Integrated network analysis of transcriptomic and protein-protein interaction data in taurine-treated hepatic stellate cells

BACKGROUND Studies show that the antifibrotic mechanism of taurine may involve its inhibition of the activation and proliferation of hepatic stellate cells(HSCs). Since the molecular mechanism of taurine-mediated antifibrotic activity has not been fully unveiled and is little studied, it is imperative to use "omics" methods to systematically investigate the molecular mechanism by which taurine inhibits liver *** To establish a network including transcriptomic and protein-protein interaction data to elucidate the molecular mechanism of taurine-induced HSC *** We used microarrays, bioinformatics, protein-protein interaction(PPI) network,and sub-modules to investigate taurine-induced changes in gene expression in human HSCs(LX-2). Subsequently, all of the differentially expressed genes(DEGs) were subjected to gene ontology function and Kyoto encyclopedia of genes and genomes pathway enrichment analysis. Furthermore, the interactions of DEGs were explored in a human PPI network, and sub-modules of the DEGs interaction network were analyzed using Cytoscape *** A total of 635 DEGs were identified in taurine-treated HSCs when compared with the controls. Of these, 304 genes were statistically significantly up-regulated, and 331 down-regulated. Most of these DEGs were mainly located on the membrane and extracellular region, and are involved in the biological processes of signal transduction, cell proliferation, positive regulation of extracellular regulated protein kinases 1(ERK1) and ERK2 cascade, extrinsic apoptotic signaling pathway and so on. Fifteen significantly enriched pathways with DEGs were identified, including mitogen-activated protein kinase(MAPK) signaling pathway, peroxisome proliferators-activated receptor signaling pathway,estrogen signaling pathway, Th1 and Th2 cell differentiation, cyclic adenosine monophosphate signaling pathway and so on. Rare earth elements(REEs)are used for the developme nt of new energy materials owing to their intrinsic physicochemical ***,excess REEs in water threaten the safety of animals,plants and *** efficient way to separate REEs from the water is therefore *** this study,a biosorbent consisting of iron oxide(Fe3 O4),persimmon tannin(PT),and graphene oxide(GO)as Fe3 O4/PT/GO was prepared,and the adsorption of trivalent erbium(Er3+)ions from aqueous solution was *** adsorption process for Er3+ions conforms to pseudo-second order kinetic and the Langmuir isotherm model *** studies indicate that the adsorption process is spontaneous and *** electron microscopy(SEM),X-ray photoelectron spectroscopy(XPS),X-ray diffraction(XRD),thermogravimetric analysis(TGA).Fourier-transform infrared(FT-IR)spectroscopy;Brunauer-Emmett-Teller(BET)analysis,and vibrating sample magnetometer(VSM)were used to assess the adsorption mechanism of Er3+ions onto the Fe3 O4/PT/GO biosorbent.A combination of electrostatic interactions,redox reactivity and chelation are responsible for adsorption of Er3+ions on the Fe3 O4/PT/GO biosorbent,The magnetic Fe3 O4/PT/GO biosorbent can be easily separated under the magnetic field for effective recycle of Er3+ions from aqueous ***,this new biomass composite holds great promise for wastewater treatment.

In this study, we propose and demonstrate a broadband polarization-independent terahertz modulator based on graphene/silicon hybrid structure through a combination of continuous wave optical illumination and electrical *** a pump power of 400 mW and the voltages ranging from-1.8 V to 1.4 V, modulation depths in a range of-23%–62% are achieved in a frequency range from 0.25 THz to 0.65 THz. The modulator is also found to have a transition from unidirectional modulation to bidirectional modulation with the increase of pump power. Combining the Raman spectra and Schottky current–voltage characteristics of the device, it is found that the large amplitude modulation is ascribed to the electric-field controlled carrier concentration in silicon with assistance of the graphene electrode and Schottky junction.

Andrographolide(AG)has favorable anti-inflammatory and antioxidative ***,it has low bioavailability due to high lipophilicity and can be easily cleared by the synovial fluid after intra-articular injection,leading to low therapeutic efficiency in osteoarthritis(OA).Herein,we designed a nano-sized pH-responsive drug delivery system(DDS)for OA treatment by using modified mesoporous silica nanoparticles(MSNs)with pH-responsive polyacrylic acid(PAA)for loading of AG to form AG@MSNs-PAA *** nanoparticles have uniform size(120 nm),high drug loading efficiency(22.3860.71%)and pH-responsive properties,beneficial to sustained release in OA *** with AG,AG@MSNs-PAA showed enhanced antiarthritic efficacy and chondro-protective capacity based on IL-1b-stimulated chondrocytes and anterior cruciate ligament transection-induced rat OA model,as demonstrated by lower expression of inflammatory factors and better prevention of proteoglycan ***,the AG@MSNs-PAA nanoplatform may be developed as a promising OA-specific and on-demand DDS.

[Objectives]This study aimed to optimize the extraction process of longan(Dimocarpus longan Lour.)leaves by multi-indicator comprehensive evaluation method.[Methods]The contents of gallic acid ethyl ester,astragalin,quercetin,luteolin and kaempferol in longan leaves were determined simultaneously by *** the multi-indicator comprehensive evaluation method,the methanol concentration,solid to liquid ratio and extraction time for extraction of longan leaves were optimized by orthogonal test.[Results]The optimal extraction process of longan leaves are as follows:methanol concentration of 100%,solid to liquid ratio of 1∶5,and extraction time of 50 min.[Conclusions]The optimized process is simple and feasible,and it can be used to determine the contents of different ingredients in longan leaves.
