The fruiting body of Ganoderma lucidum（GL） has been used as a folk medicine in the oriental country for a long time. For the past few years, the triterpenes extract from GL has been identified as an important active ingredient which possessing anti-cancer activities and many other pharmacological activities. More than 160 kinds of triterpenes had been separated form GL. However, the study and application of triterpenes were limited by the low content and hard obtainment. In order to found some methods to solve these problems, we make some efforts in our study. Firstly, supercritical CO2 extraction（SFE-CO2）was applied for extract triterpenes from the fruiting body of GL, and then anti-prostate cancer activity was used as an tracking means for separation and purification of triterpenes from GL. After that, we also had studied the mechanism of anti-prostate cancer activity of active ingredients. Single parameter and orthogonal tests were used to determine the optimal conditions for extracting triterpenes from the fruiting body of GL using SFE-CO2. Highest extraction rates were achieved when the fruiting bodies of GL were pulverized to 10 mesh particle size, an entrainer（ethanol） concentration of 95%, 35 Mpa extraction pressure, 40 ℃, 2.5 h extraction time, a carbon dioxide flow rate of 35 g/min, and an entrainer: The fruiting body of GL weight ratio of 6:1（mL :g）. Under these conditions, the yield of crude extract was 2.42 g and the triterpene content of the crude extract was 40.5%, giving a triterpene yield of 0.98%. Crude extract and triterpene yields obtained using SFE-CO2 were lower compared with levels obtained using ethanol extraction but the former extracts contained a higher percentage and a greater number of triterpenes. The comparison on bioactivities of two sorts of extracts from the fruit body of GL obtained from SFE-CO2 and ethanol extraction separately was investigated antitumor effect in vitro.
