Apple of Sodom (<em>Calatropis procera</em>) Callus Extract, a Novel Skincare Active and Its Biological Activity in Skin Models When Combined with Dead Sea Water

Background: Calotropis procera (C. procera), is an authentic plant naturally grown in the flora of Dead Sea region. Despite its toxicity, C. procera presents healing properties. However, it has not been implemented yet in cosmetics as an active ingredient. Objective: The biological effects of C. procera callus extract on skin were elucidated solely and in combination with Dead Sea water (DSW). Methods: The capability of C. procera extract to protect against skin inflammation and irritation was tested on ex vivo human skin organ culture by LPS and SDS addition respectively. Viability and cytokine secretion were evaluated. The combination of C. procera extract with Dead Sea water was tested on full thickness skin equivalents. Gene expression and relevant biochemical markers for glycolysis, hypoxia and extracellular matrix balance were tested. Results: C. procera extract exhibits a protective biological activity against skin irritation and inflammation at the biochemical level. Furthermore, a combination of C. procera extract and DSW demonstrates a potential contribution for skin wellbeing via enhance energy production, resistance to hypoxia and extracellular matrix balance. Conclusions: Topical application of C. procera callus extract might support skin balance and wellbeing at the molecular level. Hence, it is recommended for new cosmetic formulae as standalone or in combination with Dead Sea water, in the effort to achieve anti-Aging bio-activity that is working beyond skin Aging symptoms, especially via skin calming effects and skin energy enhancement.

Rutting Resistance of HMA Rehabilitated with Micro-Surfacing

The work presented here is a study on the measurement and prediction of the rutting resistance of previously rutted asphalt mixes rehabilitated with a layer of micro-surfacing manufactured with virgin and recycled aggregates at different stages of Aging. The experimental procedure consisted of rutting tests on hot mix asphalt slabs already degraded and repaired with virgin and recycled micro-surfacing. Then, the evolution of the behavior of micro-surfacing cast on the hot mix asphalt slabs is observed according to loading cycles of the pavement rutting tester MLPC. Before rutting tests, slabs are subjected to 24 hours at 50°C and aged for 2 days and 5 days at 85°C in the oven. The results showed rutting percentages of 6.3% for hot mix asphalt slabs aged for 2 days and 7.2% for 5 days. These hot mix slabs repaired with virgin micro-surfacing have rutting percentage of about 9.2 % for 2 days of Aging and 6.5% for 5 days of Aging. While, the HMA slabs repaired with recycled micro-surfacing have rutting percentage of about 8.1% for 2 days of Aging and 5.9% for 5 days of Aging. These results allowed the development of a prediction model based essentially on three predictor variables including cycle number, rutting state and percentage of water in the micro-surfacing material. The developed model shows a strong correlation between the predicted rutting values and the rutting values measured with the MLPC rut tester. Thermal Aging in oven has a positive impact on the resistance to permanent deformation of new asphalt mixes and those rehabilitated with micro-surfacing. The parameters of rutting state and contribution water are significant in the rutting prediction model, while the cycle number remains a non-significant parameter in the model but determinant.

Vitamin D3 Supplementation Improved Physical Performance in Healthy Older Adults in Japan: A Pilot Study

Japan has the highest proportion of older adults and so-called “Super-aged society” in the world. These results suggested that the prevalence of both cognitive and physical functional impairment increases with age. Therefore, the present study was designed to investigate the effect of vitamin D3 supplementation in cognitive and physical functional impairment in healthy older adults in Japan. We enrolled 7 Japanese male (age: 76.0 ± 8.7) and 5 female (age: 78.3 ± 9.3) in this study. The physical function of even a person getting on a wheelchair could be tested in all subjects. Treatment group (n = 7) consisted of a 500 IU/day vitamin D3 corresponding to twice of daily requirement for 6 months. Blood was collected by venipuncture and the serum 25-hydroxy vitamin D (25 OHD), 1,25-hydroxy vitamin D (1,25 OHD), Ca and PTH concentration were measured. The Mini-Mental State Examination and the Montreal Cognitive Assessment-Japanese version were used for the cognitive function test. Physical function was measured objectively using the Timed Up and Go (TUG), 4-mgait speed test (4 MGS), 5-repetition sit-to-stand (5 STS), 30-second chair stand test (CS-30) and measurement of upper grip strength. Our results show that a 6-month intake of vitamin D3 increased serum 25 OHD within the insufficiency and sufficiency levels without changing 1,25 OHD, Ca and PTH concentration. And it significantly decreased 4 MGS without changing TUG, 5 STS, CS-30, upper grip strength and cognitive function. Serum 25 OHD cut-off values for skeletal muscle index, grip strength and 4 MGS in diagnosis of sarcopenia are 18.6 - 23.4 ng/mL. These findings suggest that serum 25 OHD levels (23.4 ng/mL) might improve moving capacity, lower limb muscle strength, and physical balance functional impairment.

Telomeres and Telomerase: Molecular Views and Perspectives

Telomere, the nucleoprotein structure at the end of eukaryotic linear chromosomes is indispensable for maintaining the genome stability. Telomeric DNA loss is apparent with each cell division, which marks an endpoint to the indefinite replication of the cell by causing replicative senescence that may lead to the programmed cell death. The loss of telomere is normal in cell division and as such after 20 - 40 divisions, telomere becomes too short to facilitate the capping function. Telomere uncapping or chromosomal free end causes a potential threat to the genomic stability and thus leads to the accumulation of chromosomal abnormalities that have been known to play a role in Aging and cancer. Telomerase, the ribonucleoprotein complex, and its accessory proteins are required to maintain the telomere sequence. Telomerase plays a key role in maintaining the length of telomere by adding G-rich repeat sequences. Its activity has been found to be quite high in the gametes, stem cells and most importantly tumor cells. Almost 85% of tumor cells compensate for telomere loss aided by telomerase-associated protein complex and shelter in complex or telosome. However, 5% - 10% of the cells undergo telomerase-independent mechanism. This review presents the molecular view of the telomere and telomerase along with its associated complex structures. It also discusses its contrasting role in causing cellular senescence and promoting tumorigenesis.

<i>Saccharomyces cerevisiae</i>as a Model to Confirm the Ability of FTIR to Evaluate the Presence of Protein Aggregates

It is known that the presence of protein aggregates in biological samples is associated with natural Aging processes and age-related diseases. The objective of this technical study was to evaluate the potential of Fourier Transform Infrared Spectroscopy to identify the presence of protein aggregates in Saccharomyces cerevisiae containing high levels of protein aggregates. We acquired ATR-FTIR spectra at mid-infrared range (between 4000 and 600 cm-1) and used multivariate analysis to analyze the data. Significant differences between spectra of wild type and mutant strains in the spectral range assigned to proteins were observed. In particular, an increase in β-sheet structures in mutant strains (spectral signals at 1683 and 1628 cm-1) was observed, indicating the putative presence of protein aggregates. These results prove the capacity of FTIR to evaluate changes in protein conformation, mainly protein aggregation, in a fast, simple and non-expensive way, producing insights on the possible application of this technique to the detection of protein aggregates in human biological samples.

Development and Preliminary Validation of the Older Adult Service Usage Assessment

This study provides information on the development and factor scaling, validity, and reliability of a newly developed community service utilization measure (Older Adult Service Usage Assessment-OASUA). Analyzes indicate the OASUA can be used to assess current perceived community service use and satisfaction, as well as providing an indication for future service use and potential service needs with older persons. Additional data collection sites should be identified and a broader sample obtained so as to confirm the psychometric properties of the instrument in conjunction with further development of the OASUA.

Functional Capacity and Anemia in the Community Elderly

Anemia in the elderly population has been associated to clinical manifestations such as functional dependence, low cognitive performance, increased mortality and geriatric syndromes (dementia, delirium, depression and falls). Recent investigations have shown an association between low hemoglobin levels, reduction in muscular strength and physical fitness. The objective of this review was to identify, in the scientific literature, evidence of an association between anemia and functional capacity among older persons. PubMed and Bireme were used as references source. The most relevant evidence pointing towards this association was identified in the epidemiological studies Women’s Health and Aging Studies I and II (WHAS I and II). Functional capacity was evaluated by the walking, balance and sitting back and getting out of the chair tests. Elderly women with either 13 or 14 g/dl have a better performance compared to women with 12 g/dl of hemoglobin, respectively. This study also reported an increase in the mortality risk among women (65 years old and plus) living in community with less than 13.4 g/dl of hemoglobin. Participants were followed for 11 years. WHAS I and II investigators suggested a new criterion, already adopted by the World Health Organization (WHO), to define anemia among elderly women. Conclusion: anemia is common among the elderly and may have an involvement in the impairment of their functional capacity. However, in spite of the importance of these findings, it should be emphasized that an etiologic association (anemia as a cause of functional impairment) has yet to be demonstrated.

Age Invariant Face Recognition Using Convolutional Neural Networks and Set Distances

Biometric security systems based on facial characteristics face a challenging task due to variability in the intrapersonal facial appearance of subjects traced to factors such as pose, illumination, expression and Aging. This paper innovates as it proposes a deep learning and set-based approach to face recognition subject to Aging. The images for each subject taken at various times are treated as a single set, which is then compared to sets of images belonging to other subjects. Facial features are extracted using a convolutional neural network characteristic of deep learning. Our experimental results show that set-based recognition performs better than the singleton-based approach for both face identification and face verification. We also find that by using set-based recognition, it is easier to recognize older subjects from younger ones rather than younger subjects from older ones.

Introduction: The prevalence of asthma in the elderly has been increasing due to the Aging of the global population. Appropriate treatment for asthma in the elderly is now a major issue, as the disease is often fatal and incurs high medical costs in the elderly population. There have been few recent reports on asthma treatment for the elderly in the real-world clinical settings. Objectives: This study was performed to assess how respiratory physicians manage asthma in elderly patients in clinical settings in Japan. Methods: Respiratory physicians in 16 Japanese hospitals responded to a questionnaire survey on asthma and COPD treatment between December 2014 and February 2015. The analysis was performed using data collected from medical records on 2041 asthma patients. Results: The mean patient age was 61.2 years, and 36.6% of the patients were men. In the breakdown by age, 1018 (49.9%) of the patients were elderly (≥ 65 years). Compared to the non-elderly, the elderly patients had a significantly lower BMI, greater smoking history in pack-years, and poorer pulmonary function. There were no significant differences between the elderly and non-elderly in the prescription rates of common medications such as inhaled corticosteroids, long-acting beta-agonists (LABA) administered via inhaler or transdermal patch, long-acting muscarinic antagonists, or leukotriene receptor antagonists. In contrast, mucoactive drugs (18.7% vs. 12.3%, P P P = 0.02). Conclusions: According to this survey, the prescription rates of mucoactive drugs, macrolides, and transdermal LABA were significantly higher in the elderly than in the non-elderly.
