anti-hemolytic, antibacterial and anti-cancer activities of methanolic extracts from leaves and stems of Polygonum odoratum

Objective: To investigate anti-hemolytic, antibacterial and anti-cancer activities of leaf and stem extracts from Polygonum odoratum. Methods: Leaves and stems of Polygonum odoratum were extracted using methanol and their anti-hemolytic activity was assessed using 2, 2′-Azobis(2-methylpropionamidine) dihydrochloride which is known to generate free radical damage on cell membranes of red blood cells. This damage, represented by hemolysis, was measured using spectrophotometry. antibacterial activity was tested by using a broth microdilution method to find minimal inhibitory concentrations against eight bacterial strains. anti-cancer activity of the extracts was evaluated against a human promyelocytic leukemic cell line(HL-60) by using MTT assay for cell viability and flow cytometry for apoptosis induction and cell cycle analysis. Results: Both leaf and stem extracts have anti-hemolytic activity. The results showed a significantly increased percentage of inhibition in a concentration-dependent manner. Interestingly, the leaf extract showed anti-hemolytic activity to a greater extent than the stem extract. antibacterial activity of the extracts, as indicated by their minimal inhibitory concentration, using 12.5, 50, 25, 25 μg/mL, was measured against Staphylococcus epidermidis, Enterococcus faecium, Enterococcus faecalis and Staphylococcus aureus. The leaf extracts also exhibited anti-cancer activity, demonstrated by significantly decreased cell viability of human promyelocytic cells(HL-60), with an IC_(50) of(350.00±1.85) μg/mL for 48 h and(38.00±0.92) μg/mL for 72 h. Additionally, HL-60 became apoptotic and accumulated in G_1-phase after 48 hours of treatment. Conclusions: The extracts of Polygonum odoratum exhibit potential antihemolytic activity. They also have antibacterial activity by inhibiting growth of Gram-positive bacteria. Relationship between pro-and anti-inflammatory cytokines profiles and some haematological parameters in some Cameroonians infected with Onchocerca volvulus

Objective:To investigate the relationship between white blood cells,lymphocytes,monocytes, and Interleukin(IL)-1α,***-10 and IL-13 production in Cameroonians with Onchocerca volvulus(***) ***:A lolal of 357 individuals from five sites at Upper *** and Sanaga Maritime divisions and located along Sanaga valley of Sanaga River in South Cameroon were screened for the presence of *** using the skin *** levels of the interleukins(IL-) namely IL-1α,IL-6,IL-10 and IL-13 were evaluated using enzyme linked immunoabsorbent assay *** parameters were evaluated using standard laboratory automated ***:*** microfilariae were found in skin tissues of 85(23.81%) *** mean interleukin(IL-) levels in the 0,volvulus control and infected individuals were IL-1αin(1.65±0.79 and 2.31±0.5) pg/ml.:IL-6 in(278.36±55.34 and 20l.74±34.56) pg/ml.:IL-10 in(436.03±208.64 and 418.49±I47.88) pg/*** IL-13 in(8.98±7.28 and 3S.06±11.92) pg/*** was a negative correlation between monocyte counts and IL-10 concentration in positive individuals.A negative correlation of IL-6 with while blood cell and lymphocyte counts was observed(P<0.05).The level of IL-13 was positively associated with microfilaria]load(P<0.05).Conclusions:We observed depressed IL-6 and raised IL-13 concentrations in the sera of individuals with onchocerciasis which implicate these interleukins in the immunological responses of the ***,these IL-6 and IL-13 are associated with *** infection among Cameroonians.

On the Construction of Deepening anti - corruption Mechanism

It needs the foundation of system and the guarantee of organizational system for anti-corruption,but it is more necessary to build and form an effective anti-corruption mechanism,so that the anti-corruption can be really put into practice. anti-corruption mechanism refers to a organic operation system of the interaction,interconnection and constraint between the constituent elements( parts) and elements of national anti-corruption,and as a system,anti-corruption mechanism should have the characteristics of system aticness,comprehensiveness,transparency,legalization,public participation,scientific dynam ic,and internationalism. The construction of deepening anti-corruption mechanism is the need for reconstructing the ruling legitimacy of the party and the governm ent. Adhering to the principle of treating both root causes and symptoms is necessary in the construction of anti-corruption m echanism,com bating and punishing corruption is an important part of anti-corruption,and the prevention and control of corruption is the basic project of anti-corruption. Therefore,the construction of prevention and control mechanism in the anti-corruption mechanism has a more far-reaching significance.

STUDY ON anti-NOISE SCHEME FOR DETECTING PPM INFORMATION IN SHALLOW WATER CHANNELS

Detecting exactly information from pulse position modulation (PPM) is difficult in shallow water channels because of the high noise level there. This paper reports results of analyses of some statistical characteristics of shallow water channel noise and its adverse effect on detecting PPM information; and proposes some efficient countermeasures (the method of statistical decision, use of pulse width discriminator and digital selective-frequency filter) to overcome noise interference. The results have been applied to PPM detection systems and proved to be efficient, and are of reference value for other models of signal detection.

Role of anti Angiogenic Therapy in Prevention of Recurrence in Hormonal Positive Breast Cancer: A Secondary Prevention Strategy and Method of Therapy

Existing literature supports the role of signaling protein vascular endothelial growth factor (VEGF) in tumor growth and metastasis and furthers its involvement in recurrence. In both experimental and clinical studies, VEGF has been shown to be a significant factor involved for aberrant blood vessel growth, and in fact is the target of several classes of antineoplastic drugs [1] [2] [3] [4]. That said, the current standard of care for estrogen receptor positive breast cancer (although improved over the last decade), has not provided a “meaningful preventive shift” since the discovery of angiogenesis and its role in induction of recurrence. In this article, we discuss an anti angiogenic therapy implementing natural compounds to inhibit the production of VEGF. We applied our preclinical data to justify the predicted effect on VEGF. We used liquid biopsy to monitor patients response to therapy as a surrogate for recurrence. We hypothesize that by inhibition of angiogenesis through this protocol, we are able to positively impact tumor recurrence. It is our experience that patients in our sample even with high recurrence scores (based on Oncotype Dx testing) had a major reduction in recurrence when estrogen blockers were combined with this protocol. We also propose longitudinal studies to compare outcomes with combinational therapies with estrogen blockers in highly expected to recur disease.

Expression of TTV-ORF2 Protein for Detection of anti-TTV IgG antibodies in Human Sera

The present study describes the cloning and expression of ORF-2 region of TTV genome and the use of expressed peptide in developing immunoassay for detection of anti-TTV antibodies in serum. Presence of TTV-DNA in serum was detected by PCR amplifying N-22 region of ORF-1 of TTV genome. This was followed by genotyping of TTV by RFLP using N-22 amplicon. Using genotype-1 positive serum as the source of TTV, the ORF-2 region was amplified by PCR and subsequently cloned and expressed in pET-19b vector. The expressed protein, identified as 20 kDa protein on SDS-PAGE gel, was purified by affinity chromatography and then used as antigen to develop western blot assay for detection of anti-TTV antibodies in serum. Analysis of sera for anti-TTV antibodies and their comparison with presence of TTV-DNA, produced encouraging results. There was a good relation between presence of anti-TTV and TTV-DNA in these sera samples. anti-TTV antibodies could be detected in all TTV-DNA positive sera irrespective of the presence of TTV-genotype. This investigation demonstrates that ORF-2 peptide may be used in developing immunoassay for identification of TTV infection.
