Cyclic dimeric GMP(c-di-GMP)has emerged as the nucleotide second messenger regulating both development and antibiotic production in high-GC,Gram-positive ***,a diguanylate cyclase(DGC),CdgD,encoded by SCO5345 from the model strain Streptomyces coelicolor,was functionally identified and characterized to be involved in c-di-GMP synthesis through genetic and biochemical *** overexpression resulted in significantly reduced production of actinorhodin and undecylprodigiosin,as well as completely blocked sporulation or aerial mycelium formation on two different solid *** the cdgD-overexpression strain,intracellular c-di-GMP levels were 13-27-fold higher than those in the wild-type *** vitro enzymatic assay demonstrated that CdgD acts as a DGC,which could efficiently catalyze the synthesis of c-di-GMP from two GTP *** overproduction of cdgD in two industrial Streptomyces strains could similarly impair developmental transitions as well as antibiotic ***,our results combined with previously reported data clearly demonstrated that c-di-GMP-mediated signalling pathway plays a central and universal role in the life cycle as well as secondary metabolism in streptomycetes.

As the diagnostic criteria for Kawasaki disease (KD) consist of the clinical features that are shared by other childhood febrile illnesses [1, 2], KD is sometimes confused with a bacterial infection, leading to clinical situations in which children with the disease receive antibiotics (3–5)Here, we report the extent of antibiotic use in children with KD and the clinical parameters confusing KD with a bacterial infection.

Streptococcus agalactiae is one of the most common pathogens that cause bovine mastitis worldwide. Identifying pathogen prevalence and virulence factors is critical for developing prevention and control approaches. Herein, 1 161 milk samples from various dairy farms in China(n=558) and Pakistan(n=603) were collected between 2019–2021 and were subjected to S. agalactiae isolation. Prevalence, serotyping, virulence genes, and antibiotic-resistant genes of S. agalactiae were evaluated by PCR assay. All isolates were characterized for haemolysis, biofilm production, cytotoxicity, adhesion, and invasion on bovine mammary epithelial cells. The prevalence of S. agalactiae-induced mastitis in cattle was found to be considerably higher in Pakistan than in China. Jiangsu and Sindh provinces had the highest area-wise prevalence in China and Pakistan, respectively. Serotypes Ia and II were prevalent in both countries, whereas serotype III was found only in Pakistan. Moreover, all isolates tested positive for PI-2b gene but negative for PI-1 and PI-2a genes. All isolates harboured cfb, cylE, hylB, and fbsB virulent genes, whereas many of them lacked bibA, rib and bca. However, the absence of bac and scp genes in Chinese isolates and cspA in Pakistani isolates was noted, while spb1 and lmb were not detected in isolates of both countries. Pakistani isolates, particularly serotype Iapositive, had a considerably higher ability to produce biofilm, haemolysis, cytotoxicity, adhesion, and invasion than Chinese isolates. Most of the isolates were phenotypically resistant to tetracycline, erythromycin, and clindamycin and genotypic resistance was confirmed by the presence of ermA, ermB, tetM and tetO genes. Our study highlights the antimicrobial resistance profile and virulence-related factors contributing to the epidemiological spread of mastitis-causing S. agalactiae in China and Pakistan. Microdialysis is a technique used to measure the unbound antibiotic concentration in the interstitial spaces, the target site of action. In vitro recovery studies are essential to calibrating the microdialysis system for in vivo studies. The effect of a combination of antibiotics on recovery into microdialysate requires investigation. In vitro microdialysis recovery studies were conducted on a combination of vancomycin and tobramycin, in a simulated in vivo model. Comparison was made between recoveries for three different concentrations and three different perfusate flow rates. The overall relative recovery for vancomycin was lower than that of tobramycin. For tobramycin, a concentration of 20μg/mL and flow rate of 1.0μL/min had the best recovery. A concentration of 5.0μg/mL and flow rate of 1.0μL/min yielded maximal recovery for vancomycin. Large molecular size and higher protein binding resulted in lower relative recoveries for vancomycin. Perfusate flow rates and drug concentrations affected the relative recovery when a combination of vancomycin and tobramycin was tested. Low perfusate flow rates were associated with higher recovery rates. For combination antibiotic measurement which includes agents that are highly protein bound, in vitro studies performed prior to in vivo studies may ensure the reliable measurement of unbound concentrations.

In this article, antibiogram and heavy metal resistance profile of bacteria isolated from giant freshwater prawn （Macrobrachium rosenbergii） hatchery in Malaysia are described. Although giant freshwater prawn was introduced into Malaysia since the 1980s, there was no database information on antibiogram and heavy metal resistance profile of bacteria from giant freshwater prawn （34. rosenbergii） hatchery in Malaysia. Therefore, this study was carried out to determine the effectiveness of antibiotic and heavy metal resistance profile to control bacterial diseases in M. rosenbergii hatchery. The results can provide valuable information for local M. rosenbergii post-larval producer. antibiotic sensitivity test was carried out by disk-diffusion method against 15 types of antibiotics as follows： oxolinic acid （2 μg）, ampicillin （10μg）, erythromycin （15μg）, furazolidone （15 μg）, lincomycin （15μg）, amoxicillin （25 μg）, colistin sulphate （25 μg）, doxycycline （30μg）, florfenicol （30 μg）, flumequine （30 lag）, nalidixic acid （30μg）, tetracycline （30μg）, oleandomycin （15μg）, fosfomycin （50μg）, and spiramycin （100 μg）, whereas heavy metal resistance profile of the present bacterial isolates was determined by 2-fold agar dilution technique. In this study, 5 types of bacteria were successfully isolated; they were Aeromonas spp. （n = 77）, Escherichia coli （n = 73）, Edwardsiella spp. （n = 62）, Salmonella spp. （n = 75）, and Vibrio spp. （n = 43）. The result showed that furazolidone was the most effective antibiotic to control the bacteria isolated in this study, approximately 89.7% of the bacterial isolates were sensitive to this antibiotic. Multiple antibiotic resistance （MAR） index indicated that the hatchery water source and M. rosenbergii post-larval and sediment tanks were at high-risk exposure to the tested antibiotic. Furthermore, all the tested heavy metals （Cd2＋, Cr6＋, Hg2＋, and Cu2＋） failed to inhibit the growth of the bacterial isolates. Therefore, it indicated that the water

The rationale for the antibiotic treatment of viral community-acquired pneumonia(CAP)in adults was analyzed to develop a clinical reference standard for this *** data from 166 patients diagnosed with viral pneumonia across 14 hospitals in Beijing from November 2010 to December 2017 were *** indications for medications were evaluated,and the rationale for the use of antibiotics was analyzed.A total of 163(98.3%)patients with viral pneumonia were treated with antibiotics.A combination of C-reactive protein(CRP)and procalcitonin(PCT)was used as markers to analyze the possible indications for antibiotic *** threshold levels set at 0.25µg/L for PCT and 20 mg/L for CRP,the rate of unreasonable use of antibiotics was 55.2%.By contrast,at a CRP level threshold of 60 mg/L,the rate of antibiotic misuse was 77.3%.A total of 39 of the 163(23.9%)patients did not meet the guidelines for drug selection for viral CAP in *** unreasonable use of antibacterial drugs for the treatment of viral CAP in adults is a serious *** must reduce the unnecessary use of antibiotics.

Emerging antibiotic resistance is a major global health threat. The analysis of nucleic acid sequences linked to susceptibility phenotypes facilitates the study of genetic antibiotic resistance determinants to inform molecular diagnostics and drug development. We collected genetic data (11,087 newly-sequenced whole genomes) and culture-based resistance profiles (10,991 out of the 11,087 isolates comprehensively tested against 22 antibiotics in total) of clinical isolates including 18 main species spanning a time period of 30 years. Species and drug specific resistance patterns were observed including increased resistance rates for Acinetobacter baumannii to carbapenems and for Escherichia coli to fluoroquinolones. Species-level pan-genomes were constructed to reflect the genetic repertoire of the respective species,including conserved essential genes and known resis-tance factors. Integrating phenotypes and genotypes through species-level pan-genomes allowed to infer gene–drug resistance associations using statistical testing.
