Vaccines are one of the most practical means to stop the spreading of Aeromonas veronii in *** this study,virulence factor aerolysin mutant NTaer which has lost its hemolytic activity was used as a target *** pastoris constitutive secretory expression NTaer(GS115-NTaer)was used as a potential safe oral vaccine to evaluate its effectiveness on zebrafish *** result shows that vaccination of GS115-NTaer for four weeks did not affect the growth performance of the host,while eliciting an effective immune protective *** with the control group,the GS115-NTaer could significantly up-regulate the relative expression level of the intestinal tight junction protein 1α(TJP1α)gene,and significantly increased the contents of lysozyme(LYZ),complement C3 and C4 in the gut,indicating that the innate immune response of the fish was *** relative gene expression levels of macrophage-expressed gene 1(MPEG1)and T cell receptor(TCR-α)in the gut,and MPEG1,CD4,CD8,TCR-α,GATA3,and T-bet in the spleen were all increased significantly,indicating that the cellular immune response of the fish was ***,the contents of serum IgM and intestinal mucosa IgZ antibodies were significantly increased,which showed that humoral immunity was also ***,inoculation with GS115-NTaer significantly changed the structure of gut *** particular,the relative ratio of(Firmicutes+Fusobacteriota+Bacteroidota)/Proteobacteria was significantly higher than that of the control and GS115 ***,the vaccinated fish were challenged with ***,and the relative percent survival of GS115 and the GS115-NTear groups was 14.28%and 33.43%.This improvement of immunity was not only due to the specific immune response but also attributed to the improvement of innate immunity and the gut microbiota which was demonstrated by the germ-free zebrafish ***,this study provides information on the effectiveness of GS115-NTear

Verticillium wilt(VW),induced by the soil-borne fungus Verticillium dahliae(Vd),poses a substantial threat to a diverse array of plant *** molecular breeding technology for the development of cotton varieties with heightened resistance to VW stands out as one of the most efficacious protective *** this study,we successfully generated two stable transgenic lines of cotton(Gossypium hirsutum L.),VdThitRNAi-1 and VdThit-RNAi-2,using host-induced gene silencing(hiGS)technology to introduce double-stranded RNA(dsRNA)targeting the thiamine transporter protein gene(VdThit).Southern blot analysis confirmed the presence of a single-copy insertion in each *** examination showed marked reductions in the colonization and spread of Vd-mCherry in the roots of VdThit-RNAi cotton compared to wild type(WT).The corresponding disease index and fungal biomass of VdThit-RNAi-1/2 also exhibited significant ***-time quantitative PCR(qRT-PCR)analysis demonstrated a substantial inhibition of VdThit expression following prolonged inoculation of VdThit-RNAi *** RNA sequencing(sRNA-Seq)analysis revealed the generation of a substantial number of VdThit-specific siRNAs in the VdThit-RNAi transgenic ***,the silencing of VdThit by the siVdThit produced by VdThit-RNAi-1/2 resulted in the elevated expression of multiple genes involved in the thiamine biosynthesis pathway in *** field conditions,VdThit-RNAi transgenic cotton exhibited significantly enhanced disease resistance and yield compared with *** summary,our findings underscore the efficacy of hiGS targeting VdThit in restraining the infection and spread of Vd in cotton,thereby potentially enabling the development of cotton breeding as a promising strategy for managing VW.

Two alternative modes for diffuse pollution control in Wuhan City Zoo

Pollution in urban zoos arises from diffuse and small point sources. However, its control has received little attention in past decades. Online and ofttine modes of ecological engineering technology were designed to control pollution from small point and diffuse sources in Wuhan City Zoo, China. Their characteristics and performances were investigated in sixteen runoff events from 2003 to 2005. The results showed that the two modes both improved runoff water quality and had high retention rates for water and pollutants. In the outflows, the event mean concentrations （EMCs） of total suspended solids （TSS）, chemical oxygen demand （COD）, total nitrogen （TN） and total phosphorus （TP） were reduced by 88%, 59%, 46% and 71% for the online mode, and those were 77%, 42%, 50% and 66% for the ofttine mode. The annual retention rates of pollutant loads for the online mode were 94.9%-98.5% in the three study years; those for the ofitine mode were 70.5%-86.4%. Based on calculation, the online mode was able to store the runoff of 66.7 mm rainfall completely, and the ofttine mode could store that of 31.3 mm rainfall. In addition, the online mode can provide an effective way for rainwater utilization and good habitats for aquatic wildlives, and has an excellent aesthetics value for recreationsal pastimes. The ofiline mode can save land resources and may be an effective and economical measure for diffuse pollution control in urban areas.

The density-based clustering algorithm presented is different from the classical Density-Based Spatial Clustering of Applications with Noise (DBSCAN) (Ester et al., 1996), and has the following advantages: first, Greedy algorithm substitutes for R*-tree (Bechmann et al., 1990) in DBSCAN to index the clustering space so that the clustering time cost is decreased to great extent and I/O memory load is reduced as well; second, the merging condition to approach to arbitrary-shaped clusters is designed carefully so that a single threshold can distinguish correctly all clusters in a large spatial dataset though some density-skewed clusters live in it. Finally, authors investigate a robotic navigation and test two artificial datasets by the proposed algorithm to verify its effectiveness and efficiency.

The plasma-neutral gas structure generated in a magnesium cathodic arc operated with oxygen gas at a constant current of 50 A has been investigated by employing a simplified one-dimensional fluid model. The model includes elastic collisions and charge-exchange reactions between metallic particles and gas molecules, and also generation and recombination of gaseous ions by electron impact. The distribution profiles of density and velocity of species along the axial direction were obtained at different background gas pressures （in the range of 0.7-3.0 Pa） by this model. A comparison with the experiments was made. At lower gas pressures, the depositing particles were mainly the metallic ions with a larger kinetic energy. As the gas pressure increased, the magnesium atoms with smaller kinetic energy acted as the dominant depositing species. Determined by the minimization of the system＇s total energy, MgO（100） or/and MgO（110） orientation appeared easily in the MgO films at lower gas pressures, and at higher gas pressures, the film preferred orientation was MgO（111）.

A kinetic study on the sulfuric acid leaching of multi-metal oxide, which is the product of multi-metal copper alloy with iron trioxide roasted in oxygen, was carried out. The effects of leaching time, stirring speed, sulfuric acid concentration, reaction temperature, and particle size of the multi-metal oxide on the kinetics and mechanism of copper extraction were studied. It was found that the reaction kinetic model about the copper extraction from multi-metal oxide follows the mixed kinetic shrinking core mode： 1/31n（1-X）＋（1-X）-l/3-1=680.5C（H2SO4）0.4297dp0.75115exp（-Ea/RT）t.

Two biological aerated filters （BAF） were setup for ammonia removal treatment of the circulation water in a marine aquaculture. One of the BAFs was bioaugmented with a heterotrophic nitrifying bacterium, Lutimonas sp. H10, where the ammonia removal was not improved and the massive inoculation was even followed by a nitrification breakdown from day 9 to 18. The nitrification was remained stable in control BAF operated under the same conditions. Fluorescent in situ hybridization （FISH） with rRNA-targeted probes and cultivable method revealed that Lutimonas sp. H10 almost disappeared from the bioaugomented BAF within 3 d, and this was mainly due to the infection of a specific phage as revealed by flask experiment, plaque assay and transmission electron observation. Analyses of 16S rRNA gene libraries showed that bacterial groups from two reactors evolved differently and an overgrowth of protozoa was observed in the bioaugmented BAF. Therefore, phage infection and poor biofilm forming ability of the inoculated strain are the main reasons for bioaugmentation failure. In addition, grazing by protozoa of the bacteria might be the reason for the nitrification breakdown in bioaugmented BAF during day 9-18.

FEM Analysis on Acoustic Performance of Wall Flow Diesel Particulate Filters

Diesel powered vehicles, in compliance with the more strict exhaust emission standards such as Euro V, is likely to require a diesel particulate filter （DPF）. A DPF used on a vehicle will affect the acoustic emission of the diesel engine, so it is important to investigate the sound propagation rule in DPF and further to propose the optimum DPF design. However, due to the geometrical complexity of the DPF, the traditional analysis method, such as analytical method, can not assess the acoustic performance of DPF accurately in medium and high frequency band. In this paper, a combined approach of finite element analysis and viscosity correction is proposed to predict acoustic performance of DPF. A simplified model of the full DPF is established and is used to analyze the sound propagation characteristic of the DPF. The distribution of the sound pressure and velocity, the transmission matrix of the DPF are obtained using the finite element method. In addition, the method of the viscosity correction is used in the transmission matrix of the DPF to evaluate the acoustic performance of DPF. Based on the FEM computation and the viscosity correction, the transmission losses under the rated load and idle condition of a diesel engine are calculated. The calculation results show that DPF can effectively attenuate exhaust noise, and sound attenuation increase with the rise of the frequency. Sound attenuation is better under rated condition than idle condition of diesel engine, particularly in frequency above 1 000 Hz.
