ascorbic acid synthesis and transportation capacity in old laying hens and the effects of dietary supplementation with ascorbic acid

Background: Laying hens over 75 weeks of age commonly show great declines in immunity and production performance.It is unclear whether these declines can be relieved by supplementing with ascorbic acid(AA) in feed.Two trials were conducted to investigate the synthesis and metabolism of AA in layers of different ages and the effects of dietary supplemental AA on the performance and the immune and antioxidant statuses of 78 weeks old hens.Methods: In Exp.1,equal numbers(24 hens) of 35 weeks old(Young) and 75 weeks old(Old) layers were fed the same diet without AA supplementation for 4 weeks.In Exp.2,360 healthy 78 weeks old laying hens were randomly assigned to 4 treatments(basal diet supplemented with 0,0.25,0.5,or 1 g AA/kg diet) in an 8-week feeding trial.Results: The old hens tended to have decreased L-gulonolactone oxidase(GLO) synthase activity in the kidney and liver than that of the young hens(P = 0.07 and P = 0.05,respectively).Compared with the young hens,the old hens had lower hepatic antioxidant capacity allowing for the lower thioredoxin(TXN),thioredoxin reductase(TXNR) and cytochrome b5 reductase(CYB5 R) gene expression(P < 0.05),whereas increased sodium-dependent vitamin C transporter(SVCT) 1 expression levels in the ileum and kidney and enhanced splenic and hepatic AA concentrations(P < 0.05).Dietary supplementation with AA significantly decreased GLO enzyme activity but increased splenic AA concentration and anti-bovine serum albumin IgG levels(P < 0.05) and tended to increase CD4+T lymphocyte numbers(P = 0.06) in serum.Supplementation of 0.25 g AA/kg diet significantly increased hepatic total antioxidant capacity(T-AOC,P < 0.05) relative to the control group.Conclusions: Laying hens could synthesize AA in both the kidney and the liver,though the GLO enzyme activities were 100 times greater in kidneys than in livers.The old laying hens had greater absorption and reabsorption capacity and higher AA retention in some tissues that did the young hens.Dietary supplementation of AA can improve the health

BACKGROUND The current case report describes successful phacoemulsification with the aid of perioperative topical ascorbic acid(AA) in two patients with corneal endothelial disorders to prevent postoperative corneal endothelial decompensation.CASE SUMMARY Two eyes of two patients underwent phacoemulsification with pre-existing corneal endothelial disorders including Fuchs corneal endothelial dystrophy(Patient 1) and endotheliitis(Patient 2). Topical AA was applied to both patients at least one month before and after with a frequency of four times per day. After the surgery, both eyes improved best-corrected visual acuity(BCVA) and there was limited human corneal endothelial cell loss without signs of corneal endothelial decompensation, such as deteriorated BCVA or persistent corneal edema during the follow-up of at least two years.CONCLUSION Perioperative administration of topical AA may be an alternative therapy to the triple procedure in patients expecting to undergo cataract surgery.

Efficacy of 1.2L polyethylene glycol plus ascorbic acid for bowel preparations

BACKGROUND A low-volume polyethylene glycol(PEG) solution that combines ascorbic acid with PEG-based electrolyte solution(PEG-ASC) is gaining mainstream acceptance for bowel preparation due to reduced volume and improved taste.Although several reports showed that bowel preparation with PEG-ASC volume lower than 2.0 L with laxative agents could be an alternative to traditional preparation regimen, the cleansing protocols have not been fully investigated.AIM To evaluate the cleansing efficacy of 1.2 L PEG-ASC solution comparing with 2.0 L PEG electrolyte(PEG-ELS) for bowel preparations.METHODS A randomized, single-blinded, open-label, single-center, non-inferiority study was conducted. In total, 312 Japanese adult patients(aged > 18 years) who underwent colonoscopy were enrolled. Patients were randomly allocated to bowel lavage with either 1.2 L of PEG-ASC solution with at least 0.6 L of an additional clear fluid(1.2 L PEG-ASC group) or 2.0 L of PEG-ELS(PEG-ELS group). Then, 48 mg of sennoside was administered at bedtime on the day before colonoscopy, and the designated drug solution was administered at the hospital on the day of colonoscopy. Bowel cleansing was evaluated using the Boston Bowel Preparation Scale(BBPS). The volume of fluid intake and required time for bowel preparation were evaluated. Furthermore, compliance, patient tolerance,and overall acceptability were evaluated using a patient questionnaire, which was assessed using a visual analog scale.RESULTS In total, 291 patients(1.2 L PEG-ASC group, 148; PEG-ELS group, 143) completed the study. There was no significant difference in successful cleansing, defined as a BBPS score ≥ 2 in each segment, between the two groups(1.2 L PEG-ASC group, 91.9%; PEG-ELS group, 90.2%; 95%CI:-0.03-0.09). The required time for bowel preparation was significantly shorter(164.95 min ± 68.95 min vs 202.16 min± 68.69 min, P < 0.001) and the total fluid intake volume was significantly lower(2.23 L ± 0.55 L vs 2.47 L ± 0.56 L, P < 0.001) in the 1.2 L PEG-ASC group than in the

In-Situ Microscopic FTIR Spectroelectrochemistry of ascorbic Acid in Poly(ethylene glycol)/LiClO_4 Electrolyte Paste and in the Presence of Dispersed Cobalt Hexacyanoferrate Microcrystalline Powder

In-situ microscopic FTIR spectroelectrochemical technique(MFTIRs) was applied to studying the electrochemical oxidation of ascorbic acid(AA) in poly(ethylene glycol)(PEG) paste at a 100 μm diameter Pt disk electrode. Using this technique, the catalytic ability of cobalt hexacyanoferrate(CoHCF) microcrystalline toward AA oxidation was also studied. It was found that the dispersed CoHCF powder in the PEG paste can generate well shaped thin layer cyclic voltammetric waves with the peak height proportional to the scan rate, corresponding to the Fe centered redox reactions. This oxidation step catalyzed the AA oxidation. Also, this pasted CoHCF powder generated well resolved in situ MFTIRs spectra, by which a chemical interaction between CC bond of AA ring and CoHCF lattice was revealed. A corresponding surface docking mechanism for the catalytic reaction has been proposed.

Sepsis and septic shock remain a major cause of morbidity and mortality among patients admitted in the intensive care unit.Diabetes is a major risk factor for the development of sepsis.The global mortality of sepsis remains high,despite significant interventions and guidelines.It has been known for decades that patients with sepsis have reduced levels of antioxidants,most notably vitamin C.Furthermore,experimental data has demonstrated multiple beneficial effects of vitamin C in sepsis.In addition,corticosteroids and thiamine may have synergistic biological effects together with vitamin C.Preliminary data suggests that therapy with hydrocortisone,ascorbic acid and thiamine improves the outcome of patients with sepsis with the potential to save millions of lives.However,this intervention has met with much resistance and has not been widely adopted.Ultimately,we await the final jury verdict on this simple,safe and cheap intervention.

ascorbic acid and curcumin alleviate abnormal estrous cycle and morphological changes in cells induced by repeated ultraviolet B radiations in female Wistar rats

Objective: To study the protective effect of ascorbic acid and curcumin against the abnormal estrous cycle and morphological changes in cells induced by repeated ultraviolet B(UVB)radiation in female Wistar rats.Methods: Sixteen female sexually mature Wistar rats weighing 130-150 g and aged 12-16 weeks were randomly divided into four groups.The control group received normal food and water ad libitum.The UVB group was exposed to a dose of 280 nm of UVB radiation for 2 h daily.The UVB+curcumin group received a dose of 280 nm of UVB radiation for 2 h daily and also an oral dose of curcumin(25 mg/kg body weight)daily.The UVB+ascorbic acid group received a dose of 280 nm of UVB radiation for 2 h daily and also an oral dose of ascorbic acid(250 mg/kg body weight)daily.All the treatments last for 15 consecutive days.Body and ovary weight and gonadosomatic index were measured.The stages(proestrus,estrus,metaestrus and diestrus)of the estrous cycle were determined by the cell types observed in the vaginal smear.Results: UVB radiation caused irregular alterations on the estrous cycle and morphological changes of the female Wistar rat as compared with the control group.ascorbic acid and curcumin protected UVB-induced estrous phases and their cells.But curcumin showed greater protection than ascorbic acid.Conclusions: ascorbic acid and curcumin at low doses can alleviate abnormal estrous cycle and morphological changes in cells induced by UVB radiations in female Wistar rats.

RADIATION EFFECT ON BIOSYNTHESES OF ascorbic ACID AND RIBOFLAVIN IN GERMINATING CHICKPEA

The influence of irradiation on biosyntheses of ascorbic acid and riboflavin in germinating chickpea seeds at ambient (25-35℃) conditions, was investigated. The rate of syntheses of these vitamins significantly increased with increasing germination time upto 120 h depending upon the treatment (Pascorbic acid, 22.32 and 16.84 mg/100g, were found in the 0.10 kGy sample after 120 h of germination in tap and distilled water respectively. However, a radiation dose of 0.15 kGy resulted in the development of maximum values of riboflavin, 11.40 and 11.38 μ g/g, on germination in tap and distilled water respectively. A significant linear relation (r = 0-954 to 0.997) was observed between the biosyntheses of these vitamins and the germination time upto 120 h of irradiated and unirradiated chickpea seed (P<0.05).

Effect of shading on ascorbic acid accumulation and biosynthetic gene expression during tomato fruit development and ripening

The purpose of the current study was to determine the effect of leaf shading,fruit shading,and a combination of both,on the accumulation of ascorbic acid(AsA)and the expression levels of AsA biosynthetic genes at the immature green,mature green,breaker,and red ripe stages of Ailsa craig tomato during fruit development.Shading(72%reducing of light intensity)imposed on the leaves significantly reduced AsA content and AsA biosynthetic gene expression in the fruits.Leaf shading,fruit shading,and a combination of both significantly decreased the amount of total AsA and reduced AsA to a range of 18.5%−31.5%at mature green,breaker,and red ripe stages of tomato fruits,with no significant change at the immature green stage of fruits.Moreover,reducing the light intensity in tomato leaves,fruits or both resulted in reduced expression of most AsA biosynthetic genes in the fruits,except for PMM,cAPX,tAPX,and APX7 genes under leaf shading,GPI,PMI,PMM,GP1,GP2,cAPX,and tAPX genes under fruit shading,and PMM,cAPX,APX1,and APX7 genes under both shading.The expression level of GMP,GP1,and GalDH showed an upregulation at the red ripe stage in fruits with leaf shading,and also an up-regulation at the immature green and red ripe stages with both shading.Furthermore,positive correlations between expression of AsA biosynthetic genes and AsA accumulation were recorded under leaf shading,fruit shading,and both types of shading,while a negative correlation was recorded under normal conditions without shading.

ascorbic Acid Attenuates Acute Ethanol-Induced Liver Injury in SMP30 Knockout Mice

ascorbic acid (AA) is recognized as a free radical scavenger that protects cells from oxidative stress-induced damage. However, no studies have investigated the role of AA in acute alcoholic liver disease using senescence marker protein-30 (SMP30) knockout (KO) mice. SMP30 is a novel 34-kDa protein involved in AA biosynthesis. The present study aimed to elucidate the physiological functions of AA in acute ethanol-induced liver injury using SMP30 KO mice, which cannot synthesize AA in vivo. After a 4-week experimental period, mice were divided into six groups. The following three groups comprised the ethanol treatment groups: WT-E group (wild-type), KV-E group (AA-supplemented), and KT-E group (AA-deficient). Mice were exposed to an acute dose of ethanol (6 g ethanol/kg) administered by gavage once a day for three days. The other three control groups, namely, WT-C, KV-C, and KT-C control groups, received an equal volume of water via oral administration. Analysis of changes in body weight showed that mice in the KT-E group had significant loss of body weight compared to the control, KV-E, and WT-E groups. Behavioral analysis revealed that alcohol exposure significantly increased alcohol sensitivity in the KT-E group, whereas the WT-E, KV-E, and control groups developed ethanol tolerance. Aspartate transaminase (AST) levels in the KT-E group were significantly higher than those in the control, KV-E, and WT-E groups. The number of large and binucleated hepatocytes was significantly higher in the KT-E group than in the KV-E and WT-E groups. In addition, cytochrome P450 2E1 (CYP2E1) was over expressed in the central vein in the KT-E group when compared to the KV-E and WT-E groups. Our current findings indicate that AA supplementation in SMP30 KO mice can alleviate alcohol-induced liver damage by down regulating CYP2E1 expression. These results suggest that reduced CYP2E1 expression is a novel mechanism responsible for AA-induced reduction of ethanol-mediated oxidative stress.

Cytoprotective effects of amifostine,ascorbic acid and N-acetylcysteine against methotrexate-induced hepatotoxicity in rats

AIM:To investigate the potential role of oxidative stress and the possible therapeutic effects of N-acetyl cysteine(NAC),amifostine(AMF)and ascorbic acid(ASC)in methotrexate(MTX)-induced hepatotoxicity.METHODS:An MTX-induced hepatotoxicity model was established in 44 male Sprague Dawley rats by administration of a single intraperitoneal injection of20 mg/kg MTX.Eleven of the rats were left untreated(Model group;n=11),and the remaining rats were treated with a 7-d course of 50 mg/kg per day NAC (MTX+NAC group;n=11),50 mg/kg per single dose AMF(MTX+AMF group;n=11),or 10 mg/kg per day ASC(MTX+ASC group;n=11).Eleven rats that received no MTX and no treatments served as the negative control group.Structural and functional changes related to MTX-and the various treatments were assessed by histopathological analysis of liver tissues and biochemical assays of malondialdehyde(MDA),superoxide dismutase(SOD),catalase,glutathione(GSH)and xanthine oxidase activities and of serum levels of aspartate aminotransferase,alanine aminotransferase,alkaline phosphatase and total bilirubin.RESULTS:Exposure to MTX caused structural and functional hepatotoxicity,as evidenced by significantly worse histopathological scores[median(range)injury score:control group:1(0-3)vs 7(6-9),P=0.001]and significantly higher MDA activity[409(352-466)nmol/g vs 455.5(419-516)nmol/g,P<0.05].The extent of MTX-induced perturbation of both parameters was reduced by all three cytoprotective agents,but only the reduction in hepatotoxicity scores reached statistical significance[4(3-6)for NAC,4.5(3-5)for AMF and 6(5-6)for ASC;P=0.001,P=0.001 and P<0.005vs model group respectively].Exposure to MTX also caused a significant reduction in the activities of GSH and SOD antioxidants in liver tissues[control group:3.02(2.85-3.43)μmol/g and 71.78(61.88-97.81)U/g vs model group:2.52(2.07-3.34)μmol/g and 61.46(58.27-67.75)U/g,P<0.05];however,only the NAC treatment provided significant increases in these antioxidant enzyme activities[3.22(2.54-3.62)μmol/g and 69.22(61.13-1
