Prevalence and risk factors of cardiovascular diseases and psychological distress among female scientists and technicians

This study aimed to explore the prevalence and risk factors of cardiovascular disease(CVD)and psychological distress among female scientists and technicians in ***,we included scientists and technicians from representative research institutions,medical institutions,colleges,universities,and businesses in china,and the data were collected from July 1,2019 to March 31,2021 via online *** parameters evaluated in this study included age,sex,marital status,educational background,monthly income,sleep hours,sleep problems,smoking,alcohol consumption,work-related stress,work burnout,cardiovascular symptoms,CVD,family history,and depressive and anxiety symptoms.A total of 14530 scientists and technicians were included,comprising 7144 men and 7386 *** found 34.9%men and 16.6%women with CVD,35.1%men and 21.4%women with depressive symptoms,28.7%men and 13.8%women with anxiety symptoms,and 22.0%men and9.5%women with CVD combined with depressive or anxiety *** study focused on the details of *** women(age≤35 years)had the highest prevalence of depressive symptoms(24.9%),anxiety symptoms(16.2%),and comorbidity(11.2%).It was established that,despite traditional risk factors,unmanageable work burnout,depressive symptoms,and anxiety symptoms were associated with a higher risk of CVD in women;insomnia,overwhelming work stress,unmanageable work burnout,and CVD were linked to a higher risk of depressive symptoms and anxiety;insomnia,overwhelming work stress,and unmanageable work burnout were related to CVD combined with depressive or anxiety symptoms.A bidirectional relationship was noted between CVD and depression or anxiety in female scientists and technicians,and insomnia and overwhelming work stress were positively associated with *** is suggested that effective measures should be taken to protect female scientists and technicians from CVD and psychological distress.

Epithelial to mesenchymal transition (EMT) plays a crucial role in cancer metastasis, accompanied with vast epigenetic ***-activated protein kinase (AMPK), a cellular energy sensor, plays regulatory roles in multiple biological processes. Although afew studies have shed light on AMPK regulating cancer metastasis, the inside epigenetic mechanisms remain unknown. Herein weshow that AMPK activation by metformin relieves the repressive H3K9me2-mediated silencing of epithelial genes (e.g., CDH1)during EMT processes and inhibits lung cancer metastasis. PHF2, a H3K9me2 demethylase, was identified to interact with AMPKα*** deletion of PHF2 aggravates lung cancer metastasis and abolishes the H3K9me2 downregulation and anti-metastasis effectof metformin. Mechanistically, AMPK phosphorylates PHF2 at s655 site, enhancing PHF2 demethylation activity and triggering thetranscription of CDH1. Furthermore, the PHF2-s655E mutant that mimics AMPK-mediated phosphorylation status further reducesH3K9me2 and suppresses lung cancer metastasis, while PHF2-s655A mutant presents opposite phenotype and reverses the antimetastasiseffect of metformin. PHF2-s655 phosphorylation strikingly reduces in lung cancer patients and the higherphosphorylation level predicts better survival. Altogether, we reveal the mechanism of AMPK inhibiting lung cancer metastasis viaPHF2 mediated H3K9me2 demethylation, thereby promoting the clinical application of metformin and highlighting PHF2 as thepotential epigenetic target in cancer metastasis.

Improving genetic diagnosis of Mendelian disease with RNA sequencing:a narrative review

Targeted sequencing and whole exome sequencing are the most common approaches used to detect causative variants in Mendelian diseases;however, using DNA-based sequencing techniques, the current molecular diagnostic yield is at best 50%. In recent years, RNA sequencing has been shown to be able to provide a genetic diagnosis in patients whose conditions were previously unable to be identified by DNA analysis. RNA sequencing can reveal expression outliers, aberrant splicing events, allele-specific expression, and new pathogenic variants, and as such can complement and expand on the traditional genomic methods used to diagnose Mendelian diseases. Therefore, RNA sequencing is expected to become a routine method for genetic diagnosis in the future. This article reviews the applications and challenges of RNA sequencing in the genetic diagnosis of Mendelian diseases.

Background This study aimed to explore the relationship of 25-hydroxyvitamin D[25(OH)D]in three trimesters and at birth with neurodevelopment at 24 months of *** From 2013 to 2016,pregnant women from the shanghai Birth Cohort in china were recruited for the ***,649 mother-infant pairs were *** 25(OH)D was measured with mass spectrometry in three trimesters,and cord blood was divided into deficiency(spectively),insufficiency(20–30 and 12–20 ng/mL,respectively),and sufficiency(≥30 and≥20 ng/mL,respectively).Bayley-III scale was used to assess cognitive,language,motor,social-emotional,and adaptive behavior development at 24 months of *** Bayley-III scores were grouped into quartiles,and scores within the lowest quartile were defined as suboptimal *** After adjusting for confounding factors,cord blood 25(OH)D in the sufficient group was positively correlated with cognitive[β=11.43,95%confidence interval(CI)=5.65–17.22],language(β=6.01,95%CI=1.67–10.3),and motor scores(β=6.43,95%CI=1.73–11.1);cord blood 25(OH)D in the insufficient group was also positively correlated with cognitive scores(β=9.42,95%CI=3.74–15.11).Additionally,sufficient vitamin D status in the four periods and persistent 25(OH)D≥30 ng/mL throughout pregnancy were associated with a lower risk of suboptimal cognitive development in adjusted models,although the effects were attenuated after applying the false discovery rate *** Cord blood 25(OH)D≥12 ng/mL has a significant positive association with cognitive,language,and motor development at 24 months of *** vitamin D status in pregnancy might be a protective factor for suboptimal neurocognition development at 24 months of age.

Correlation between symmetry Energy and Collective Flow in Heavy-Ion Collisions Induced by High Energy Radioactive Beams

Using an isospin- and momentum-dependent hadronic transport model, we investigate effects of the symmetry energy on several collective flows in heavy-ion collisions induced by radioactive beams at intermediate energies. It is found that the neutron-proton differential directed flow and the neutron-proton differential elliptic flow are strongly correlated with the symmetry energy, while the position averaged radial flow is weakly correlated with the symmetry energy.

stanene(sn)-based materials have been extensively applied in industrial production and daily life,but their potential biomedical application remains largely unexplored,which is due to the absence of the appropriate and effective methods for fabricating sn-based ***,we explored a new approach combining cryogenic exfoliation and liquid-phase exfoliation to successfully manufacture two-dimensional(2D)sn nanosheets(snNss).The obtained snNss exhibited a typical sheet-like structure with an average size of~100 nm and a thickness of~5.1 *** PEGylation,the resulting PEGylated snNss(snNss@PEG)exhibited good stability,superior biocompatibility,and excellent photothermal performance,which could serve as robust photothermal agents for multi-modal imaging(fluorescence/photoacoustic/photothermal imaging)-guided photothermal elimination of ***,we also used first-principles density functional theory calculations to investigate the photothermal mechanism of snNss,revealing that the free electrons in upper and lower layers of snNss contribute to the conversion of the photo to *** work not only introduces a new approach to fabricate 2D snNss but also establishes the snNss-based nanomedicines for photonic cancer *** new type of snNss with great potential in the field of nanomedicines may spur a wave of developing sn-based biological materials to benefit biomedical applications.

Cancer chemotherapy can be hindered by drug resistance which leads to lower drug ***,we have developed a drug delivery system that tethers doxorubicin to the surface of gold nanorods via a pHsensitive linkage(AuNRs@DOX),for a combined photothermal and chemical therapy for ***,AuNRs@DOX is ingested by HepG2 liver cancer *** endocytosis,the acidic pH triggers the release of doxorubicin,which leads to chemotherapeutic *** gold nanorods are not only carriers of DOX,but also photothermal conversion *** the presence of an 808 nm near-infrared laser,AuNRs@DOX significantly enhance the cytotoxicity of doxorubicin via the photothermal effect,which induces elevated apoptosis of hepG2 cancer cells,leading to better therapeutic effects in vitro and in vivo.

Objective: Venlafaxine is a common antidepressant and its therapeutic effect varies among people with different genetic backgrounds. The aim of this study was to investigate whether single nucleotide polymorphisms (sNPs) in thesLC17A7 gene are associated with the treatment outcome of venlafaxine in a Chinese Han population with major depressive ***: This prospective pharmacogenetic case-control study that involved genotyping of four sNPs ofsLC17A7 was conducted on 175 major depressive disorder patients of Chinese Han origin, aged 18 to 65 years, participated in the study from April 2005 to september 2006. Comparisons of allele and genotype frequencies of all sNPs were performed between the responder/remission group and the nonresponder/nonremission group. This study was approved by the Institutional Ethics Committee of sichuan University (approval No. 20151112-265).Results: The allele and genotype frequencies of the four candidate sNPs insCL17A7 showed no significant difference between responders and nonresponders. Meanwhile, no significant difference was detected in the four investigatedsLC17A7 sNPs between patients who did and did not exhibit remission. Although one of the investigatedsLC17A7 variants (rs1578944) demonstrated a significant association (P=0.022) with a response to venlafaxine after 6 weeks of treatment in the survival analysis, the association was unclear after a Bonferroni multiple comparisons test was ***: No significant association exists between the four candidate sNPs (rs1043558, rs1320301, rs1578944, and rs74174284) insLC17A7 and venlafaxine treatment in the Chinese Han population.

Platinum-based chemotherapy remains the main systemic treatment of ovarian cancer(OC).However,the inevitable development of platinum and poly(adenosine diphosphate-ribose)polymerase inhibitor(PARPi)resistance is associated with poor outcomes,which becomes a major obstacle in the management of this *** present study developed“all-in-one”nanoparticles that contained the PARPi olaparib and gallium(Ga)(III)(olaparib-Ga)to effectively reverse PARPi resistance in platinum-resistant A2780-cis and sKOV3-cis OC cells and in sKOV3-cis tumor ***,the olaparib-Ga suppressed sKOV3-cis tumor growth with negligible ***,the suppression effect was more evident when combining olaparib-Ga with cisplatin or carboplatin,as evaluated in A2780-cis and sKOV3-cis ***,the combined treatment induced DNA damage,which elicited the activation of ataxia telangiectasia mutated(ATM)/AMT-and Rad3-related(ATR)checkpoint kinase 1(Chk1)/Chk2 signal transduction *** led to the arrest of cell cycle progression at s and G2/M phases,which eventually resulted in apoptosis and cell death due to unrepairable DNA *** addition,effective therapeutic responses to olaparib-Ga and cisplatin combination or olaparib-Ga and carboplatin combination were observed in sKOV3-cis tumor-bearing animal ***,the present findings demonstrate that olaparib-Ga has therapeutic implications in platinum-resistant OC cells,and the combination of olaparib-Ga with cisplatin or carboplatin may be promising for treating patients with OC who exhibit resistance to both PARPi and platinum.
