To explore the similarities and differences of molecular mechanism of wheat defense against Puccinia striiformis ***（Pst） and Blumeria graminis ***（Bgt）,quantitative proteomic analysis of Xingmin318（XM318）,a wheat cultivar highly resistant to both wheat stripe rust and powdery mildew,was performed by the TMT technology.A total of 741 proteins were significantly different and identified as differentially accumulated proteins（DAPs）.Bioinformatics analysis indicated that all these DAPs relate to reactive oxygen species metabolic process,stimulus response,immune system process and other basal *** addition,results of bioinformatics showed that the main functional or metabolism categories for DAPs was identical between Pst and Bgt treatments,but some categories were existed obvious difference,such as process of ***,only 42 DAPs responded to both Pst and Bgt *** proteins of DAPs were randomly selected and performed qRT-PCR *** results showed that the mRNA expression levels of eleven proteins were consistent with the protein expression ***,two proteins,member of Glutathione Stransferase family and member of aba/wds induced protein family,which was in response to both Pst and Bgt treatments,were performed gene silencing using the virus-induced gene silencing（VIGS） *** results indicated that the Glutathione S-transferase protein mainly played an important role in anti-Bgt and had no function in resistance to *** to the aba/wds induced protein,this protein was only reflected resistance in wheat-Pst *** study revealed that wheat cultivar was likely to have different response mechanisms under the stress of Pst and *** also provide a new vision to better understand the complex regulation of the gene expression and numerous metabolic pathways between wheat and Pst or Bgt interaction.
