anti-obesity and antioxidant activity of dietary flavonoids from Dioscorea steriscus tubers

Objective:To investigate antioxidant and anti-obesity activity of flavonoids extracted by solvent cold percolation and preparative thin liquid chromatography from Dioscorea steriscus ***:1-diphenyl-2-picrylhydrazyl(DPPH)antiradical activity was employed to investigate antioxidant activity while chromogenic method was used to determine alpha amylase inhibition activity and spectrophotometric methods using triolein as a substrate was used to investigate lipase ***:Thin liquid chromatography profiling revealed eight different flavonoid *** acetate extract yielded two types,R_(f) values 0.38 and 0.40;chloroform extract also yielded two types R_(f) values 0.06 and 0.51,while ethanol extract yielded four types with R_(f) values 0.16,0.33,0.65 and *** the extracted flavonoids exhibited antioxidant activity with ethanol extracts exhibiting the greatest antiradical *** order of enzyme inhibition capacity was ethyl acetateanti-obesity activity as compared to herbex,a commercially anti-obesity medication sold in drug ***-α amylase activity and anti-lipase activity for herbex was(78.38±0.02)%and(76.07±0.09)%respectively,while that for ethanolic extract(R_(f)=0.96)was(93.66±0.00)%and(95.88±0.13)%.Conclusions:Results of the present study show that Dioscorea steriscus consists of bioactive compounds that can act as lipase andα-amylase inhibitors and therefore can be useful for the development of functional foods against *** can also be used as a source of lead compounds for designing new ant-obesity therapeuticals.

Rising consumer concerns with synthetic drugs to treat non-communicable diseases(NCDs)have promoted a shift towards using natural biological active constituents that offer similar health *** canary seed(Phalaris canariensis L)is an emerging crop traditionally used in Mexico to treat *** liberated during simulated digestion of canary seed protein are believed to be responsible for their biological activity;however,no studies have shown the effect of controlled protein hydrolysis using commercial proteases on canary seed protein’s biological ***,this study aimed to explore the in vitro antihypertensive,antidiabetic,and anti-obesity activity of canary seed peptides derived from proteolysis with Alcalase®.Protein fractions were primarily composed of prolamins(54.07±1.8%),glutelins(32.19±3.18%),globulins(5.97±0.52%)and albumins(5.97±0.52%).Theantioxidant activity against DPPH(134.22–150.66μmol TE/mg)and ABTS(520.92–813.33μmol TE/mg).These results provide an insight into the potential development of functional foods using commercial enzymatic hydrolysis of canary seed proteins for treating hypertension,type-2 diabetes,and obesity.
