tumor immunotherapy,particularly cancer vaccines,holds promise for combating cancer by harnessing tailored immune responses against malignant ***,conventional approaches face challenges in efficiently delivering antigens for optimal immune *** adjuvants,like MF59,enhance cellular uptake but struggle to induce robust CD8^(+)T cell ***,we introduce a novel strategy employing a water-in-oil-in-water(W/O/W)multiple Pickering emulsion(mPE)for antigen *** mPE,utilizing biocompatible,pH-sensitive particles,encapsulates antigens within the inner water phase,ensuring enhanced intracellular processing and dictating the intracellular fate of antigens for improved *** vitro and in vivo studies demonstrated that mPEs induced robust dendritic cells activation and antigen cross-presentation,leading to a significantly enhanced immune ***,calcium phosphate-stabilized mPE(CaP-mPE)illustrated the more robust IFN-γ^(+)T cell *** comparison with traditional surfactant-stabilized multiple emulsions,CaP-mPE significantly inhibit tumor growth and effectively prolong the survival of tumor-bearing *** innovative approach offers a promising avenue for the development of effective cancer vaccines with potent cellular immune responses.

The paracaspase Malt1 is a key molecule in mediating antigen receptor-induced nuclear factor kB(NF-kB) activation in *** it has been well studied in conventional T cells and adaptive immune response, the role of Malt1 in Treg cells function is still ***, we report that specific deletion of Malt1 in Treg cells would lead to Scurfy-like lethal autoimmune disease, which was caused by Treg cells dysfunction but not number ***, Foxp3Malt1 mice, in which Malt1 protease is specifically inactivated in Treg cells, also displayed spontaneous inflammatory disorders, with severe hair loss and skin ***, Foxp3Malt1 mice showed enhanced anti-tumor response due to their decreased Treg cells function and infiltration, as well as reduced CD8 T cells exhaustion, in the tumor *** expression profiling results revealed dysregulated expression pattern of Treg effector genes upon Malt1 deletion or its protease inactivation, which was due to dampened TCR-induced tonic and NF-kB signaling in Treg ***, our data unraveled a critical role of Malt1, especially its protease activity, in maintaining Treg cells homeostasis and function.

Age and obesity are two major global problems that significantly increase the incidence of *** intra-tumor exhausted CD8+T cells(Tex) due to chronic cancer stimulation,are major obstacles to anti-tumor *** and obesity affect Tex and a better understanding of the heterogeneity of these cells is necessary to improve tumor immune responses and to increase the efficacy of immune checkpoint blockade(ICB) *** fat diet(HFD) promoted tumor growth only in relatively old mice,even though the young and old mice had similar obesity *** HFD-fed old mice but not in young mice,increased tumor growth was associated with the increased numbers of Tex expressing PD-1 and *** old obese mice,reduced numbers of stem-like Tex expressingTCF1 and increased numbers of effector-like Tex lacking TCF1 were shown to remodel the heterogeneity of Tex *** results collectively indicate age and obesity remodel the heterogeneity of Tex.
