Sheng-jiang powder ameliorates obesity-induced pancreatic inflammatory injury via stimulating activation of the AMPK signalling pathway in rats

AIM To investigate the mechanisms by which Sheng-jiang powder(SJP) ameliorates obesity-induced pancreatic inflammatory *** Sprague-Dawley rats were randomized into three groups: normal group(NG), obese group(HLG), or SJP treatment group(HSG). Obesity was induced by feeding a high-fat diet in the HLG and HSG, while the NG received standard chow. Rats were euthanized after 12 wk, and blood and pancreatic tissues were collected for histopathological analyses. Nuclear factor kappa-light-chain-enhancer of activated B cells(NF-κB) and transforming growth factor beta(TGF-β) expression, serum triglyceride and adiponectin levels, and apoptosis in pancreatic acinar cells were assessed. A high-fat AR42 J acinar cell injury model was established using very low-density lipoprotein(VLDL). AR42 J acinar cell culture supernatant, treated with different interventions, was applied to seven groups of pancreatic stellate cells(PSCs). AMPK-associated signaling to bridge the gap between fuel metabolism and hepatocyte viability

The adenosine monophosphate-activated protein kinase (AMPK) and p70 ribosomal S6 kinase-1 pathway may serve as a key signaling flow that regulates energy metabolism; thus, this pathway becomes an attractive target for the treatment of liver diseases that result from metabolic derangements. In addition, AMPK emerges as a kinase that controls the redox-state and mitochondrial function, whose activity may be modulated by antioxidants. A close link exists between fuel metabolism and mitochondrial biogenesis. The relationship between fuel metabolism and cell survival strongly implies the existence of a shared signaling network, by which hepatocytes respond to challenges of external stimuli. The AMPK pathway may belong to this network. A series of drugs and therapeutic candidates enable hepatocytes to protect mitochondria from radical stress and increase cell viability, which may be associated with the activation of AMPK, liver kinase B1, and other molecules or components. Consequently, the components downstream of AMPK may contribute to stabilizing mitochondrial membrane potential for hepatocyte survival. In this review, we discuss the role of the AMPK pathway in hepatic energy metabolism and hepatocyte viability. This information may help identify ways to prevent and/or treat hepatic diseases caused by the metabolic syndrome. Moreover, clinical drugs and experimental therapeutic candidates that directly or indirectly modulate the AMPK pathway in distinct manners are discussed here with particular emphasis on their effects on fuel metabolism and mitochondrial function.

EFFECTS OF SODIUM SALICYLATE ON THE FEBRILE RESPONSE AND INCREASED LEVELS OF CYCLIC AMP IN CEREBRO-SPINAL FLUID DURING ENDOGENOUS PYROGEN-INDUCED FEVER IN RABBITS

To further evaluate the causality between endogenous pyrogen (EP)-induced fever andcyclic adenosine- 3’, 5’- monophosphate (cyclic AMP) level. the effects of sodium salicylate(SS) on the febrile response and increased levels of cyclic AMP in both cerebrospinal fluid(c.s.f.) and plasma during EP- induced fever in rabbits were observed. The results suggestthat cyclic AMP is probably involved in the central mediation of EP-induced fever and thatincreased concentration of cyclic AMP in c.s.f. associated with EP- induced fever is not theresult of temperature elevation but appears to be caused by the increased synthesis in the cen-tral nervous system. In addition it is confirmed that blood is impossibly a contributorysource of increased cyclic AMP in c.s.f. during EP fever, and that SS may act subsequentto the increase in cyclic AMP.

Urinary analysis based on surface-enhanced Raman scattering for the noninvasive screening of lung cancer

We developed a reliable simple analysis of adenosine,creatinine and other nucleosides(cytidine and guanosine)with single measurement based on surface-enhanced Raman spectroscopy(SERS)using DSP-AuNPs with Cu^(2+)ions probe and applied it to achieve noninvasive screening of lung *** gold nanoparticles(AuNPs)were modified with 3,3′-dithiodipropionic acid di(N-hydroxysuccinimide ester)(DSP),which could react with above mentioned molecules followed with the aggregation of AuNPs at the presence of Cu^(2+),thus form the plasmonic hot spots to dramatically increase their fingerprint Raman *** probe of DSP-AuNPs was applied for the detail analysis of urinary adenosine of healthy people and lung cancer *** SERS measurement indicated a higher concentration of urinary adenosine for lung cancer patients than that of healthy people,which was consistent with the results measured by high-performance liquid chromatography(HPLC).In combination of Raman spectroscopy with the orthogonal partial least squares discriminant analysis(OPLS-DA)model,we are able to discriminate the lung cancer patients from healthy people with the urinary *** results indicated that besides of adenosine,other metabolites including uric acid,guanine and creatinine may also be the potential tumor markers in urine for the noninvasive lung cancer *** a method paves a way for the noninvasive cancer screening and it can be further modified for the detection of other molecules on medical diagnosis.

Sevoflurane pretreatment inhibits the myocardial apoptosis caused by hypoxia reoxygenation through AMPK pathway:An experimental study

Objective:To study whether sevoflurane pretreatment inhibits the myocardial apoptosis caused by hypoxia reoxygenation through AMPK ***:H9c2 myocardial cell lines were cultured and divided into control group(C group),hypoxia reoxygenation group(H/R group),sevoflurane pretreatment+hypoxia reoxygenation group(SP group) and sevoflurane combined with Compound C pretreatment+hypoxia reoxygenation group(ComC group),and the cell proliferation activity and apoptosis rate,myocardial enzyme levels in culture medium as well as the expression of apoptosis genes and p-AMPK in cells were ***:p-AMPK expression in cells of H/R group was significantly lower than that of C group,SP group was significantly higher than that of H/R group;cell proliferation activity value and Bcl-2 expression in cells of H/R group were significantly lower than those of C group,SP group were significantly higher than those of H/R group,Com C group were significantly lower than those of SP group;apoptosis rate,LDH,CK and AST levels as well as the Bax and Caspase-3 expression in cells of H/R group were significantly higher than those of C group,SP group were significantly lower than those of H/R group,ComC group were significantly higher than those of SP ***:Sevoflurane pretreatment can activate AMPK signaling pathway to inhibit the myocardial apoptosis caused by hypoxia reoxygenation.

Induction of inosine containing mRNA during the inflammatory stress in C57BL/6 mouse

A-to-I RNA editing is a recently discovered process of post-transcription modification of pre-mRNA by adenosine deamination that results in the production of pro-teins not inherent in the genome. The present study aimed to identify a role for A-to-I RNA editing in the acute inflamma-tion. It was found that A-to-I edtiting activities of RNA edi-tases, ADARs, were upregulated in lung, spleen, thymus and lymph node of mouse during endotoxin stimulation. Impor-tantly, the number of inosine in poly(A) RNA isolated from mouse lung and spleen was significantly increased in corre-lating with the induction of ADARs?editing activity. The in vitro synthesized RNA which did not contain inosine was edited by thymus extracts and the generation of inosine was greatly increased after editing in LPS treated thymus extract. Take together, these data suggest that A-to-I RNA editing by ADARs may play an important role in pathogenesis of in-flammation. The existence of high level of I-mRNA also sug-gests that more protein isoforms might be generated from a single gene via adenosine deamination by ADARs during inflammatory stress.

A-to-I RNA editing: A new mechanism of genomic information modification

A-to-I RNA editing, the important event of gene modification, which takes place at post-transcriptional level, was firstly reported in 1991. The molecular mechanism of A-to-I RNA editing involves site-selective deamination of adenosine to inosine in pre-mRNA, which leads to altering translation codons and splicing in nuclear transcripts, thereby functionally distinct proteins can be produced from a single gene. The mammalian editing enzymes ADARs (adenosine deaminases acting on RNA) are widely expressed in brain and other tissues, however, up to date their sub-strates are mainly found in the central nervous system. It has recently been noticed that imperfect editing of these RNA substrates play critical roles in corresponding diseases, indi-cating that A-to-I RNA editing may be quite important in physiological or pathophysiological processes. Finding more new substrates of ADARs, especially in peripheral tissues, and performing functional research on new genes will be helpful to elucidate the biological significance of A-to-I RNA editing.

To the Editor： Current treatment guidelines for patients with acute coronary syndrome （ACS） undergoing percutaneous coronary intervention （PCI） recommend dual antiplatelet therapy, a combination of aspirin and a P2YI2 inhibitor （i.e., clopidogrel, prasugrel, and ticagrelor） for a minimum of 12 months. Ticagrelor, an oral reversibly binding platelet P2Y 12 receptor inhibitor, generates a greater and more consistent inhibitory effect with rapid onset of action as compared to clopidogrel.

BACKGROUND Atrial arrhythmias such as paroxysmal supraventricular tachycardia(PSVT)and atrial flutter(AF)are common in the perioperative *** commonly resolve ***,occasionally,they may continually progress to fatal arrhythmias or cause ***,prompt and appropriate management is *** SUMMARY A 46-year-old female patient diagnosed with cervical C6-7 radiculopathy characterized by decreased sensation in the right third,fourth and fifth fingers underwent C6-7 anterior cervical disc fusion *** showed PSVT and ventricular tachycardia during C6-7 disc ***,the patient remained *** treatment with esmolol and lidocaine for ventricular tachycardia was *** massage and Valsalva maneuver were attempted but PSVT did not *** surgery was paused,and the patient’s fraction of inspired oxygen was set to 100%.adenosine was administered for pharmacological management of *** arrhythmia temporarily ***,it then transformed into *** was administered,which briefly decreased blood pressure,which immediately *** resumed while the patient was in normal sinus *** was discharged safely on postoperative day 6 without complications or ***,she is living a healthy life without arrhythmia *** Ganglia associated with cardiac arrhythmias in the surgical site should be identified during cervical spine surgery.
