Bone morphogenetic protein-6 suppresses TGF-β_(2)-induced epithelial-mesenchymal transition in retinal pigment epithelium

AIM:To evaluate the effect of bone morphogenetic protein-6(BMP-6)on transforming growth factor(TGF)-β_(2)-induced epithelial-mesenchymal transition(EMT)in retinal pigment epithelium(RPE).METHODS:Adult retinal pigment epithelial cell line(ARPE-19)were randomly divided into control,TGF-β_(2)(5μg/L),and BMP-6 small interfering RNA(siRNA)group.The cell morphology was observed by microscopy,and the cell migration ability were detected by Transwell chamber.The EMT-related indexes and BMP-6 protein levels were detected by Western blotting.Furthermore,a BMP-6 overexpression plasmid was constructed and RPE cells were divided into the control group,TGF-β_(2)+empty plasmid group,BMP-6 overexpression group,and TGF-β_(2)+BMP-6 overexpression group.The EMT-related indexes and extracellular regulated protein kinases(ERK)protein levels were detected.RESULTS:Compared with the control group,the migration of RPE cells in the TGF-β_(2) group was significantly enhanced.TGF-β_(2) increased the protein expression levels ofα-smooth muscle actin(α-SMA),fibronectin and vimentin but significantly decreased the protein levels of E-cadherin and BMP-6(P<0.05)in RPE.Similarly,the migration of RPE cells in the BMP-6 siRNA group was also significantly enhanced.BMP-6 siRNA increased the protein expression levels ofα-SMA,fibronectin and vimentin but significantly decreased the protein expression levels of E-cadherin(P<0.05).Overexpression of BMP-6 inhibited the migration of RPE cells induced by TGF-β_(2) and prevented TGF-β_(2) from affecting EMT-related biomarkers(P<0.05).CONCLUSION:BMP-6 prevents the EMT in RPE cells induced by TGF-β_(2),which may provide a theoretical basis for the prevention and treatment of proliferative vitreoretinopathy.

An advanced 2D/3D g-C_(3)N_(4)/TiO_(2)@MnO_(2) multifunctional membrane for sunlight-driven sustainable water purification

Graphitic carbon nitride(g-C_(3)N_(4))nanosheets have attracted widespread interest in the construction of advanced separation membranes.However,dense stacking and a single functionality have limited the membrane development.Here,an advanced two-/three-dimensional(2D/3D)g-C_(3)N_(4)/TiO_(2)@MnO_(2) membrane is constructed by intercalating 3D TiO_(2)@MnO_(2) nanostructures into g-C_(3)N_(4) nanosheets.The 3D flower-like nanostructures broaden the transport channels of the composite membrane.The membrane can effectively separate five oil-in-water(O/W)emulsions,with a maximum flux of 3265.67±15.01 L·m^(-2)·h^(-1)·bar^(-1) and a maximum efficiency of 99.69%±0.45%for toluene-in-water emulsion(T/W).Meanwhile,the TiO_(2)@MnO_(2) acts as an excellent electron acceptor and provides positive spatial separation of electrons–holes(e^(-)–h^(+)).The formation of 2D/3D heterojunctions allows the material with wider light absorption and smaller bandgap(2.10 eV).These photoelectric properties give the membrane good degradation of three different pollutants,with about 100%degradation for methylene blue(MB)and malachite green(MG).The photocatalytic antibacterial efficiency of the membrane is also about 100%.After cyclic experiment,the membrane maintains its original separation and photocatalytic capabilities.The remarkable multifunctional and self-cleaning properties of the g-C_(3)N_(4) based membrane represent its potential value for complex wastewater treatment.

BACKGROUND The magnetic compression technique has been used to establish an animal model of tracheoesophageal fistula(TEF),but the commonly shaped magnets present limitations of poor homogeneity of TEF and poor model control.We designed a Tshaped magnet system to overcome these problems and verified its effectiveness via animal experiments.AIM To investigate the effectiveness of a T-shaped magnet system for establishing a TEF model in beagle dogs.METHODS Twelve beagles were randomly assigned to groups in which magnets of the Tshaped scheme(study group,n=6)or normal magnets(control group,n=6)were implanted into the trachea and esophagus separately under gastroscopy.Operation time,operation success rate,and accidental injury were recorded.After operation,the presence and timing of cough and the time of magnet shedding were observed.Dogs in the control group were euthanized after X-ray and gastroscopy to confirm establishment of TEFs after coughing,and gross specimens of TEFs were obtained.Dogs in the study group were euthanized after X-ray and gastroscopy 2 wk after surgery,and gross specimens were obtained.Fistula size was measured in all animals,and then harvested fistula specimens were examined by hematoxylin and eosin(HE)and Masson trichrome staining.RESULTS The operation success rate was 100%for both groups.Operation time did not differ between the study group(5.25 min±1.29 min)and the control group(4.75 min±1.70 min;P=0.331).No bleeding,perforation,or unplanned magnet attraction occurred in any animal during the operation.In the early postoperative period,all dogs ate freely and were generally in good condition.Dogs in the control group had severe cough after drinking water at 6-9 d after surgery.X-ray indicated that the magnets had entered the stomach,and gastroscopy showed TEF formation.Gross specimens of TEFs from the control group showed the formation of fistulas with a diameter of 4.94 mm±1.29 mm(range,3.52-6.56 mm).HE and Masson trichrome staining showed scar tissue formation and hierarchical structural

BACKGROUND Magnetic anchor technique(MAT)has been applied in laparoscopic cholecystectomy and laparoscopic appendectomy,but has not been reported in laparoscopic partial hepatectomy.AIM To evaluate the feasibility of the MAT in laparoscopic left lateral segment liver resection.METHODS Retrospective analysis was conducted on the clinical data of eight patients who underwent laparoscopic left lateral segment liver resection assisted by MAT in our department from July 2020 to November 2021.The Y-Z magnetic anchor devices(Y-Z MADs)was independently designed and developed by the author of this paper,which consists of the anchor magnet and magnetic grasping apparatus.Surgical time,intraoperative blood loss,intraoperative accidents,operator experience,postoperative incision pain score,postoperative complications,and other indicators were evaluated and analyzed.RESULTS All eight patients underwent a MAT-assisted laparoscopic left lateral segment liver resection,including three patients undertaking conventional 5-port and five patients having a transumbilical single-port operation.The mean operation time was 138±34.32 min(range 95-185 min)and the mean intraoperative blood loss was 123±88.60 mL(range 20-300 mL).No adverse events occurred during the operation.The Y-Z MADs showed good workability and maneuverability in both tissue and organ exposure.In particular,the operators did not experience either a“chopstick”or“sword-fight”effect in the single-port laparoscopic operation.CONCLUSION The results show that the MAT is safe and feasible for laparoscopic left lateral segment liver resection,especially,exhibits its unique abettance for transumbilical single-port laparoscopic left lateral segment liver resection.

Temporal and environmental factors drive community structure and function of methanotrophs in volcanic forest soils

Methanotrophs,organisms that obtain oxygen by oxidizing methane,are recognized as the only known biological sink for atmospheric CH_4,and forest soil methanotrophs play crucial roles in mitigating global warming.The succession patterns of methanotrophic communities and functions in Wudalianchi volcano forest soils could provide a basis for the study of evolutionary mechanisms between soil microorganisms,the environment,and carbon cycling of temperate forest ecosystems under climate change.In this study,the characteristics and drivers of methanotrophic community structure and function of two volcanic soils at different stages of development are analyzed,including an old volcano and a new volcano,which most recently erupted 300 years and 17-19×10^(5)years ago,respectively,and a non-volcano hills as control,based on space for time substitution and Miseq sequencing and bioinformation technology.The results showed that CH_(4) fluxes were significantly higher in old-stage volcano forest soils than new-stage forest soils and non-volcano forest soils.There were significant differences in the community composition and diversity of soil methanotrophs from different volcano forest soils.Methylococcus was the dominant genus in all soil samples.Additionally,the relative abundance of Methylococcus,along with Clonothrix,Methyloglobulus,Methylomagum,Methylomonas and Methylosarcina,were the important genera responsible for the differences in methanotrophic community structure in different volcano forest soils.The relative abundance of methanotroph belonging toγ-proteobacteria was significantly higher than that belonging toα-proteobacteria(P<0.05).Chao1,Shannon and Simpson indices of soil methanotrophic community were significantly lower in new-stage volcanos and were significantly affected by bulk density,total porosity,p H,nitrate,dissolved organic carbon and dissolved organic nitrogen.There were significant differences in community structure between new-stage and old-stage volcanoes.Bulk density and p H are important soil prope

The high mortality of patients with lung squamous cell carcinoma(LUSC)results from metastasis rather than primary tumors,1 whereas the molecular pathogenesis of cancer metastasis remains poorly understood.PLAU encodes urokinase-type plasminogen activator(uPA)which is closely related to tumor diagnosis,treatment target,and prognosis of patients.2.3 We previously predicted that PLAU is a key gene negatively associated with overall survival by integrated multidimensional analyses of The Cancer Genome Atlas(TCGA)datasets from 504 samples of LUSC tumor tissues and 46 samples of adjacent non-tumorous lung tissues.4 However,whether the role of PLAU contributes to the metastasis of LUC remains unclear.Here,we analyzed the association between uPA levels and LUSC and found that uPA levels were associated with pathological variations in LUSC patients.

Selenium nanoparticles in aquaculture: Unique advantages in the production of Se-enriched grass carp(Ctenopharyngodon idella)

The production of selenium-enriched fish can contribute to alleviating selenium deficiency in human diets. However, it is still unclear which selenium source, as an additive, can efficiently and costeffectively produce high-quality selenium-enriched fish. This study evaluated the effects of selenium nanoparticles（SeNP）, selenite, and selenomethionine（SeMet） on the growth, antioxidant capacity, selenium content, selenium speciation, and meat quality of grass carp. Ten diets were prepared, including a basal diet（BD） and three concentrations（0.1, 0.3, and 0.9 mg/kg） of SeNP, selenite, and SeMet. A total of600 fish（250.79 ± 1.57 g） were randomly assigned to 30 tanks（3 tanks/group）. Fish were fed the experimental diet three times daily for 60 d. In this study, SeNP most significantly promoted the growth and antioxidant capacity of grass carp, with 0.3 mg/kg SeNP identified as the optimal additive concentration. Additionally, SeNP demonstrated equally excellent bioavailability as SeMet and significantly increased the content of SeMet in grass carp（Ctenopharyngodon idella） muscle. Furthermore, compared to SeMet and selenite, dietary SeNP could more significantly enhance the content of selenocysteine（SeCys2） and methylselenocysteine（MeSeCys） in grass carp muscle tissue. In addition, we have demonstrated that SeCys2and MeSeCys promote apoptosis of cancer cells（HeLa） through the mitochondrial apoptotic pathway（involving Bax and Bcl-2）. Furthermore, as an additive, 0.3 mg/kg SeNP significantly improved the flesh quality of grass carp by reducing crude fat and heavy metal content, as well as increasing the levels of eicosapentaenoic acid（EPA） and docosahexaenoic acid（DHA） and the ratio of n-3/n-6 polyunsaturated fatty acid（PUFA）. In summary, SeNP is the most suitable additive for producing selenium-enriched fish.

The strong pyrocatalytic dye decomposition of the BaTiO_(3)/Pr_(2)O_(3) heterojunction catalyst under cold–hot alternation conditions has been demonstrated in this work.For pure BaTiO_(3) nanofibers,~54%rhodamine B(RhB)dye is decomposed under the cold–hot alternation of 29–57℃.With the loading content of Pr_(2)O_(3) increases from 0 to 4 wt%,the pyrocatalytic decomposition ratio of RhB solution increases first and then decreases,eventually achieving a maximum of 91%at 3 wt%.The enhanced pyrocatalytic performance after loading Pr_(2)O_(3) can be attributed to an internal electric field of the heterojunction,which effectively separates positive and negative charges.The strongly pyrocatalytic performance of BaTiO_(3)/Pr_(2)O_(3) makes it hopeful for applications in the dye wastewater treatment through harvesting the environmental cold–hot temperature alternation thermal energy in future.

Learning to select the recombination operator for derivative-free optimization

Extensive studies on selecting recombination operators adaptively,namely,adaptive operator selection(AOS),during the search process of an evolutionary algorithm(EA),have shown that AOS is promising for improving EA's performance.A variety of heuristic mechanisms for AOS have been proposed in recent decades,which usually contain two main components:the feature extraction and the policy setting.The feature extraction refers to as extracting relevant features from the information collected during the search process.The policy setting means to set a strategy(or policy)on how to select an operator from a pool of operators based on the extracted feature.Both components are designed by hand in existing studies,which may not be efficient for adapting optimization problems.In this paper,a generalized framework is proposed for learning the components of AOS for one of the main streams of EAs,namely,differential evolution(DE).In the framework,the feature extraction is parameterized as a deep neural network(DNN),while a Dirichlet distribution is considered to be the policy.A reinforcement learning method,named policy gradient,is used to train the DNN.As case studies,the proposed framework is applied to two DEs including the classic DE and a recently-proposed DE,which result in two new algorithms named PG-DE and PG-MPEDE,respectively.Experiments on the Congress of Evolutionary Computation(CEC)2018 test suite show that the proposed new algorithms perform significantly better than their counterparts.Finally,we prove theoretically that the considered classic methods are the special cases of the proposed framework.

Formatted PVDF in lamellar composite solid electrolyte for solidstate lithium metal battery

Solid polymer electrolytes(SPEs)hold great application potential for solid-state lithium metal battery because of the excellent interfacial contact and processibility,but being hampered by the poor room-temperature conductivity(~10^(−7)S·cm^(−1))and low lithium-ion transference number(tLi+).Here,a lamellar composite solid electrolyte(Vr-NH_(2)@polyvinylidene fluoride(PVDF)LCSE)withβ-conformation PVDF is fabricated by confining PVDF in the interlayer channel of-NH_(2)modified vermiculite lamellar framework.We demonstrate that the conformation of PVDF can be manipulated by the nanoconfinement effect and the interaction from channel wall.The presence of-NH_(2)groups could induce the formation ofβ-conformation PVDF through electrostatic interaction,which serves as continuous and rapid lithium-ion transfer pathway.As a result,a high room-temperature ionic conductivity of 1.77×10^(−4)S·cm^(−1)is achieved,1-2 orders of magnitude higher than most SPEs.Furthermore,Vr-NH_(2)@PVDF LCSE shows a high tLi+of 0.68 because of the high dielectric constant,~3 times of that of PVDF SPE,and surpassing most of reported SPEs.The LiNi_(0.8)Co_(0.1)Mn_(0.1)O_(2)||Li cell assembled by Vr-NH_(2)@PVDF LCSE obtains a discharge specific capacity of 137.1 mAh·g^(−1)after 150 cycles with a capacity retention rate of 93%at 1 C and 25℃.This study may pave a new avenue for high-performance SPEs.
