Discovery and structure-activity relationship studies of novel tetrahydro-β-carboline derivatives as apoptosis initiators for treating bacterial infections

Developing and excavating new agrochemicals with highly active and safe is an important tactic for protecting crop health and food *** this paper,to discover the new bactericide candidates,we designed,prepared a new type of1,2,3,4-tetrahydro-β-carboline(THC)derivatives and evaluated the in vitro and in vivo bioactivities against the Xanthomonas oryzae ***(Xoo),Xanthomonas axonopodis ***(Xac),and Pseudomonas syringae ***(Psa).The in vitro bioassay results exhibited that most title molecules possessed good activity toward the three plant pathogenic bacteria,the compound A17 showed the most active against Xoo and Xac with EC50 values of 7.27 and 4.89 mg mL^(-1)respectively,and compound A8 exhibited the best inhibitory activity against Psa with EC50value of 4.87 mg mL^(-1).Pot experiments showed that compound A17 exhibited excellent in vivo antibacterial activities to manage rice bacterial leaf blight and citrus bacterial canker,with protective efficiencies of 52.67 and 79.79%at 200 mgmL^(-1),***,compound A8 showed good control efficiency(84.31%)against kiwifruit bacterial canker at 200 mg mL^(-1).Antibacterial mechanism suggested that these compounds could interfere with the balance of the redox system,damage the cell membrane,and induce the apoptosis of Xoo *** together,our study revealed that tetrahydro-β-carboline derivatives could be a promising candidate model for novel broadspectrum bactericides.

Organic conjugated polymers have received extensive attention due to their unique electronic ***,there have been relatively few reports on the dark photocatalytic reactions utilizing organic conjugated ***,we report the successful synthesis of an organic conjugated polymer based on poly(heptazine imide)nanocrystals(CNNCs)for H_(2)O_(2)evolution and biomedical applications using a simple salt molten method and sonication-centrifugation *** results show that these colloid CNNCs have the characteristics of photogenerated electrons accumulation and realize dark photocatalysis with high reducibility under visible light ***,these accumulating photogenerated electrons can reduce O_(2)in darkness to produce H_(2)O_(2).In addition,cytotoxicity tests were conducted and it was found that H_(2)O_(2)produced under dark conditions could oxidize L-arginine(L-Arg)to NO,which effectively killed tumors in the *** work provides an important strategy to construct organic conjugated semiconductor nanocrystals and applying them to future energy and biomedical fields.

Although CRISPR/Cas9 has been widely used in insect gene editing,the need for the microinjection of preblastoderm embryos can preclude the technique being used in insect species with eggs that are small,have hard shells,and/or are difficult to collect and maintain outside of their normal *** is the case with Sogatella furcifera,the white-backed planthopper(WBPH),a significant pest of Oryza sativa(rice)that oviposits inside rice *** extraction from the stem runs the risk of mechanical damage and hatching is heavily influenced by the micro-environment of the rice *** bypass these issues,we targeted embryos prior to oviposition via direct parental(DIPA)-CRISPR,in which Cas9 and single-guide RNAs(sgRNAs)for the WBPH eye pigment gene tryptophan 2,3-dioxygenase were injected into the hemocoel of adult *** at varying numbers of days posteclosion were evaluated to determine at what stage their oocyte might be most capable of taking up the gene-editing *** evaluation of the offspring indicated that the highest G0 gene-edited efficacy(56.7%)occurred in females injected 2 d posteclosion,and that those mutations were heritably transmitted to the G1 *** study demonstrates the potential utility of DIPA-CRISPR for future gene-editing studies in non-model insect species and can facilitate the development of novel pest management applications.

Simultaneously Enhancing Strength, Ductility and Corrosion Resistance of a Martensitic Stainless Steel via Substituting Carbon by Nitrogen

Two martensitic stainless steels of 2Cr12Ni6 type hardened and tempered at 773 K have been studied:the first with 0.2%carbon content and the second with partial replacement of carbon by nitrogen(C0.1N0.1)in the first *** is found that the partial substitution of carbon with nitrogen contributed to an increase in ductility and strength of the steel,presumably due to the formation of more dispersive ***,the addition of nitrogen suppressed the precipitation of carbonitrides,so that the solid solution strengthening effect of C0.1N0.1 did not decrease significantly after tempering *** addition,the partial replacement of carbon by nitrogen contributed to improved ability against pitting corrosion(PC)in chloride-containing medium(3.5%NaCl at 303 K).The higher resistance to PC of tempered nitrogen-containing steel is apparently due to the lower content of massive carbonitrides,especially the reduced aggregation at grain *** leads to a lower acidity and aggressiveness of the test solution near the sample surface due to the accumulation of NH4^(+) ammonium ions in *** a result of nitrogen addition,exception for Cr_(23)C_(6) and VC,Cr_(2)N and(Cr,V)N type precipitates have also been found in C0.1N0.1 steel and this is consistent with the thermodynamic calculation *** conclusion,substituting carbon by nitrogen in traditional martensitic stainless steel could realize the simultaneous improvement of multiple properties of martensitic stainless *** result provides a promising composition optimization route to develop novel martensitic stainless steels.

Numerical Study on the Energy Extraction Performance of Coupled Tandem Flapping Hydrofoils

Tidal current energy is a promising renewable energy source for future electricity *** flapping hydrofoil is regarded as a useful tool to extract the tidal current energy in shallow water.A concept of coupled tandem flapping hydrofoils under semi-activated mode was proposed in the present study.A two-dimensional numerical model,based on the computational fluid dynamics software ANSYS-Fluent,was established to investigate the power extraction performance of the coupled tandem flapping *** effects of the reduced frequency,pitching amplitude,moment of inertia,damping coefficient,and longitudinal distance between hydrofoils were *** vortices,pressure distribution,and kinetic characteristics of hydrofoils under various conditions were analyzed to reveal the interaction between the shedding vortex and *** energy extraction mechanism and hydrodynamic performance were *** positive interactions for energy harvesting were identified for improvements of the further *** peak values of efficiency and power coefficient were achieved at 0.69 and 2.13,respectively.

The time-delay problem,which is introduced by the response time of hardware for correction,is a critical and nonignorable problem of adaptive optics(AO)*** will result in significant wavefront correction errors while turbulence changes severely or system responses *** AO is proposed to alleviate the time-delay problem for more accurate and stable corrections in the real time-varying ***,the existing prediction approaches either lack the ability to extract non-linear temporal features,or overlook the authenticity of spatial features during prediction,leading to poor robustness in ***,we propose a mixed graph neural network(MGNN)for spatiotemporal wavefront *** MGNN introduces the Zernike polynomial and takes its inherent covariance matrix as physical *** takes advantage of conventional convolutional layers and graph convolutional layers for temporal feature catch and spatial feature analysis,*** particular,the graph constraints from the covariance matrix and the weight learning of the transformation matrix promote the establishment of a realistic internal spatial pattern from limited ***,its prediction accuracy and robustness to varying unknown turbulences,including the generalization from simulation to experiment,are all discussed and *** experimental verification,the MGNN trained with simulated data can achieve an approximate effect of that trained with real *** comparing it with two conventional methods,the demonstrated performance of the proposed method is superior to the conventional AO in terms of root mean square error(RMS).With the prediction of the MGNN,the mean and standard deviation of RMS in the conventional AO are reduced by 54.2%and 58.6%at most,*** stable prediction performance makes it suitable for wavefront predictive correction in astronomical observation,laser communication,and microscopic imaging.

Separating overlapping bat calls with a bi-directional long short-term memory network

Acquiring clear acoustic signals is critical for the analysis of animal *** studies commonly face the problem of overlapping signals,which can impede the structural identification of vocal units,but there is currently no satisfactory *** study presents a bi-directional long short-term memory network to separate overlapping echolocation-communication calls of 6 different bat species and reconstruct *** separation quality was evaluated using 7 temporal-spectrum *** the echolocation pulses and syllables of communication calls in the overlapping signals were separated and parameter comparisons showed no significant difference and negligible deviation between the extracted and original *** analysis was conducted with separated echolocation calls from each bat species to provide an example of practical application of the separated and reconstructed *** result of clustering analysis showed high corrected rand index(82.79%),suggesting the reconstructed waveforms could be reliably used for species *** results demonstrate a convenient and automated approach for separating overlapping *** study extends the application of deep neural networks to separate overlapping animal sounds.

Data-driven unsupervised anomaly detection and recovery of unmanned aerial vehicle flight data based on spatiotemporal correlation

Anomaly detection is crucial to the flight safety and maintenance of unmanned aerial vehicles(UAVs)and has attracted extensive attention from ***-based approaches rely on prior knowledge,while model-based approaches are challenging for constructing accurate and complex physical models of unmanned aerial systems(UASs).Although data-driven methods do not require extensive prior knowledge and accurate physical UAS models,they often lack parameter selection and are limited by the cost of labeling anomalous ***,flight data with random noise pose a significant challenge for anomaly *** work proposes a spatiotemporal correlation based on long short-term memory and autoencoder(STCLSTM-AE)neural network data-driven method for unsupervised anomaly detection and recovery of UAV flight ***,UAV flight data are preprocessed by combining the Savitzky-Golay filter data processing technique to mitigate the effect of noise in the original historical flight data on the ***-based feature subset selection is subsequently performed to reduce the reliance on expert ***,the extracted features are used as the input of the designed LSTM-AE model to achieve the anomaly detection and recovery of UAV flight data in an unsupervised ***,the method's effectiveness is validated on real UAV flight data.
