relationship,chronic electrodes were implanted into the auditory cortex(AC) and sound evoked activities were measured from awake rats before and after noise *** auditory brainstem response(ABR) was used to assess the degree of noise-induced hearing *** was evaluated by measuring gap-induced prepulse inhibition(gap-PPI).Rats were exposed monaurally to a high-intensity narrowband noise centered at 12 kHz at a level of 120 dB SPL for 1 *** the noise exposure,all the rats developed either permanent(>2 weeks) or temporary(<3 days) hearing loss in the exposed ear(s).The AC amplitudes increased significantly 4 h after the noise *** of the exposed rats also showed decreased *** post-exposure AC enhancement showed a positive correlation with the amount of hearing *** onset of tinnitus-like behavior was happened after the onset of AC enhancement.

Objective Blood oxygenation level-dependent functional magnetic resonance imaging (BOLD-fMRI) was used to study activation signals in the brain cortex evoked by tone stimulation in patients with tinnitus for its potential utility as an objective indicator of tinnitus. Methods BOLD-fMRI examination was conducted in 7 patients with chronic tinnitus and 15 control subjects. The activation signal in the brain cortex was recorded. Results Significant activation was found in temporal lobe in control subjects, with greater signal volume and intensity on the contralateral than ipsilateral auditory cortex (P auditory cortex.

The relationship of attention and sensory coding is complicated and is an area of active investigation. Previous studies have revealed that an animal’s behavioral state can play a crucial role in shaping the characteristics of neural responses in the auditory cortex（AC）. However, the behavioral modulation of auditory response has not been well studied on the brain areas outside AC. In this study, we investigated the attention effects on the neural activities in both the subcortical structures（amygdala and striatum） and AC of cats. Single-unit spike activities were recorded when cats were actively performing a tone detection task and when they were passively listening to the same set of pure-tone stimuli. We reported that, task engagement had different effects（enhancive or suppressive） on the tone-evoked neural response in different brain areas. Task engagement had stronger modulation effects on the amygdala and striatum than on the AC. More importantly, the neural activities in amygdala, which showed a longer tonedriven response latency were tended to be enhanced, while those in striatum, which had a shorter latency, were tended to be suppressed. In AC, there was still a tendency that auditory response was suppressed in the primary auditory cortex（A1） and enhanced in the posterior auditory field（PAF）. Our results suggest that top-down attention activates the neurons in the upstream areas of auditory hierarchy, and inhibits the auditory response in the downstream areas.

auditory steady-state responses（ASSRs） are electrophysiological responses entrained to the frequency and phase of a periodic auditory stimulus generated by auditory pathway and auditory cortex activity. ASSR has been suggested as a potential biomarker for schizophrenia, in which the power and phase-locking of ASSR are reduced. Rats with neonatal ventral hippocampal lesion（NVHL） have been widely used as a neurodevelopmental model of schizophrenia, which exhibits several behavioral, molecular and physiological abnormalities similar to schizophrenics. However, clear abnormalities of ASSR have not been reported in NVHL rats. The aim of the present study was to compare the ASSRs of adult NVHL rats versus adult sham rats. We recorded local field potential（LFP） evoked by 40 and 80 Hz click-trains through microelectrodes inserted into the primary auditory cortex（A1） and posterior auditory field（PAF）. Mean trial power（MTP） and phase-locking factor（PLF） were obtained from the spectrogram analysis of click-train evoked LFPs. Both MTP and PLF results suggest that A1 showed a strong and synchronizing ASSR, which was less affected by NVHL, while PAF showed a relatively weak and non-synchronizing ASSR, which was impaired by NVHL. Our results, for the first time, reveal that NVHL may influence the neural activities of primary and non-primary regions of auditory cortex in different ways.

Band-passed noise（BPN） stimuli are prevalent in communication sounds. Tuning of auditory neurons for bandwidth of BPN may be functionally important for the perception of these sounds. In awake mouse primary auditory cortex（A1）, we found that excitatory neurons exhibit decreasing, increasing or flat bandwidth tuning, and there is an enhancement of tuning strength from layer 4 to layer 2/3. On the other hand, parvalbumin（PV） positive inhibitory neurons had flat tuning. Sequential cell-attached and whole-cell voltage clamp recordings from the same cells revealed that the tuning properties of excitatory neurons were inherited from excitation they received, whereas in all neurons inhibition had flat tuning. Importantly, tuning strength of spike response was enhanced as compared with excitation. Our neuron modeling work demonstrate an important role of flat（untuned） inhibition in enhancing bandwidth tuning of spike responses. Our study, for the first time, elucidated synaptic mechanisms underlying bandwidth tuning of auditory cortical neurons.

How activity of sensory neurons leads to perceptual decisions remains a challenge to understand. Correlations between choices and single neuron firing rates have been found in visual, vestibular and somatosensory systems. In the auditory system, however, how neuronal activities correlate with auditory perception, for instance, the spatial perception, remains unclear. To investigate choice-related activity, we have recorded from auditory cortex（AC） and auditory thalamus（MGB） while animals performed a fine sound location discrimination task. We find that AC has similar discrimination thresholds as thalamic cells, and the most sensitive neurons have thresholds that are only twofold or threefold greater than perceptual thresholds. The fact that minimum thresholds measured in MGB are of the same order of magnitude as behavioral thresholds suggests that the MGB might determine the limit on our ability to discriminate small differences in sound location, with little noise added during downstream processing. We also tested the hypothesis that spatial tuning sharpens when an animal engages in an auditory task by comparing the neuronal threshold between behavior condition and non-behavior condition. Preliminary data indicated neuronal firing in both AC and MGB neuron were modulated by the task, reflecting a top-down control of dynamic spatial tuning.

＜正＞In natural acoustic environment,sound stimuli often arrive at the two ears in *** aim of the present study was to determine how a preceding sound influences the frequency tuning of neurons in the rat primary auditory cortex to a subsequent sound.A forward masking paradigm was used to determine the frequency receptive fields of rat auditory cortical neurons in quiet and in sequential sound *** frequency and the level of both the preceding sound stimulus and the subsequent sound stimulus were changed systematically,and the interstimulus intervals between the stimuli also *** frequency receptive fields of a cortical neuron were modulated dynamically by a preceding sound *** most neurons,if the level of a preceding sound stimulus was constant,the receptive fields were mostly influenced when the frequency of the preceding sound was at or near the characteristic frequency of the neuron;if the frequency of the preceding sound stimulus was fixed,the effect of modulation was increased with increasing sound stimulus *** both the frequency and the level of the preceding sound stimulus were fixed,the effect of modulation decreased with increasing interstimulus *** results of the present study suggest that the frequency tuning of auditory cortical neurons is plastic and dynamically modulated in a reverberant acoustical environment,and the degree of modulation depends on the contextual acoustical stream.
