Objective Hyperphosphorylated tau protein is one of the hallmarks of Alzheimer’s disease（AD）. Glycogen Synthase Kinase 3 beta（GSK3 B）is the major kinase phosphorylating tau protein. In patients with AD, increased systemic expression of active GSK3 B has been detected. We previously have shown that aerobic exercise induce neuronal plasticity to rescue brain structure in AD models. Here we test the effect of 12 weeks aerobic exercise on GSK3 B in 9-month-old APP/PS1 transgenic mice to reduce neuropathology and restore brain function. Methods C57 BL/6 J（6-month-old） and APP/PS1 transgenic mice（6-month-old） were randomly divided into exercise group（CE/AE） and sedentary group（CS/AS） with food and water made available ad libitum, respectively. Animals were subjected to treadmill exercise for 12 weeks from 6 months of age to 9-month old. The changes of behavior were detected by eight-arm radial maze test to evaluate the cognitive function consisting of spatial and food motivations. The phosphorylation levels of the Akt, GSK3 B and tau protein were measured using Western Blotting. Results 1) In the eight-arm radial maze, using food as motivation, animals use visual spatial cues for place learning to execute their spatial working and reference memory. Learning performance in the eight-arm radial maze showed that mice of all four groups, even those in AS group, improved their time required to retrieve all baits after training. Among the control groups, the rate of learning of C57 BL/6 J mice was faster than those of APP/PS1 transgenic mice;The aerobic exercise improved the rate of learning of C57 BL/6 J and APP/PS1 transgenic mice. For reference memories（remembering the arms where baits were located so not to enter the unbaited arm）, all groups showed clear learning curves and APP/PS1 transgenic mice were significantly poorer than C57 BL/6 J mice in not making errors entering unbaited arms. Effects of Teriparatide and aerobic exercise on Lumbar Spine Microstructure in Ovariectomized and Tail-Suspended Rats

Osteoporosis is an increasingly prevalent malady of the elderly that is associated with bone fragility and increased risk of fractures. Osteoporosis treatments focus on restoring bone strength and quality. Teriparatide (TPTD) is a therapeutic agent that has been shown to increase bone strength by improving the volume and connectivity of trabecular bone. Exercise is also known to have pro-osteogenic effects. Here we used a rat model of severe osteoporosis (ovariectomized and tail-suspension) to evaluate the effects of TPTD, exercise and a combination of TPTD and exercise on the microstructure of trabecular bone. TPTD mono-therapy and TPTD combined with exercise treatment significantly increased bone mineral density (BMD) in the whole body. Micro-computed tomography analysis revealed that a combination of exercise and TPTD treatment significantly decreased bone surface to volume and trabecular separation compared with those of the control and exercise groups. Node-strut analysis indicated that exercise or TPTD alone did not affect trabecular bone connectivity. However, the combination of exercise and TPTD treatment significantly decreased measures of trabecular bone connectivity (node number) that are consistent with a transition from rod-like to plate-like of trabecular bone microstructures. The combination treatment with exercise and TPTD improved microstructure of trabecular bone in the OVX and tail-suspended rats. These results indicate that combining exercise with TPTD represents a viable means to improve cancellous bone strength in osteoporosis populations.

BACKGROUND Pulmonary hypertension is a serious complication in the treatment of maintenance hemodialysis patients,which seriously affects the quality of life of patients and threatens their life ***,treatment and improvement of pulmonary hypertension are of great significance to improve the quality of life of *** To investigate the intervention and control of pedal-powered bicycle in maintaining quality of life and pulmonary hypertension in hemodialysis *** 73 patients with maintenance hemadialysis combined with pulmonary arterial hypertension at a hemodialysis center in a certain hospital from May 2021 to May 2022 are *** are divided into two groups,37 cases in the control group(group C)and 36 cases in the intervention group(group I).Patients are divided into two groups,group C is treated with oral administration of betaglandin sodium combined with routine nursing *** on group C,group I conducts power cycling *** After treatment,group I patients had higher muscle strength,36-Item Short Form Health Survey scores,and Kidney Disease Targets Areas scores;The 6-minute walk distance test index level was higher and the Borg score was lower;The group I had lower systolic blood pressure,greater vital capacity,higher positive emotion,lower systolic pulmonary artery pressure index level,higher arterial partial oxygen pressure level,lower pulmonary vascular resistance index level,and higher blood oxygen saturation level[158.91±11.89 vs 152.56±12.81,1795.01±603.18 vs 1907.20±574.15,24.00(22.00,29.00)vs 24.00(22.00,28.00),Paerobic exercise combined with Western medicine treatment can effectively improve patients'pulmonary hypertension,alleviate their negative emotions,and enable them to achieve a higher level of quality of life.

Objective:To investigate the impact of resistance training and aerobic exercise before doula-assisted delivery on the psychological well-being and labor duration of ***:105 cases of parturients who gave birth in our hospital from February 2021 to April 2023 were included in this *** patients were divided into a control group of 52 cases and an observation group of 53 *** patients in the control group received no particular intervention before delivery and only received doula *** patients in the observation group received resistance training,aerobic exercise,and doula assistance prior to giving *** delivery,the mothers were observed for 2 hours in the delivery room and then returned to the maternal and infant *** observation indicators of the two groups were ***:In comparison to the control group,the observation group exhibited shorter durations in the first,second,and third stages of labor,as well as the total delivery ***,the amount of postpartum blood loss at 2 hours was smaller in the observation *** rate of cesarean section was lower,and the rate of vaginal natural delivery was higher in the observation *** the intervention,both groups of postpartum women showed decreased SAS and SDS scores compared to before the ***,the observation group had lower scores than the control *** differences were all statistically significant(Paerobic exercise before doula-assisted delivery can effectively improve the psychological state of parturients,shorten labor duration,increase the rate of natural vaginal delivery,and reduce pain and postpartum hemorrhage.

Objective: To explore the intervention effect of aerobic exercise on the model building process of Alzheimer’s in mice. To provide experimental data for Alzheimer’s disease’s mechanism of production and the preventive and therapeutic effect through aerobic exercise. Methods: The experiment chose healthy kunming mice（about 20 g weight）, divided into control group and the AD quiet group, AD exercise *** injection of 0.9% saline in the control group daily was conducted for a continuous injection of 40 days;5% D-galactose saline 0.5 ml was injected daily for the AD quiet group for a continuous injection of 40 days in Alzheimer’s model building;AD exercise group carried out daily subcutaneous injection of 5% D-galactose saline 0.5 ml for 40 days consecutively, and in the meantime AD exercise group took treadmill aerobic exercise. After 40 days, the SOD and the ACh E activity was tested for all mice in the experiment, the number of neurons in hippocampus CA1 and CA3 area and the number of positive cells of the epidermal growth factor was also observed for all three groups. Finally, the effect of aerobic exercise in the intervention and prevention for Alzheimer’s disease was explored. Results: 1. The SOD and the ACh E activity for the AD quiet group was declining in the model building process.2. The number of neurons in hippocampus CA1 and CA3 area for AD exercise group significantly exceeded the number for the AD quiet group（P ＜ 0.05）.3. The number of positive cells of the epidermal growth factor for the AD exercise group significantly exceeded the number for the AD quiet group（P ＜ 0.05）. Conclusion: aerobic exercise intervention may modulate AD mice’ hippocampal damage and repair function, which is of great theoretical significance to intervene and prevent familial Alzheimer’s disease in early exercise.

OBJECTIVE:The crosstalk between mTORC1/S6K1 signaling and AMPK is emerging as a powerful and highly regulated way to gauge cellular energy and nutrient *** aim of the current study was to elucidate the mechanism by which exercise training reverses lipid-induced insulin resistance and the role of AMPK/m TOR/S6K1 signaling axis in mediating this response in skeletal *** AND RESULTS:Insulin resistant mouse model was produced by feeding high-fat diet,which was associated with decreased Akt expression and increased intramuscular triglyceride *** in lipid metabolism were accompanied by increased total protein and phosphorylation of S6K1,SREBP-1cleavage,and decreased AMPK phosphorylation in skeletal *** training reversed these impairments by inhibiting SREBP-1 cleavage through AMPK/m TOR/S6K1 signaling,resulting in decreased intramyocellular lipid *** determine the role of AMPK/m TOR/S6K1 axis in insulin sensitization regulation in response to lipid oversupply,C2C12 myotubes were exposed to palmitate,resulting in an increased insulin-dependent Akt Ser473 phosphorylation,associated with increased phosphorylation of S6K1 on *** these changes were reversed by activation of *** with this,inhibition of AMPK by compound C induced an enhanced phosphorylation of both S6K1 and Akt,and silencing of S6K1 with si RNA showed no effect on Akt phosphorylation in cultured myotubes with or without *** addition,compound C led to an elevated SREBP-1 cleavage but this action was blocked by S6K1 si ***:Our results indicate that exercise training inhibits SREBP-1cleavage through AMPK/m TOR/S6K1 signaling,resulting in decreased de novo lipogenesis and intramyocellular lipid accumulation.

Changes of DNA methylation level on promoter region of HSD11β2 gene(11β hydroxysd-hydroxyl dehydrogenase,11βHSD) are important mechanisms controlling hypertension epigenetics in peripheral blood of patients with primary *** purpose of this study was to investigate the effect of HSD11β2 gene promoter methylation level on hypertensive vascular function before and after aerobic exercise *** the beginning of the study and after 6 weeks of aerobic exercise intervention,the hypertension group was selected,and the control group was measured for signs and blood pressure *** research subjects fasted within hours before blood drawing,collected venous blood,and tested relevant biochemical *** was used to detect plasma protein levels between hypertension and control *** expression of 11βHSD in endothelial cells and the changes in the level of hypertension-related molecules were measured by real-time PCR,western bolt and immunoflourence *** results showed that the methylation level of the promoter region of HSD11β2 gene in the hypertension group was higher than that in the control group(P aerobic exercise intervention,the proportion and methylation level of were decreased,but higher than that in the control group,and the metabolite of cortisol and the metabolite of corticosterone in the blood of the hypertension group *** study shows that aerobic exercise can effectively inhibit the expression of 11βHSD caused by hypertension and reduce blood pressure by reducing the methylation level of HSD11β2 promoter region.

Purpose:Whether IGF-1 and its receptor IGF-1 R are involved in exercise-induced improvement of glycolipid metabolism in obesity remain unclear,although the important roles of IGF-1/IGF-1 R in regulating glycolipid *** the purpose of the study was to explore the effects of IGF-1/IGF-1 R on exercise-induced improvement of glycolipid metabolism in ob/ob ***:Male ob/ob mice were randomly divided into ob/ob group（OB,n=5） and exercised ob/ob group（EOB,n=5）,and C57 BL/6 mice used as control（Con,n=5）.All of the mice were fed with normal diet,and EOB mice participated in a 6-week moderate aerobic exercise on a treadmill with gradually intensity,1 h a day and 5 days per *** 6-week exercise,glycolipid metabolism index（fasting insulin,fasting blood glucose,TC,TG,LDL and HDL） were *** IGF-1 and the protein levels of IGF-1 and IGF-1 R in the liver and gastrocnemius were detected by ELISA and Western blot,***:（1） Compared with Con, serum IGF-1 and the protein levels of IGF-1 and IGF-1 R in the gastrocnemius of ob/ob mice were abnormally increased accompanied with the disorder of glycolipid metabolism,which might be related to leptin deficient（because ob/ob mice is lack of leptin and leptin inhibits IGF-1 level）.（2） 6-week exercise decreased the IGF-1 in serum and IGF-1/IGF-1 R in gastrocnemius of ob/ob mice,meanwhile,the disorder of glycolipid metabolism were ***:The retrieves of IGF-1 in serum and IGF-1/IGF-1 R in gastrocnemius by 6-week aerobic exercise might be associated with the exercise-induced improvement of glycolipid metabolism in ob/ob mice.

The aging trend of the global population makes more and more people pay attention to the influencing factors and prevention methods of all kinds of elderly *** cognitive impairment(MCI) is a precursory stage between normal aging and Alzheimer’s disease(AD), characterized by memory and executive function decline, gray matter atrophy and white matter *** conversion rate to AD and other dementia is as high as 10% ～ 15%, but it is also the best stage to prevent Alzheimer’s *** patients show poor exercise control ability in different *** research shows that aerobic dance, swimming, power cycling, tai chi, jogging, finger exercise and other aerobic exercises can improve the cognitive status of the elderly, delay the development of the disease, and improve the quality of life of the *** them, finger exercise is a kind of aerobic program widely used by the *** the brain functional location area, finger movement can activate a wide range of cortical motor sensory areas, which can feedback the activation of the cerebral cortex and enhance the flow of cerebral blood *** the fingers perform simple exercise, the cerebral blood flow increases by 10%, when the fingers perform fine exercise, the cerebral blood flow can increase by more than 35%.Therefore, the long-term fine hand movement can activate the nervous system and exercise *** with the fMRI research results of pre exercise control, the laboratory developed finger exercises to improve the cognitive function of the elderly, which were popularized in different community units and widely used by the *** with the motor control function of brain basal ganglia, this paper summarizes the effects of different aerobic exercises on the cognitive function of the elderly, and provides theoretical reference for the later exercise intervention of the elderly.
