Bioprinting of inorganic-biomaterial/neural-stem-cell constructs for multiple tissue regeneration and functional recovery

Tissue regeneration is a complicated process that relies on the coordinated effort of the nervous, vascular and immune systems. While the nervous system plays a crucial role in tissue regeneration, current tissue engineering approaches mainly focus on restoring the function of injury-related cells, neglecting the guidance provided by nerves. This has led to unsatisfactory therapeutic outcomes. Herein, we propose a new generation of engineered neural constructs from the perspective of neural induction, which offers a versatile platform for promoting multiple tissue regeneration. Specifically, neural constructs consist of inorganic biomaterials and neural stem cells(NSCs), where the inorganic biomaterials endows NSCs with enhanced biological activities including proliferation and neural differentiation. Through animal experiments, we show the effectiveness of neural constructs in repairing central nervous system injuries with function recovery. More importantly, neural constructs also stimulate osteogenesis, angiogenesis and neuromuscular junction formation, thus promoting the regeneration of bone and skeletal muscle, exhibiting its versatile therapeutic performance. These findings suggest that the inorganic-biomaterial/NSC-based neural platform represents a promising avenue for inducing the regeneration and function recovery of varying tissues and organs.

Nd^(3+)-sensitized upconversion nanoparticle coated with antimony shell for bioimaging and photothermal therapy in vitro using single laser irradiation

Combining treatment and diagnosis,called theranostics,which is achieved within single nanoparticle is an ultimate goal of many ***,we developed a new nanotheranostic agent-Nd^(3+)-sensitized upconversion nanoparticles core for dual modal imaging(i.e.,upconversion luminescence imaging and magnetic resonance imaging) and antimony nanoshell for photothermal therapy(PTT).The core-shellshell upconversion nanoparticles(NaYF_(4):Yb,Er@NaYF_(4):Yb,Nd@NaGdF_(4):Nd,named as UCNP) were firstly synthesized using thermal decomposition method and then were coated by antimony shell over the surface of UCNP using simple cost and time effective new ***,the surface of UCNP@Sb nanostructures was modified with DSPE-PEG in order to enhance the water solubility and *** final nanotheranostic agent,named as UCNP@Sb-PEG,exhibits very low toxicity,good biocompatibility,very good photothermal therapeutic effect,and efficient upconversion luminescence(UCL) imaging of HeLa cells under only one laser(808 nm) *** antimony shell is quenching the upconversion emission in pristine nanotheranostic agent,but interestingly,the UCL intensity of the agent recovers progressively under 808 nm laser irradiation due to light induced degradability of antimony ***,high longitudinal relaxivity(r_(1)) obtained from the experiment approves excellent potential of the nanotheranostic agent for T_(1)-weighted magnetic resonance imaging application.

Bioinspired laminated bioceramics with high toughness for bone tissue engineering

Activation of autophagy by in situ Zn^(2+) chelation reaction for enhanced tumor chemoimmunotherapy

Chemotherapy can induce a robust T cell antitumor immune response by triggering immunogenic cell death(ICD),a process in which tumor cells convert from nonimmunogenic to immunogenic ***,the antitumor immune response of ICD remains limited due to the low immunogenicity of tumor cells and the immunosuppressive tumor *** autophagy is involved in activating tumor immunity,the synergistic role of autophagy in ICD remains elusive and ***,we report an autophagy amplification strategy using an ion-chelation reaction to augment chemoimmunotherapy in cancer treatments based on zinc ion(Zn^(2+))-doped,disulfiram(DSF)-loaded mesoporous silica nanoparticles(DSF@Zn-DMSNs).Upon pH-sensitive biodegradation of DSF@Zn-DMSNs,Zn2+and DSF are coreleased in the mildly acidic tumor microenvironment,leading to the formation of toxic Zn2+chelate through an in situ chelation ***,this chelate not only significantly stimulates cellular apoptosis and generates damage-associated molecular patterns(DAMPs)but also activates autophagy,which mediates the amplified release of DAMPs to enhance *** vivo results demonstrated that DSF@Zn-DMSNs exhibit strong therapeutic efficacy via in situ ion chelation and possess the ability to activate autophagy,thus enhancing immunotherapy by promoting the infiltration of T *** study provides a smart in situ chelation strategy with tumor microenvironment-responsive autophagy amplification to achieve high tumor chemoimmunotherapy efficacy and biosafety.

Analyzing of mixing performance determination factors for the structure of radial multiple jets-in-crossflow

The radial multiple jets-in-crossflow mixing structure(RMJCMS) is extensively used in industrial manufacture. In this research, the effects of thickness of injection ring on mixing performance and factors influencing the mixing performance of RMJCMS were discussed based on the results of computational fluid dynamics. The simulation results showed that the dimensionless mixing distance, with the increase of the thickness of injection ring, drops from 1.1 to 0.18 first and then increases to 0.27 while the uniformity of flux monotonously improves, manifesting that the consistency of flux is not the single element determining the mixing performance. Analyzing the simulation results, a conclusion was drawn that the consistency of flux, penetration mode and interaction among injection flows which can be altered by adjusting the thickness of injection ring, determine the mixing performance of RMJCMS jointly. That is to say, in RMJCMS an injection ring with a suitable thickness can realize the function of injection and rectification simultaneously, which not only improves the mixing performance but also reduces the complexity of RMJCMS as well.

Nano-preparation promoting effectively luminescent properties of one-dimensional rare earth oxides

Rare earth （La, Nd and Tb） oxides with one-dimensional micro/nanostmcatres were synthesized hydrothermally under facile and mild conditions with two surfactants, and characterized by X-ray diffraction （XRD）, thermal gravimetry （TG）, scanning electron microscopy （SEM）, transmission electron microscopy （TEM） and photoluminescence. The results showed that the synthesized rare earth oxides behaved regular nano- and micro-scale structures. And the morphologies of samples depended on the radii of rare earth ions, with the help of two sur- factants of sodium dodecyl sulfonic and PEG 600. Nanocrystalline La2O3：Eu^3＋ possessed good photoluminescence （PL） property and might be used as a nanosized phosphor, its PL intensity was akered by the doping Eu^3＋ concentration and the optimum concentration of Eu^3＋ was 3 mol.%. In comparison with bulk Eu^3＋/La2O3, Eu^3＋/nano-La2O3 showed better photoluminescence property, nearly equal to that of nanocrtstalline La2O3：Eu^3＋. Tb4O7 microwires showed interesting photoluminescence properties.

A LINEAR DYNAMIC MODEL DESCRIBING LYMPH CIRCULATION

The lymph circulation plays an important role in interstitial fluid balance, protein transport, and immune system. However, its mechanism is poorly understood, and associated pathologies are mostly untreatable. By following Sungawa＇s method of discussing the heart output, a hemodynamic model of lymph circulation was set up. The model was used to demonstrate how the lymphatic fluid formation, investigate the relationship between the lymph flow rate QL and physiological parameters, and explore how local massage influences QL Computer simulation results show that QL is linearly proportional to the interstitial fluid pressure P~, which is similar to experimental observations. Comparing the simulated value of QL during the massage of different frequency with the experimental data from Ikomi shows that they are identical, which indicates that the presented model is rational to explain the mechanism of massage treatment in traditional Chinese medicine （TCM）.

Propagation properties of electromagnetic fields in elliptic dielectric hollow fibres and their applications

We numerically calculate and analyse the electromagnetic fields, optical intensity distributions, polarization states and orbital angular momentum of some elliptic hollow modes in an elliptic dielectric hollow fiber （EDHF） by using Mathieu functions, and also calculate the optical potential of the blue-detuned eHE11 mode evanescent-light wave for ^85Rb atoms, including the position-dependent van der Waals potential, and discuss briefly some potential applications of our EDHF in atom and molecule optics, etc. Our study shows that the vector electric field distributions of the odd modes in the cross section of the EDHF are the same as that of the even modes and with different boundary ellipses by rotating an angle of π/2, and the orbital angular momentum （OAM） of single HE （EH） mode is exactly equal to zero, while that of dual-mode in the EDHF is fractional in h, and has a sinusoidal oscillation as z varies. The EDHF can be used to produce various elliptic hollow beams, even to generate and study various atomic vortices with a fractional charge and its fractional quantum Hall effect in atomic Bose Einstein condensate, and so on.

Structural insight into mechanisms for dynamic regulation of PKM2

Pyruvate kinase isoform M2 （PKM2） converts phospho- enolpyruvate （PEP） to pyruvate and plays an important role in cancer metabolism. Here, we show that post- translational modifications and a patient-derived muta- tion regulate pyruvate kinase activity of PKM2 through modulating the conformation of the PKM2 tetramer. We determined crystal structures of human PKM2 mutants and proposed a ＂seesaw＂ model to illustrate confor- mational changes between an inactive T-state and an active R-state tetramers of PKM2. Biochemical and structural analyses demonstrate that PKM2^Y105E （phos- phorylation mimic of Y105） decreases pyruvate kinase activity by inhibiting FBP （fructose 1,6-bisphosphate）- induced R-state formation, and PKM2K^3305Q （acetylation mimic of K305） abolishes the activity by hindering tet- ramer formation. K422R, a patient-derived mutation of PKM2, favors a stable, inactive T-state tetramer because of strong intermolecular interactions. Our study reveals the mechanism for dynamic regulation of PKM2 by post- translational modifications and a patient-derived muta- tion and provides a structural basis for further investi- gation of other modifications and mutations of PKM2 yet to be discovered.
