Polyethylenimine-cyclodextrin-tegafur conjugate shows anti-cancer activity and a potential for gene delivery

Polyethylenimine-cyclodextrin-tegafur(PEI-CyD-tegafur) conjugate was synthesized as a novel multifunctional prodrug of tegafur for co-delivery of chemotherapeutic agent tegafur and enhanced green fluorescent protein(EGFP) reporter plasmid *** of tegafur to PEI-CyD via chemical linkage was characterized by 1 H NMR spectrometry and ultraviolet(UV) ***-CyD-tegafur was able to condense plasmid DNA into complexes of around 150 nm with positive charge at the N/P ratio of 25,in accordance with electron microscopy observation of compact and monodisperse *** results of in vitro experiments showed enhanced cytotoxicity and considerable transfection efficiency in B16F10 cell ***,PEI-CyD-tegafur may have great potential as a co-delivery system with anti-cancer activity and potential for gene delivery.

Preparation of a novel biodegradable β-cyclodextrin-containing polymer

A novel cyclodextrin-containing polymer was prepared by graftingβ-cyclodextrin onto the backbone of poly(D,L-lactic acid)(PLA).First,mono(6-(2-aminoethyl)amino-6-deoxy)-β-cyclodextrin(β-CD-6-en)was prepared by sulfonylation and amination ofβ-cyclodextrin and modified poly(D,L-lactic acid)(MPLA)was prepared by free radical polymerization of maleic anhydride and ***,grafting ofβ-cyclodextrin derivative to MPLA backbone was carried out by N-acylation reaction of MPLA andβ-CD-6-en in dimethyl *** synthetic polymer(PLA-β-CD)was characterized by FTIR(Fourier transform infrared)spectroscopy and proton nuclear magnetic resonance(1H NMR)*** results demonstrated that,in the context of remaining the chain structure of PLA,the content ofβ-CD-6-en is 1.6%by the developed synthetic method.
