The regulation of agmatine on MMP expression in the cerebral endothelial cells overexpressed ADC gene

Objective In this study we will investigate the effect and possible mechanism of agmatine on MMP expression.Methods We established a transfection of human ADC gene into the murine bEnd.3 cells(bEnd.3-ADC)by the retroviral packaging cell line PT67.The bEnd.3-ADC cells were maintained in the hypoxia chamber for 6 hours,then returned to a normoxic incubator with glucose for 16 hours.We performed RT-PCR and western blot analysis against MMP-2,MMP-9,eNOS and ATF3,and measured the production of NO using Griess reagent.To determine the possible mechanism of agmatine on MMP expression we used inhibitory study.Results The expression of MMP was decreased by agmatine administered exogenously and endogenously,while eNOS was increased.We showed L-NAME(NOS inhibitor)altered the suppression of MMP-9 by exogenously administered agmatine.It seems that MMP-9 suppression by exogenously administrated agmatine is mediated,at least in part,via eNOS.We also found that ATF3 was increased significantly in ADC overexpression cells,but it was attenuated by NOS inhibitor.Furthermore,we found the suppression of MMP by agmatine was attenuated in cells transfected with ATF3 siRNA.Conclusions Our study suggested that endogenously administered agmatine suppress the MMP-2 and MMP-9 expression via eNOS-ATF3-MMPs pathway.

目的筛选出与尿道下裂发病相关的基因。方法尿道下裂病人的尿道或包皮组织,年龄配对的包皮过长患儿组织作为对照,提取组织mRNA,合成双股cDNA并纯化,再逆转录成cRNA,生物素标记的cRNA打成碎片后与人类基因芯片进行杂交,计算机工作站自动扫描并记录芯片上的信号强度,转化为EXCEL数据库,总共分析了22000个基因的表达差异。结果发现一组表达有显著差异的基因,包括CYR61,CTGF,ATF3和GADD45。结论这些表达差异的基因可能参与尿道下裂的发生和发展,为进一步研究关键基因提供了依据。

＜正＞Objective Bone cancer pain,one of the most common types of cancer pain,seriously affects the quality of life of patients,however,the basic neurobiological mechanism of bone cancer pain remains unclear.Therefore,much more effort should be required for further understanding and development of therapeutics of bone cancer pain.The present study is to explore the selective change of TRPV1 expression on different segments of dorsal root ganglion during bone cancer pain.Methods A rat model of bone cancer pain was built by injection of Syngeneic Walker 256 mammary gland carcinoma cells into the tibia medullary cavity.The expression of the ATF3,a marker of nerve injury,and capsaicin receptor TRPV1 was measured by immunofluorescence.Results（1） ATF3 was stained positively in the ipsilateral L2,L3,L4 and L5 dorsal root ganglion（DRG） neurons from cancer animals.（2）In the L4 and L5 DRG neurons,the percentage of TRPV1 - positive neurons was increased on the fourteenth day[（30.02±0.86）%in sham group and（35.26±0.84）%in cancer group] and the twenty-first day[（30.84±0.77）%in sham group and（37.92±1.25）%in cancer group].（3）In the L2 and L3 DGR neurons which had been proved to be injury,had been proved to be injury the level of TRPV 1 expression were similar in sham group and cancer group,whatever in the fourteenth day[（33.07±0.68）%in sham group and（34.07±0.93）%in cancer group]or in the twenty-first day[（32.86±0.8）%in sham group and（33.14±0.84）%in cancer group].Conclusion TRPV 1 expression is selectively enhanced on different segments of dorsal root ganglion during bone cancer pain.

ATF3 rSNPs, transcriptional factor binding sites and human etiology

Three rSNPs (rs3125289, rs1877474 and rs11119982) in one intron of the activating transcription factor 3 (ATF3) gene have been significantly associated with the human etiology of hypospadias and may be associated with human disease. These rSNP alleles alter the DNA landscape for potential transcriptional factors (TFs) to attach resulting in changes in transcriptional factor binding sites (TFBS). These TFBS changes are examined with respect to the human etiology of hypospadias which has been found to be significantly associated with the rSNPs.
