Osteogenic peptides in collagen hydrolysates:Stimulate differentiation of MC3T3-E1 cells viaβ1 integrin-FAK-ERK1/2 signaling pathway and Smad1 protein

Our previous study demonstrated that orally administration of collagen hydrolysates from silver carp skin(SCH)could increase bone remodeling in chronologically aged ***,the active compounds and molecular mechanism were still *** this study,SCH showed strong activity in promoting osteogenic differentiation of MC3T3-E1 cells and the active components in it were separated by ultrafiltration and YMC ODS-A C18 *** then,a series of peptides containing potential osteogenic sequences were identified by LC-MS/MS and two novel peptides,GARGDKGETGEAGE(GE-14)and TGPRGPVGPA(TA-10),were synthesized for further *** results indicated that peptides GE-14 and TA-10 could promote the differentiation and mineralization of MC3T3-E1 cells by positively regulatingβ1 integrin-FAK-ERK1/2 and Smad1 pathway,which might be due to the RGD or GPR sequences in *** findings suggest that SCH has the potential to be developed as an effective diet supplement to prevent osteoporosis.

Expressions of type I collagen, α2 integrin and β1 integrin in sclera of guinea pig with defocus myopia and inhibitory effects of bFGF on the formation of myopia

AIM:To investigate the expressions of type I collagen, α2 integrin and β1 integrin in the posterior sclera of guinea pigs with defocus myopia and whether basic fibroblast growth factor (bFGF) injection inhibits the formation and development of myopia by upregulating the expression of type I collagen, α2 integrin and β1 integrin. METHODS:After 14 days of treatment, the refractive state and axial length were measured and the levels of type I collagen, α2 integrin and β1 integrin were assayed in the posterior sclerae of groups of guinea pigs that wore a monocular-7D polymethylmethacrylate (PMMA) lens or had -7D lens wear followed by the peribulbar injection of Phosphate Buffer Solution (PBS) or bFGF. The untreated fellow eye served as a control. Guinea pigs with no treatment served as normal group. ·RESULTS:The results showed that 14 days of monocular defocus increased axial eye length and refraction, while bFGF delivery inhibited them markedly. Further, it was also found that the monocular-7D lens could decrease the levels of type I collagen, α2 integrin and β1 integrin expressions, while, unlike PBS, bFGF increased them significantly in comparison to contralateral control eyes and normal eyes. CONCLUSION:bFGF can prevent the formation anddevelopment of defocus myopia by upregulating the expressions of type I collagen, α2 integrin and β1 integrin. Taken together, our results demonstrate that bFGF promotes sclera remodeling to prevent myopia in guinea pigs.

Effect of Astragalin on matrix secretion and β1 integrin mRNA expression in human mesangial cells

Objective To investigate the effect of Astragalin on human renal mesangial cells Methods Cultured human mesangial cells were treated with Astragalin and Astragalin serum in different concentrations in the presence or absence of PDGF BB, the proliferation and type Ⅳ collagen secretion of mesangial cells were measured by MTT assay and ELISA, and expression of β1 integrin gene was estimated by reverse transcription polymerase chain reaction (RT PCR) method, sespectively Results After 72 hours Astragalin or Astragalin serum treatment, the proliferation of mesangial cells induced by PDGF BB was inhibited significantly in a dose dependent manner compared with untreated controls ( P 1) After 24 hours of Astragalin or Astragalin serum treatment, the secretion of type Ⅳ collagen protein in presence of PDGF BB was significantly decreased and β1 integrin mRNA level decreased significantly compared with untreated control ( P 1) Conclusions Astragalin inhibits cell proliferation and matrix over synthesis which might be mediated, at least, partly by decrease of β1 integrin gene over expression The study suggested that Astragalin might play a role in preventing the progression of chronic renal diseases
