Rapid assembly of 1,3-indanedione-based spirocyclic tetrahydroquinolines for inducing human lung cancer cell apoptosis

The rapid collection of a library of biologically relevant molecules is essential in new drug *** we show a variety of bioactive 1,3-indanedione-based spirocyclic tetrahydroquinolines that have been concisely and efficiently constructed under the guidance of biology-oriented synthesis(BIOS).Using Pd(PPh_(3))_(4) as a catalyst,various 2-arylidene-1,3-indanediones can react with vinyl benzoxazinanones under the mild condition to afford a new class of spirocyclic tetrahydroquinolines in moderate to excellent yields(45%-99%)with high stereocontrol(>95:5 diastereoselectivity).Their structures were unambiguously confirmed by X-ray single crystal *** biological evaluation revealed that two of the 1,3-indanedione-based spirocyclic tetrahydroquinolines show remarkable apoptosis-inducing activities over A549 human lung cancer cells,exceeding that of *** antitur activities were triggered by reactive oxygen species,commonly overproduced in cancer *** clinical demand for antitumor chemotherapy for lung cancer has encouraged an all-out search for analogues with lower toxcity,improved therapeutic index and increased activity.

Indanone-containing compounds are very important components of various natural products,chemical drugs,agrochemicals and functionalized material fields.1,3-indanedione is a typical cyclic 1,3-dicarbonyl compound with three contiguous reactive electrophilic and nucleophilic *** easily prepared 2-arylidene-1,3-indanediones are activeα,β-unsaturated carbonyl compounds and could act as 1,3-dipolarophile,1,4-dienophile and 1-oxa-1,3-diene,*** the other hand,the homodimer(bindone)and cyclotrimer(truxenone)derived from basecatalyzed self-condensation of 1,3-indanedione are also reactive carbonyl ***,1,3-indanedione and its derivatives can be employed as key substrates in various domino and multicomponent *** this review,we summarized the recent progress on the domino and multicomponent reactions of 1,3-indanedione,especially on our achievements on the synthetic applications for the complex indanone-containing carbocyclic and heterocyclic compounds.

Synthesis and Crystal Structure of 5-Cyclopropyl- 10-(4-fluorophenyl)-7,7-dimethyl-7,8-dihydro-5H- indeno[1,2-b]quinoline-9,11(6H,10H)-dione

The title compound 5-cyclopropyl-10-（4-fluorophenyl）-7,7-dimethyl-7,8-dihydro- 5H-indeno[1,2-b]quinolne-9,11（6H,10H）-dione was obtained by the reaction of 4-fluorobenzal- dehyde, 2H-indene-1,3-dione and 3-（cyclopropylamino）-5,5-dimethylcyclohex-2-enone in the presence of acetic acid under microwave irradiation. Its structure was confirmed by IR and 1H- NMR spectra. The crystal is of monoclinic, space group P21/c with a = 14.138（3）, b = 8.952（2）, c = 17.140（3）A, β = 102.253（3）°, C27H24FNO2, Mr = 413.47, Z = 4, V = 2119.9（8）A^3, Dc = 1.296 g/cm^3, μ（MoKα） = 0.087 mm^-1, F（000） = 872, the final R = 0.0425 and wR = 0.0905 for 2336 observed reflections （I 〉 2σ（I））. X-ray analysis revealed that the pyridine ring adopts a boat conformation and the six-membered ring fused with it assumes a twist boat conformation.

Convenient construction of spiro[indoline-3,5’-pyrrolo[3,4-c]carbazole]and spiro[indene-2,5’-pyrrolo[3,4-c]carbazole]via acid-catalyzed Diels-Alder reaction

p-TsOH catalyzed Diels-Alder reaction of 3-(indol-3-yl)maleimides with 3-phenacylideneoxindoles in toluene at 80℃for two hours afforded cis/trans isomers of 3 a’,4’,6’,10 c’-tetrahydrospiro[indoline-3,5’-pyrrolo[3,4-c]carbazoles]in nearly comparable yields,which could be easily converted to the corresponding 4’,6’-dihydrospiro[indoline-3,5’-pyrrolo[3,4-c]carbazole]in high yields and with high diastereoselectivity by further DDQ ***,the similar reaction of 3-(indol-3-yl)maleimides with 2-arylidene-1,3-indanediones in toluene 80℃and sequential DDQ oxidation afforded functionalized dihydrospiro[indene-2,5’-pyrrolo[3,4-c]carbazoles]as major products.

The β-enamino esters,which were easily generated in situ from addition of arylamines to electron-deficient alkynes,have widely used as convenient building blocks for synthesis of heterocyclic *** continuation of our study on the synthetic applications of reactive β-enamino ester intermediates,here,we provided an efficient synthetic protocol for the biologically important indenone-fused *** the presence of Lewis acid BF3· OEt2,cycloaddition reaction of 2-arylidene-1,3-indanediones with β-enamino esters generated from addition of arylamines to dimethyl acetylenedicarboxylate to give densely substituted 1,4-diphenyl-4,5-dihydro-1 H-indeno[1,2-b]pyridines in satisfactory *** 2-isatylidene-1,3-indanediones were employed under same conditions,the one-pot reaction afforded the corresponding spiro[indeno[1,2-b]pyridine-4,3’-indolines] in high *** the other hand,the unusual 1,2-diphenyl-2,5-dihydro-1 H-indeno[1,2-b]pyridines can be obtained in moderate to good yields in the four-component reaction.

The nucleophilic phosphine-catalyzed annulations have emerged as one of powerful tools for the construction diverse carbocyclic and heterocyclic *** the phosphine catalyzed annulations,dialkyl acetylenedicarboxylates,alkyl propionates and aroylacetylenes are the most common used electron-deficient *** recent years,a new kind of electron-deficient alkyne,dialkyl hex-2-en-4-ynedioate,emerged as new versatile reactive substrates in the *** order to further demonstrated the potential synthetic values of the nucleophilic phosphine catalyzed cyclization,we also successfully developed some mutlicomponent reaction containing triphenylphosphine and dialkyl hex-2-en-4-ynedioate for the efficient construction of diverse polycyclic and spiroheterocyclic *** three-component reaction of triphenylphosphine,dialkyl hex-2-en-4-ynedioate and arylidene-1,3-indanedione in dry DME at zero to room temperature resulted in functionalized triphenylphosphanylidene 1’,3’-dihydrospiro[cyclopentane-1,2’-inden]-2-enes in 75-92% yields.1 H NMR spectra and single crystal structures indicated the trans-isomer of the products was predominately formed and the reaction has very high diastereoselectivity.

The heteroaromatic N-ylides have been drawing much attention of the chemists due to their wide spectrum of utilities in numerous chemical transformations. The cycloaddition of heteroaromatic N-ylides with various dipolarophiles showed very interesting chemical, regio-and steric selectivity. Over the last few years, we have concentrated our works on the development of synthetic applications of heteroaromatic N-ylides for various polyheterocyclic systems. Recently, we investigated the cycloaddition reactions of various heteroaromatic N-ylides with in situ generated 2-arylidene-1,3-indanediones and disclosed that the minor difference of substrates exhibited various reactivities which afforded diverse spiro, fused and polycyclic derivatives.
