β-defensins and the epididymis： contrasting influences of prenatal, postnatal, and adult scenarios

β-defensins are components of host defense, with antimicrobial and pleiotropic immuno-modulatory properties. Research over the last 15 years has demonstrated abundant expression of a variety of β-defensins in the postnatal epididymis of different species. A gradient of region- and cell-specific expression of these proteins is observed in the epithelium of the postnatal epididymis. Their secretion into the luminal fluid and binding to spermatozoa as they travel along the epididymis has suggested their involvement in reproduction-specific tasks. Therefore, continuous attention has been given to various β-defensins for their role in sperm function and fertility. Although β-defensins are largely dependent on androgens, the underlying mechanisms regulating their expression and function in the epididymis are not well understood. Recent investigation has pointed out to a new and interesting scenario where β-defensins emerge with a different expression pattern in the Wolffian duct, the embryonic precursor of the epididymis, as opposed to the adult epididymis, thereby redefining the concept concerning the multifunctional roles of β-defensins in the developing epididymis. In this review, we summarize some current views of β-defensins in the epididymis highlighting our most recent data and speculations on their role in the developing epididymis during the prenatal-to-postnatal transition, bringing attention to the many unanswered questions in this research area that may contribute to a better understanding of epididymal biology and male fertility.

The expression of AVBDs,TLRs and vitamin D hydroxylase associated protein(VDHA) was studied following in vivo by feeding vitamin D.Healthy Silky Fowl were fed vitamin D of 0,800,1600,3200 and 6400 IU/kg feed from 1 to 6w.Real-time PCR analyses reveal that feeding of vitamin D showed significantly changed the expression of AVBDs(AVBD 1,AVBD 5,AVBD 7,AVBD 9,AVBD 10 and AVBD 12) in the thymus, duodenum,jejunum,ileum,cecum and bursa of Fabricius during different dose from 800 to 6400 IU,similarly the expression of TLRs(TLR2,TLR5),VDHA and AVBDs(AVBD-6,GAL-1) in the tissues(liver,spleen and kidney). Effect of Different Concentrations of Lipopolysaccharide on Expression of NF-κB and LAP in Mammary Epithelial Cells of Dairy Cow

The paper was to study the effects of different concentrations of lipopelysaccharide （LPS） on expression of nuclear factor Kappa B （NF-κB） and lingual antimicrobial peptide （LAP） gene in mammary epithelial ceils of dairy cow. The mammary epithelial ceils of dairy cow were stimulated by different concentrations （50, 100,200,400 and 800 ng/mL） of LPS. The total RNA of cells was extracted after stimulation for 2, 4, 8, 16, 24, 48 and 72 h, respectively, and the mRNA expression levels of NF-κB P65 and LAP were evaluated by real-time quantitative PCR. The results showed that the expression of NF-κB P65 and LAP mRNA treated with 400 ng/mL LPS for 72 h were the highest compared to the control group （ P 〈0.01 ）. The result confirmed that the expression activity of NF-κB was enhanced in inflammatory effects of inammary epithelial cells induced by LPS, which regulated the expression of defense gene LAP, with certain dose and time effects.

肝脏表达抗菌肽1(liver-expressed antimicrobial peptide1, LEAP-1)、肝脏表达抗菌肽2(LEAP-2)和p防御素(β-defensin)是一类富含半胱氨酸的阳离子抗菌肽,广泛存在于鱼类中,是鱼类先天免疫系统的重要成分,在抵御病原微生物入侵的过程中发挥着重要作用。LEAP-1在体内外具有抑制细菌、真菌、病毒和原生动物生长的作用,在机体铁平衡的调控方面也发挥着重要作用。LEAP-2作为另外一种重要的抗菌肽,具有抗菌作用。β-defensin除了能直接杀死细菌和病毒外,还可以间接促进效应细胞的补充,诱导多种细胞因子和趋化因子的表达,促进伤口愈合和血管生成。团头鲂是我国重要的淡水经济养殖鱼类。为了阐明LEAP-1、LEAP-2和β-defensin在团头鲂早期发育阶段以及细菌免疫应答中的作用,为后续研究3种抗菌肽在其先天免疫系统中的生物活性奠定理论基础,本研究通过分子克隆技术获得团头鲂LEAP-1、LEAP-2基因全长cDNA序列以及β-defensin基因部分cDNA序列,构建3种基因的时空表达谱,并对免疫注射细菌后3种抗菌肽基因在团头鲂组织中的表达变化进行了研究。主要研究结果如下： 1.团头鲂LEAP-1和LEAP-2基因分子克隆与表达分析团头鲂LEAP-1基因cDNA (GeneBank accession no. JQ308841)全长779bp,其中开放阅读框285bp,5’端非编码区115bp,3’端非编码区379bp。开放阅读框编码94个氨基酸的前体肽,包括24个氨基酸的信号肽,45个氨基酸的前结构域和25个氨基酸的成熟肽。对团头鲂LEAP-1氨基酸序列进行同源性比较,结果表明团头鲂LEAP-1氨基酸序列与草鱼、鳙的相似性最高,达到95%。实时定量PCR分析表明,在胚胎发育阶段,LEAP-1基因mRNA表达量在囊胚中期下降,在眼囊出现期显著增加到最大值,在尾鳍出现期迅速下降到中等水平。出膜后,LEAP-1基因mRNA表达量在出膜后第1和3d逐渐增加到中等水平,在出膜后第7d降到低水平。在团头鲂成鱼组织中,LEAP-1基因mRNA在肝脏中表达量最高。细菌免疫后,团头鲂肝脏中LEAP-1基因mRNA表达量逐渐增加并在注射后144h达到最高(P>0.05)。脑组织中表达量在注射后72h达到最大(P>0.05)。在脾脏和鳃中表达量分别在注射后24h、6h显著上升(P0.05),但是表达量都非常

本试验旨在研究外源L-精氨酸对脂多糖（LPS）诱导猪细胞免疫应激参数与β-defensins-1（pBD-1）,pBD-2和pBD-3 mRNA表达的影响。采用培养至4～5代的猪胎儿皮肤成纤维细胞（3orcineFetal Skin Fibroblasts,PFSF）,通过LPS诱导猪细胞,建立免疫应激模型,在DMEM/F12培养基上添加不同浓度的L-精氨酸,24h后分别收集细胞培养液和提取总RNA,检测细胞培养液乳酸脱氢酶（LDH）、一氧化氮（NO）、一氧化氮合酶（NOS）及溶菌酶（LSZ）应激参数;采用Real-timePCR法检测pBD-1、pBD-2和pBD-3mRNA的表达。结果显示,对非应激细胞,添加70μg/ml L-精氨酸培养液显著降低LDH活性（P＜0.05）;对LPS诱导应激细胞,添加140μg/ml L-精氨酸培养液时,极显著降低LDH活性（P＜0.01）,而后随L-精氨酸浓度增加而上升。与对照组比较,当140μg/ml L-精氨酸培养液培养细胞时,应激或非应激细胞N0含量达到最高,差异极显著（P＜0.01）,随着L-精氨酸浓度的增加,NO含量有下降趋势。LPS应激组细胞培养液中NOS的浓度均高于正常组细胞,L-精氨酸浓度为0μg/ml、35μg/ml和70μg/ml时,应激细胞组NOS显著或极显著高于非应激细胞组（P＜0.05,P＜0.01）。不同浓度L-精氨酸对非应激细胞LSZ无显著影响（P＞0.05）,L-精氨酸为35μg/ml时,应激细胞组LSZ活性最低,L-精氨酸为140μg/ml时,应激细胞LSZ与对照组相同;其他不同浓度L-精氨酸应激组细胞LSZ明显高于对照组。不同浓度外源L-精氨酸对PFSF细胞pBDs mRNA表达具有调节作用,70μg/ml L-精氨酸可显著提高pBD-1和pBD-2基因的mRNA表达水平,140μg/ml L-精氨酸可显著提高pBD-3基因的mRNA表达水平。综上所述,L-精氨酸影响应激或非应激细胞参数变化,猪应激细胞LDH、NO、NOS和LSZ应激参数发生明显改变,LPS诱导猪免疫应激细胞pBD-1、pBD-2和pBD-3基因mRNA表达量均明显高于正常组细胞。结果提示,LPS诱导免疫应激细胞pBD-1、pBD-2和pBD-3基因产生过表达可能是猪机体适应免疫应激的重要方式之一。适宜浓度L-精氨酸可上调应激细胞或非应激细胞pBDs基因mRNA表达,增强猪的抗病。
