Insulin receptor participates in the peripheral olfactory processes of honey bees (apis cerana cerana)

Insulin receptors(InR)are an integral component of the insulin/insulin-like growth factor signaling pathway,which plays a vital role in insect development,lifespan,reproduction,and olfactory ***,whether InR participate in the periph-eral olfactory system of insects remains ***,we found that 2-heptanone(2-HT)affects AcerlnR expression,the gene for an InR protein,in apis cerana *** then examined the spatiotemporal expression profile of the gene in *** *** mRNA of AcerlnR was primarily expressed in the antennae,wings,and legs of forager bees,which are probable chemosensory *** results of fluorescence competitive binding assays,combined with site-directed mutagenesis,demonstrated that AcerOBP6 and AcerOBP14 exhibit strong binding affinities to ***,after foragers were fed with double-stranded AcerlnR,the expression levels of AcerOBP6 and AcerOBP14 decreased significantly,as did the electroantennogram responsiveness to 2-HT and some other *** conclusion,our findings provide a foundation for understanding the in-volvement of AcerlnR in the odor perception of *** ***,they offer novel insights into the olfactory recognition mechanism in insects.

The mitogen-activated protein kinase(MAPK)cascade pathway plays an important role in regulating stress *** function of the c-Jun NH_(2)-terminal kinase(JNK),a component of the MAPK cascade pathway,in apis cerana cerana(Acc)remains ***,JNK was isolated and identified from *** analyses revealed there is a typical serine/threonine protein kinase catalytic domain in the AccJNK *** expression profile analysis showed that AccJNK was significantly induced by pesticide *** further explore the functional mechanisms of AccJNK,a yeast 2-hybrid screen was performed,activator protein-1(AP-1)was screened as the interaction partner of AccJNK,and the interaction relationship was further verified by pull-down *** real-time polymerase chain reaction showed the expression pattern of AccAP-I was similar to that of *** a knockdown of AccJNK or AccAP-I by RNA interference,the survival rate of Acc after pesticide treatments ***,the expression levels of antioxidant-related genes and the activities of antioxidant enzymes increased,suggesting that the knockdown of AccJNK or AccAP-I increased the antioxidant capacity of *** study revealed that the JNK-mediated MAPK pathway responds to pesticide stress by altering the antioxidant capacity of Acc.

Cloning and Sequence Analysis of cDNA Encoding MRJP3 of apis cerana cerana

By screening the worker （apis cerana cerana） heads cDNA library using a fragment of the mrjp3 gene of apis cerana as probe, 120 positive clones were obtained. The clone containing A. cerana cerana MRJP3 （AccMRJP3） cDNA was selected. Based on the sequencing of the inserts of the positive clone, a sequence of AccMRJP3 cDNA which is 1 887 bp long including a poly （A） tail was obtained. The AccMRJP3 cDNA encompassed an open-reading frame （ORF） with 1 779 bp encoding 593 amino acids. The un-translated regions （UTR） of the 5′ end and 3′end are 46 bp and 160 bp in length, respectively. Similar to AmMRJP3 and AdMRJP3, the putative AccMRJP3 also has a repetitive region. The comparison of the repetitive region of AccMRJP3, AmMRJP3 and AdMRJP3 shows some differences between them.

N-terminal asparagine amidohydrolase(Ntan1) is a feature of the ubiquitin-dependent N-end rule pathway of protein degradation which has been implicated in a legion of physiological functions: sensing of heme,oxygen,and nitric oxide;selective elimination of misfolded proteins,DNA *** identified the AccNtan1 gene from apis cerana cerana and investigated its role in oxidation *** sequence alignments and phylogenetic analysis revealed that AccNtan1 is highly conserved in insect *** results indicated that the expression levels of AccNtan1 in wing,honey sac and abdomen were significantly lower than in other tissues and significantly higher in early development stages,including larve,prepupae and pink-eyed *** further observed that the expression of AccNtan1 was induced by several environmental ***,disc diffusion assays result suggested that overexpression of AccNtan1 enhances the resistance of bacteria to oxidative *** results indicated that AccNtan1 may be involved in the response to oxidative ***,after knockdown of AccNtan1 using RNA interference(RNAi),the expression levels of most antioxidant genes and antioxidant enzymatic activities were significantly ***,we hypothesize that AccNtan1 plays important roles in environmental stress responses and anti-oxidative processes.

Pollinators, most often honeybees, play essential roles in maintaining healthy ecosystems and in supporting agricultural production and food security. Compared to the worldwide distribution of apis mellifera ligustica(Aml), apis cerana cerana(Acc) is indigenous to Asian distributes across variety of physiographic and climatic zones. Although closely related, Aml and Acc have some distinct characteristics in behavior. To reveal how neuropeptides are involved in the regulation of behavioral differences between Aml and Acc adult worker bees, an in-depth comparison of the brain neuropeptidomic dynamics was performed over three time points. In total, 132 nonredundant neuropeptides derived from 23 precursor proteins were identified in Aml. In Acc, 116 nonredundant neuropeptides derived from 22 precursor proteins were identified for the first time which provide novel neuropeptidomic information of Acc. The fact that 15 identical neuropeptide families changed their expression levels during the ontogeny of adult workers in both honeybee species suggests the two species of bees employ a similar neuropeptidome modification to tune their respective physiology. Noticeably, the elevated abundance of FMRFamide and myosuppressins in Acc foragers demonstrate neuropeptidic regulation enhance musculature governing to facilitate the rapid and hasty flight pattern of Acc. The up-regulated diuretic hormone and CAPA peptides in Aml nurse bees signify the fact that neuropeptides are also involved in the regulation of royal jelly secretion. These findings indicating Aml and Acc have evolved unique neuropeptidomic settings to accommodate their particular biological needs. Our research gain novel understanding of honeybee neuropeptidome correlated with physiology and behavior regulation, which is potentially important in neurobiology for honeybees and other insects.

Geographic isolation interrupted gene flow between populations leading to population differentiation during the long evolutionary *** this paper,33 colonies from Damen Island and 100 colonies from adjacent mainland populations,Juxi and Chixi,were analyzed with both mitochondrial tRNA-COII sequences and five microsatellite *** results showed that apis cerana cerana population from Damen Island significantly differentiated from its adjacent mainland *** addition,Damen island population showed a lower level of genetic diversity in terms of the number of mitochondrial haplotypes while both island and mainland populations showed a low level of genetic diversity with mutilocus *** divergent small island population *** might probably have suffered inbreeding and genetic drift as well as limited gene flow across the *** data provides useful information for management and preservation for the Damen Island population.

Myclobutanil is currently used on the flowering *** is known about how apis cerana cerana respond to myclobutanil ***,the acute toxicity of myclobutanil and its sublethal effects on respiration,flight and detoxification enzymes [7-ethoxycoumarin O-deethylase(ECOD) and glutathione S-transferases(GSTs)] in *** cerana were *** results indicated that formulation grade myclobutanil showed moderate toxicity to *** cerana either contact(LD=4.697μg/bee) or oral(LD=2.154 μg/bee) *** dose of myclobutanil significantly reduced the respiration rate of workers at 24 h and 48 h regardless of the exposure ***,myclobutanil didn’t significantly affect the take-off *** nurse bees exposure to the dose(LD) of formulation-grade myclobutanil,ECOD activity was significantly induced when compared with control,but GST activity didn’t *** the forager bees,no enzyme markers response was obtained in this *** results suggested that myclobutanil disturbed respiration and P450-mediated detoxification of the individual bees of *** ***,myclobutanil has risk for cerana,it should be cautiously used.

Antifreeze proteins(AFPs) are biological cryoprotectants with unique properties that play a crucial role in regulating the molecular mechanisms governing cold resistance in *** identify and characterize apiscerana cerana AFP(AcerAFP),we cloned the full-length cDNA of AcerA FP and examined its expression patterns in *** cerana.A nucleotide alignment analysis showed that the entire coding region of AcerAFP is 1095 bp and encodes a polypeptide of 365 amino *** amino acid sequence of this protein exhibits 63% to 96% homology with AFP homologs from other ***-helices form the main secondary and tertiary structures of AcerAFP,which is similar to the molecular structure of fish AFP *** expression profiles of AcerAFP revealed that expression was tissue,sex,and developmentally *** response to cold stress,the mRNA and protein expression of AcerA FP were both induced by low temperatures,and were also related to the concentrations of several cryoprotectants,including glucose,glycerin,glutamic acid,cysteine,histidine,alanine,and *** addition,we found that the knockdown of AcerAFP by RNA interference remarkably increased the total freezing temperature of hemolymph in apis cerana cerana,where levels of AcerAFP mRNA were correlated with the expression of most antifreeze-related *** together,these results suggest that AcerAFP plays an essential role as a biological cryoprotectant in honeybees,and is in turn regulated by small cryoprotectants and antifreeze-related proteins.

Cytochrome P450 s(P450 s) plays an important role in maintaining redox homeostasis and in protecting organisms from the accumulation of toxic reactive oxygen species(ROS).In this study,we sequenced and characterized a P450 s gene(AccCYP4 AV1)from apis cerana cerana,playing a critical role in maintaining *** open reading frame(ORF) of Acc4 AV1 is 1506 bp in length and encodes a predicted protein of 501 amino acids,57.84 kDa and an isoelectric point of 8.67.A quantitative RT-PCR analysis indicated that Acc4 AV1 was expressed higher in midgut than other tissues,with its highest expression occurring on the fifteenth day of the adult stage,followed by the first day of the larval ***,the expression of the Acc4 AV1 transcript was up-regulated by cold(4℃),ultraviolet radiation,H O,paraquat,and dichlorovos *** contrast,AccCYP4 AV1 transcription was down regulated by other abiotic stresses,including 44 ℃,deltamethrin,CdCl,and *** results were confirmed by the western blot ***,the recombinant AccCYP4 AV1 protein acted as a potent antioxidant that resisted paraquat-induced oxidative ***,when the AccCYP4 AV1 was knocked down by RNA interference,we observed that multiple oxidant genes(AccHSP22.6,AccSOD,AccTpxl,and AccTpx4) were down-regulated in the knockdown samples,and other genes(AccGST and AccTrx1) were ***(POD) and catalase(CAT) activity levels were up-regulated in the knockdown samples compared to the control *** findings suggest that the AccCYP4 AV1 protein is an antioxidant enzyme that may play a very significant role in antioxidant defense against ROS damage.
