Asip is a mammalian homologue of polarity protein Par-3 of Caenorhabditis elegans and Bazooka of Drosophila melanogaster. Asip/Par-3/Bazooka are PDZ-motif containing proteins that localize asymmetrically to the cell periphery and play a pivotal role in cell polarity and asymmetric cell division. In the present study, we have cloned human asip cDNA and its splicing variants by 5’-RACE and RT-PCR using candidate human EST clones which have a high homology to rat asip cDNA. The full-length cDNA of human asip encodes a 1,353 aa protein exhibiting 88% similarity to the rat one. Human asip is a single copy gene consisting of at least 26 exons and localizing in human chromosome 10, band p11.2, with some extraordinarily long introns. All exon/intron boundary nucleotides conform to the "gt-ag" rule. Three main transcripts were detected by Northern blot analysis, and at least five variants, from alternative splicing and polyadenylation, have been identified by RT-PCR and liver cDNA library screening. Exon 17b deleted asip mRNAs expressed ubiquitously in normal human tissues, including liver, on RT-PCR analysis. However, they were absent from most human liver cancer cell lines examined. More interestingly, the expression of exon 1 7b deleted variants was down regulated in 52.6% (10/19) clinic specimens of human hepatocellular carcinomas (HCCs), compared with the surrounding nontumorous liver tissues from the same patients. The presence of various splicing transcripts, the variation of their distribution among different tissues and cells, and their differential expressions in human HCCs suggest that human Asip isoforms may function in different context.

Using time-resolved laser-induced fluorescence(LIF)spectroscopy,the fluorescence quantum beat decay was observed in C(1B_(2))←X(1A_(1))transition of SO_(2)*** experimental results show that only the v_(3) mode and its combination band v_(1)+v_(2)+v_(3) excitation exist the beat *** the result,we attribute the quantum beat states to the coupling of the v3 vibration mode of C state with the high vibrational levels of the electronic ground state.

The 10-year climatological features related to midlatitude double tropopause events （DTs） are examined using ERA- Interim data from 2003 to 2012. The analysis is based on tropopauses defined by lapse rate. Results show that DTs are permanent or semi-permanent in the midlatitudes, and high DT frequency bands move poleward in winter and equatorward in summer, which is consistent with the seasonal movement of the subtropical jet. Based on our statistics, the second tropopause is found at about 100 hPa in the subtropics and at slightly lower altitudes in sub-polar regions. The thickness between the first and second tropopause is smaller in the subtropics and increases with latitude. Next, the origin of air sandwiched between the first and second tropopause of DTs is studied with a revised version of the UK Universities Global Atmospheric Modelling Programme Offline Trajectory Code （Version 3） diabatic trajectory model. The results show that, in the lower or middle troposphere, air is transported into the DTs from lower latitudes, mainly in the tropics. The dominant source regions are mainly areas of deep convection and steep orography, e.g., the western Pacific and Himalayan Mountains, and they show strong seasonality following the seasonal shift of these strong upwelling regions.

The genetic models are greatly important in the analysis of genetic epidemiologic studies and many of the studies are conducted using the trend test under the additive model. However, for many complex diseases and traits, the underlying genetic model for a genetic locus is usually uncertain. So a robust test free of genetic model is appropriate. In this paper, the authors propose a model-embedded trend test by incorporating Hardy-Weinberg equilibrium information and obtain the explicit formula to calculate its statistical significance. Extensive simulation studies show the proposed test is more robust than the existing procedures. Finally, a real application is further analyzed to show the performance of the proposed test.

Snow depth is a general input variable in many models of agriculture,hydrology,climate and *** study makes use of observational data of snow depth and explanatory variables to compare the accuracy and effect of geographically weighted regression kriging(GWRK)and regression kriging(RK)in a spatial interpolation of regional snow *** auxiliary variables are analyzed using correlation coefficients and the variance inflation factor(VIF).Three variables,Height,topographic ruggedness index(TRI),and land surface temperature(LST),are used as explanatory variables to establish a regression model for snow *** estimated spatial distribution of snow depth in the Bayanbulak Basin of the Tianshan Mountains in China with a spatial resolution of 1 km is *** results indicate that 1)the result of GWRK's accuracy is slightly higher than that of RK(R^2=0.55 vs.R^2=0.50,RMSE(root mean square error)=0.102 m ***=0.077 m);2)for the subareas,GWRK and RK exhibit similar estimation results of snow *** in the Bayanbulak Basin with a snow depth greater than 0.15m are mainly distributed in an elevation range of 2632.00–3269.00 m and the snow in this area comprises 45.00–46.00% of the total amount of snow in this ***,the GWRK resulted in more detailed information on snow depth distribution than the *** final conclusion is that GWRK is better suited for estimating regional snow depth distribution.

Application of a nuclear localization signal gene in transgene mice

Efficient gene transfer by cytoplasm co-injec-tion will offer a powerful means for transgenic animals. Us-ing co-injection in cytoplasm, two independent gene con-structs, including bovine a-s1-casein-hG-CSF and a mammal expression vector expressing a nuclear localization signal (mNLS), were introduced into fertilized mouse eggs. The target gene construct was docked into host nucleus probably by the nuclear localization signal. Transgene mice have been obtained at 58% (29/50) of integration ratio. Ex-pression level of the positive transgene mice was detected by Western blotting. Maximal expression of human G-CSF was estimated about 540 mg/L of milk. The expression ratio was up to 75% (9/12). The results here have important practical implications for the generation of mammary gland bioreac-tors and other transgene studies. Co-injection of a target gene with an expression vector of a mammal nuclear localization signal by cytoplasm appears to be a useful, efficient and easy strategy for generating

Cloud structure and evolution of Mesoscale Convective Systems （MCSs） retrieved from the Tropical Rainfall Measuring Mission Microwave Imager （TRMM TMI） and Precipitation Radar （PR） were investigated and compared with some pioneer studies based on soundings and models over the northern South China Sea （SCS）. The impacts of Convective Available Potential Energy （CAPE） and environmental vertical wind shear on MCSs were also explored. The main features of MCSs over the SCS were captured well by both TRMM PR and TMI. However, the PR-retrieved surface rainfall in May was less than that in June, and the reverse for TMI. TRMM-retrieved rainfall amounts were generally consistent with those estimated from sounding and models. However, rainfall amounts from sounding-based and PR-based estimates were relatively higher than those retrieved from TRMM-TMI data. The Weather Research and Forecasting （WRF） modeling simulation underestimated the maximum rain rate by 22% compared to that derived from TRMM-PR, and underestimated mean rainfall by 10.4% compared to the TRMM-TMI estimate, and by 12.5% compared to the sounding-based estimate. The warm microphysical processes modeled from both the WRF and the Goddard Cumulus Ensemble （GCE） models were quite close to those based on TMI, but the ice water contents in the models were relatively less compared to that derived from TMI. The CAPE and wind shear induced by the monsoon circulation were found to play critical roles in maintaining and developing the intense convective clouds over SCS. The latent heating rate increased more than twofold during the monsoon period and provided favorable conditions for the upward transportation of energy from the ocean, giving rise to the possibility of inducing large-scale interactions.
