Nested rT-PCr method for the detection of European avian-like H1 swine influenza A virus

Swine influenza A virus（swine IAV） circulates worldwide in pigs and poses a serious public health threat, as evidenced by the 2009 H1N1 influenza pandemic. Among multiple subtypes/lineages of swine influenza A viruses, European avian-like（EA） H1N1 swine IAV has been dominant since 2005 in China and caused infections in humans in 2010. highly sensitive and specific methods of detection are required to differentiate EA H1N1 swine IAVs from viruses belonging to other lineages and subtypes. In this study, a nested reverse transcription（rT）-PCr assay was developed to detect EA H1 swine IAVs. Two primer sets（outer and inner） were designed specifically to target the viral hemagglutinin genes. Specific PCr products were obtained from all tested EA H1N1 swine IAV isolates, but not from other lineages of H1 swine IAVs, other subtypes of swine IAVs, or other infectious swine viruses. The sensitivity of the nested rT-PCr was improved to 1 plaque forming unit（PFU） m L^（-1） which was over 10~4 PFU m L^（-1） for a previously established multiplex rT-PCr method. The nested rT-PCr results obtained from screening 365 clinical samples were consistent with those obtained using conventional virus isolation methods combined with sequencing. Thus, the nested rT-PCr assay reported herein is more sensitive and suitable for the diagnosis of clinical infections and surveillance of EA H1 swine IAVs in pigs and humans.

H9 s ubtype avian influenza virus（AIV） and infectious bronchitis virus（IBV） are major pathogens circulating in poultry and have resulted in great economic losses due to respiratory disease and reduced egg production. As similar symptoms are elicited by the two pathogens, it is difficult for their differential diagnosis. So far, no reverse transcription-polymerase chain reaction（rT-PCr） assay has been found to differentiate between H9 AIV and IBV in one reaction. Therefore, developing a sensitive and specific method is of importance to simultaneously detect and differentiate H9 AIV and IBV. In this study, a duplex rT-PCr（d rT-PCr） was established. Two primer sets target the hemagglutinin（HA） gene of H9 AIV and the nucleocapsid（N） gene of IBV, respectively. Spec ific PCr products were obtained from all tested H9 AIVs and IBVs belonging to the major clades circulating in China, but not from AIVs of other subtypes or other infectious avian viruses. The sensitivity of the d rT-PCr assay corresponding to H9 AIV, IBV and mixture of H9 AIV and IBV were at a concentration of 1×10^1, 1.5×10^1 and 1.5×10^1 50% egg infective doses（EId_（50）） m L^–1, respectively. The concordance rates between the d rT-PCr and virus isolation were 99.1 and 98.2%, respectively, for detection of samples from H9N2 AIV or IBV infected chickens, while the concordance rate was 99.1% for detection of samples from H9N2 AIV and IBV co-infected chickens. Thus, the d rT-PCr assay reported herein is specific and sensitive, and suitable for the differential diagnosis of clinical infections and survei llance of H9 AIVs and IBVs.

Genome-wide analysis of the calcium-dependent protein kinase gene family in Gossypium raimondii

Plant calcium-dependent protein kinases （CdPKs） play important roles in diverse physiological processes by regulating the downstream components of calcium signaling. To date, only a few species of the plant CdPK gene family have been functionally identified. In addition, there has been no systematic analysis of the CdPK family in cotton. Here, 41 putative cotton CdPK （GrCdPK） genes were identified via bioinformatics analysis of the entire genome of Gossypium raimondii and were classified into four groups based on evolutionary relatedness. Gene structure analysis indicated that most of these GrCdPK genes share a similar intron-exon structure （7 or 8 exons）, strongly supporting their close evolutionary relationships. Chromosomal distributions and phylogenetics analysis showed that 13 pairs of GrCdPK genes arose via segmental duplication events. Furthermore, using microarray data of upland cotton （G. hirsutum L.）, comparative profiles analysis of these GhCdPKs indicated that some of the encoding genes might be involved in the responses to multiple abiotic stresses and play important regulatory roles during cotton fiber development. This study is the first genome-wide analysis of the CdPK family in cotton, and it will provide valuable information for the further functional characterization of cotton CdPK genes.

In this paper,a prediction model is developed that combines a Gaussian mixture model(GMM) and a Kalman filter for online forecasting of traffic safety on *** time-to-collision(TTC) samples are divided into two categories:those representing vehicles in risky situations and those in safe ***,the GMM is used to model the bimodal distribution of the TTC samples,and the maximum likelihood(ML) estimation parameters of the TTC distribution are obtained using the expectation-maximization(EM) *** propose a new traffic safety indicator,named the proportion of exposure to traffic conflicts(PETTC),for assessing the risk and predicting the safety of expressway traffic.A Kalman filter is applied to forecast the short-term safety indicator,PETTC,and solves the online safety prediction problem.A dataset collected from four different expressway locations is used for performance *** test results demonstrate the precision and robustness of the prediction model under different traffic conditions and using different *** results could help decision-makers to improve their online traffic safety forecasting and enable the optimal operation of expressway traffic management systems.

A comparative study of differential evolution and genetic algorithms for optimizing the design of water distribution systems

The differential evolution （dE） algorithm has been received increasing attention in terms of optimizing the design for the water distribution systems （WdSs）. This paper aims to carry out a comprehensive performarice comparison between the new emerged dE algorithm and the most popular algorithm-the genetic algorithm （GA）. A total of six benchmark WdS case studies were used with the number of decision variables ranging from 8 to 454. A preliminary sensitivity analysis was performed to select the most effective parameter values for both algorithms to enable the fair comparison. It is observed from the results that the dE algorithm consistently outperforms the GA in terms of both efficiency and the solution quality for each case study. Additionally, the dE algorithm was also compared with the previously published optimization algorithms based on the results for those six case studies, indicating that the dE exhibits comparable performance with other algorithms. It can be concluded that the dE is a newly promising optimization algorithm in the design of WdSs.

Morphological,Biochemical and Genetic Analysis of a Brittle Stalk Mutant of Maize Inserted by Mutator

Mutants on stalk strength are essential materials for the studies on the formation of plant cell *** this study,a brittle stalk mutant of maize,designated as Bk-x,was screened from a Mutator inserted mutant *** the germination and early seedling stage,the mutant plants were indistinguishable from the normal ***,all of the plant organs were brittle after the 5th-leaf stage and remained brittle throughout the rest of the growing *** observation showed that the cell wall in vascular bundle sheath of Bk-x was thinner than that in normal *** leaf mechanical strength in Bk-x was 77.9% of that in normal plants growing at Xishuangbanna（BN）,Yunnan province and that was 61.7% in Wuhan（WH）,Hubei Province,*** proportion of cellulose was 12.3% in Bk-x,which was significantly lower than that in normal plants（26.7%）,while the soluble sugar content was 36.1% in Bk-x,which is significantly higher than that in normal plants（12.4%）.Genetic analysis using two F 2 populations and one F 2：3 families demonstrated that the trait of brittle stalk is controlled by a single recessive gene.

Polypropylene composite nonwovens containing rare-earth strontium aluminates Sr Al2O4：Eu2＋,dy3＋ and functional additives were fabricated by the spun-bonded *** optical properties, morphology and mechanical properties of the samples were *** from scanning electron microscopy photographs（SEM） indicated that the surface of the fiber was destroyed by the addition of rare earth luminescent materials lightly but the thickness of the fiber was *** scanning calorimetry results showed that pure polypropylene has the double crystallization peak at 162.3 and 165.1 °*** from X-ray diffraction showed that the nonwoven prepared with the luminescent materials contained the α-monoclinic crystal and β crystalline ***, the afterglow properties were tested, which showed that the afterglow curve of the luminous nonwoven was similar to that of strontium aluminate, and the intensity was more intensive than luminous nonwoven at the *** nonwoven fabricated with the luminescent material did not affect the crystal lattice of the polymer making the materials have potential applications in fluorescent lamps and field emission displays（FEds）.
