Objective Activation of voltage-gated sodium channels(VGSCs) has been related to disorders of neuronal excitability such as epilepsy and *** I,a site-3-specific modulator of VGSCs from the Chinese scorpion Buthus martensi Karsch,has been confirmed to induce spontaneous nociception and hyperalgesia and generate epileptiform responses in ***,the exact mechanism of modulation of BmK I on VGSCs in never cells is still not yet *** present study is to explore the modulation of BmK I on sodium currents eliciting from differentiated SHSY5Y *** Immunofluorescence revealed that two VGSCs subtypes Navl.2,Navl.6 were expressed with highest abundance in SHSY5Y cells treated with retinoic acid for 6 ***,patch-clamp was performed in SHSY5Y cells treated with retinoic acid for 6 days to record the whole-cell *** The inward currents eliciting from differentiated SHSY5Y cells were totally blocked by 100 nM *** inactivation of I was dramatically delayed in the presence of 500 nM BmK I in SHY5Y cells,and the persistent proportion of I was obviously *** I at500 nM can shift the steady-activation curve toward hyperpolarization direction by 4.8 mV(n=24,P<0.05),additionally,after a series of pulse stimuli the steady-inactivation curve shift toward hyperpolarization direction by ***,BmK I at 500 nM shift the fast inactivation curve toward hyperpolarization direction by 9.1mV(n=15,P<0.05).Conclusion BmK I is an effective and novel molecule in the study of VGSCs-related neuronal hyper-excitability.
