Effects of 17beta-estradiol on cell migration in male chicks distribution of primordial germ

Aim： To assess whether exogenous estradiol has any effect on migration of primordial germ cells （PGCs） in the chick. Methods： Fertilized eggs were treated with 17beta-estradiol （E2）（80 lag/egg） at stage X （day 0 of incubation）, stages 8-10 （incubation 30 h） and 13-15 （incubation 55 h）. Controls received vehicle （emulsion） only. Changes in PGC number were measured on different days according to developmental stages. Results： In male right gonads, but not in female left gonads, at stages 28-30 （incubation 132 h） significant decreases in the mean number of PGCs aggregating were observed compared with the controls （P 〈 0.05） while the total PGC number in the right and left gonads at each stage did not change （P 〉 0.05）. Conclusion： The present study provides evidence that E2 has significant effects on the localization of PGCs in male right, but not female left, gonads of chicken embryos at stages 28-30, compared with controls. （Asian J Andro12008 Mar; 10： 243-248）

Objective:Estrogen plays an important role in the osteogenic differentiation of mesenchymal stem cells while stem cells from apical papilla(SCAPs) can contribute to the formation of dentin/bone-like *** date,the effects of estrogen on the differentiation of SCAPs remain ***:SCAPs were isolated and treated with 10-7 M 17beta-estradiol(E2).Their odonto/osteogenic potency and the involvement of mitogen-activated protein kinase(MAPK) signaling pathway were subsequently investigated by using methylthiazolyl-tetrazolium(MTT) assay,***:MTT and flow cytometry results demonstrated that E2 treatment had no effect on the proliferation of SCAPs in vitro,while alkaline phosphatase(ALP) assay and alizarin red staining showed that E2 could significantly promotes ALP activity and mineralization ability in ***-time reverse transcription polymerase chain reaction(RT-PCR) and western blot assay revealed that the odonto/osteogenic markers(ALP,DMP1/DMP1,DSPP/DSP,RUNX2/RUNX2,OSX/OSX and OCN/OCN) were significantly up-regulated in E2-treated *** addition,the expression of phosphor-p38 and phosphor-JNK in these stem cells was enhanced by E2 treatment,so was the expression of the nuclear downstream transcription factors including phosphor-Sp1,phosphor-Elk-1,phosphor-c-Jun and phosphor-c-Fos,indicating the activation of MAPK signaling pathway during the odonto/osteogenicdifferentiation of E2-treated ***,the differentiation of E2-treated SCAPs was inhibited in the presence of MAPK specific ***:10-7 M 17beta-estradiol enhanced the odonto/osteogenic differentiation of SCAPs via the activation of MAPK signaling pathway.

Effect of 17beta-estradiol and Progesterone on Superovulatory Response of Donor and Pregnancy of Recipient

To study the effects of 17beta-estradiol （E2 ） and progesterone （P4） levels on superovulatory response and recipients conception rate, the E2 and P4 levels of embryo donor （Borderdale） and recipient ewes （Inner Mongolian sheep） were determined during multiple ovulation and em- bryo transfer （MOET）. The results showed that the E2 level increased from the day of initiation by CIDR （Controlled internal drug release devices） treatment and decreased to 3.18± 1.32 pg/ml on the day of superovulatory estrus. On the day of embryo collection, the ewes having higher levels of estrogenic peak showed better superovulatory response than the other groups having comparatively lower levels. On the day of initiation of super- ovulatory treatment, the ewes having lower levels of P4 showed better superovulatory response than those having higher levels of P4. The E2 level of the early abortion group increased from Day 0 to Day 45, while that of two pregnant recipient groups came down from Day 0 to Day 30. The ser- um P4 level of the early abortion group was significantly lower than that of the pregnant recipient groups on Day 45 （ P 〈 0.05）, but no significant difference was found between the two pregnant groups after Day 30. Therefore, the levels of E2 and P, on definite time affect superovulatory re- sponse of donors and conception rate of recipients during MOET.

Effects of long-term estrogen replacement therapy on beta-amyloid precursor protein and mRNA expression in ovariectomized rat hippocampus

BACKGROUND： In vitro cultures of neural stem cells have shown that estrogen can regulate beta-amyloid precursor protein （β-APP） metabolism and reduce amyloid-beta production. OBJECTIVE： To investigate the effects of long-term oral administration of compound nylestriol or low-dose 17beta-estradiol on β-APP and mRNA expression in the hippocampus of ovariectomized （OVX） rats. DESIGN, TIME AND SETTING： This randomized and controlled experiment was performed at the Animal Laboratory and Laboratory of Endocrine and Metabolic Disease, Xiangya Second Hospital of Central South University between April 2003 and May 2004. MATERIALS： According to body mass, 50 six-month-old female Sprague-Dawley rats were randomly divided into five groups （n = 10 per group）： normal control, sham operation, OVX model, 17beta-estradiol （Sigma, USA）, and compound nylestriol tablet （Laboratory of Endocrine and Metabolic Disease, Xiangya Second Hospital of Central South University） groups. METHODS： Rats in OVX plus 17beta-estradiol and OVX plus compound nylestriol tablet groups underwent ovariectomy. On the second day after surgery, rats were intragastrically given 17beta-estradiol （100 μg/kg）, once per day or compound nylestriol tablet （0.5 mg/kg） and levonorgestrel （0.15 mg/kg） every 2 days. MAIN OUTCOME MEASURES： β-APP expression in the hippocampus of OVX rats was determined using immunohistochemistry （SABC method） and β-APP mRNA expression was analyzed by in situ hybridization. The results were quantitatively analyzed using cell counting and average optical density. RESULTS： The number and optical density of β-APP-positive neurons in every subregion of the hippocampus of OVX rats was dramatically increased compared with normal and sham operation groups following 35 weeks of administration （P 〈 0.05）.
