Simultaneous identification and quantification of tetrodotoxin in fresh pufferfish and pufferfish-based products using immunoaffinity columns and liquid chromatography/quadrupole-linear ion trap mass spectrometry

In this study, we established a comprehensive method for simultaneous identification and quantification of tetrodotoxin （TTX） in fresh pufferfish tissues and pufferfish-based products using liquid chromatography/quadrupole-linear ion trap mass spectrometry （LC-QqLIT-MS）. TTX was extracted by 1% acetic acid-methanol, and most of the lipids were then removed by freezing lipid precipitation, followed by purification and concentration using immunoaffinity columns （IACs）. Matrix effects were substantially reduced due to the high specificity of the IACs, and thus, background interference was avoided. Quantitation analysis was therefore performed using an external calibration curve with standards prepared in mobile phase. The method was evaluated by fortifying samples at 1, 10, and 100 ng/g, respectively, and the recoveries ranged from 75.8%--107%, with a relative standard deviation of less than 15%. The TTX calibration curves were linear over the range of 1-1 000 ~tg/L, with a detection limit of 0.3 ng/g and a quantification limit of 1 ng/g. Using this method, samples can be further analyzed using an information- dependent acquisition （IDA） experiment, in the positive mode, from a single liquid chromatography-tandem mass spectrometry injection, which can provide an extra level of confirmation by matching the full product ion spectra acquired for a standard sample with those from an enhanced product ion （EPI） library. The scheduled multiple reaction monitoring method enabled TTX to be screened for, and TTX was positively identified using the IDA and EPI spectra. This method was successfully applied to analyze a total of 206 samples of fresh pufferfish tissues and pufferfish-based products. The results from this study show that the proposed method can be used to quantify and identify TTX in a single run with excellent sensitivity and reproducibility, and is suitable for the analysis of complex matrix pufferfish samples.

Surface-enhanced Raman scattering(SERS)spectroscopy has been employed as a rapid analysis technology for food security inspection ***,it is still a great challenge to rapidly quantify multiple trace antibiotics potentially abused in aquaculture *** this work,a magnetic Ti_(3)C_(2)T_(x)/Fe_(3)O_(4)/Ag substrate was prepared for the development of a reliable rapid SERS quantification method for multiple trace sulfonamides in aquatic *** magnetic substrate had good uniformity,reproducibility,stability and SERS ***,this substrate could integrate the magnetic separation-enrichment and matrix clean-up without cross contamination,which endowed it with good selectivity and antiinterference capability during real sample *** electromagnetic enhancement and chemical enhancement mechanism of this magnetic substrate were studied in detail to reveal its good separationenrichment performance and SERS ***,a rapid SERS quantification method was established and practically applied for trace phthalic sulfathiazole(PST)and silver sulfadiazine(SSD)in aquatic products by using Ti_(3)C_(2)T_(x)/Fe_(3)O_(4)/Ag magnetic *** PST and SSD could be actually detected and quantified as 55.9μg/kg and 64.0μg/kg in aquatic products,*** recoveries of 83.9%-116%with relative standard deviations(RSDs)of 0.5%-3.2%for PST and 80.2%-102%with RSDs of 1.3%-5.8%for SSD were *** work proposed an efficient and reliable method for rapid quantification of trace multiple sulfonamides in complex aquatic samples during food security inspection.

Carbon dots/graphite carbon nitride(CDs/g-C_(3)N_(4)),a novel composite photocatalyst,has shown great po-tential for applications in energy regeneration and environmental remediation owing to its following advantages:metal-free,low cost,easily tunable,and excellent photocatalytic ***,we reviewed the development of synthetic strategies,photocatalytic enhancement mechanisms,and pho-tocatalytic applications of CDs/g-C_(3)N_(4) in this ***,the three composite strategies of CDs and g-C_(3)N_(4)-self-assembly,solvothermal,and calcination polymerization are outlined,and their advantages and disadvantages are described in ***,the photocatalytic enhancement mechanism of the com-posite strategies was elucidated according to the variation trends of CDs/g-C_(3)N_(4) band structure,electronic properties,light absorption range,and interfacial charge ***,the applications of CDs/g-C_(3)N_(4) in hydrogen evolution,pollutant degradation,CO_(2) reduction,and bacterial disinfection in recent years are reviewed ***,the current obstacles and future research directions of CDs/g-C_(3)N_(4) are discussed from the perspective of preparation technology and practical applications,respectively.

Phylogenetic and Comparative Genomic Analysis of Lactobacillus fermentum Strains and the Key Genes Related to their Intestinal Anti-Inflammatory Effects

Emerging evidence shows that some Lactobacillus fermentum(***)strains can contribute to the prevention and treatment of ulcerative colitis(UC).In this study,105 isolates of *** strains were separated from fecal samples of populations in different regions in china and their draft genomes were ***-genomic and phylogenetic characterizations of these strains and four model strains(*** 3872,CECT5716,IF03956,and VRI003)were *** analysis ind icated that there was no significant adaptive evolution between the genomes of *** strains and the geographical location,sex,ethnicity,and age of the *** *** strains(FWXBH115,FGDLZR121,and FXJCJ61)from different branches of the phylogenetic tree and strain type *** CECT5716 were selected and their anti-inflammatory and immune modulatory activities in a dextran sulphate sodium(DSS)-induced colitis mouse model were further *** *** FXJCJ61 and CECT5716 significantly alleviated UC by reducing all colitis-associated histological indices,maintaining mucosal integrity,and stimulating replenishment of short-chain fatty acids(SCFAs),while the other two strains failed to offer similar *** anti-inflammato ry mechanisms of *** FXJCJ61 and CECT5716 were related to the inhibition of nuclear factor kappa-B(NF-κB)signaling pathway activation and enhancement of interleukin 10(IL-10)*** genomic analysis of these strains identified candidate genes that may contribute to the anti-inflammatory effects of specific *** strains.

Irritable bowel syndrome with diarrhea(IBS-D)is chronic intestinal dysfunction with diarrhea and othercomplicated clinical symptoms,and it has a great impact on the daily life and mental state of *** studies have reported that ingestion of probiotics can significantly alleviate a variety of *** pupose of this study was to investigate the IBS-D-alleviating effects of a probiotic strain,lactobacillus plantarum(***)CCFM8610,with multiple health-promoting *** studywas a 12-week,randomized,double-blind,placebo-controlled,pilot clinical ***-five patientswere randomly assigned to receive the placebo,oligosaccharides,or *** CCFM8610(1×10^(10) colony-forming units(CFU)per day),with a 2-week run-in period,an 8-week intervention period,anda 2-week follow-up observation *** patients'clinical symptoms and quality of life were exam-ined by the IBS symptom severity scale(IBS-SsS)and the IlBS quality of life scale(IBS-QOL).Changes ingut microbiota composition and diversity were measured at the end of the intervention *** oraladministration of *** CCFM8610 significantly decreased the IBS-SSS and IBS-QOL scores,reduced IBS-D symptom severity,recovered gut microbiota diversity,decreased the relative abundanceof bloating-related genus Methanobrevibacter,and increased the relative abundance of butyric acid-producing genera,including Anaerostipes,Anaerotruncus,Bifidobacterium,Butyricimonas,*** findings suggest that ingestion of *** CCFM8610 can significantly alleviate clinical symptoms and gut microbiota dysbiosis in IBS-D *** IBS-D-alleviating effect of ***-tarum CCFM8610 may be related to theincrease in the relative abundance of butyric acid-producing genera in the intestine.

DEMC: A Deep Dual-Encoder Network for Denoising Monte Carlo Rendering

In this paper, we present DEMC, a deep dual-encoder network to remove Monte Carlo noise efficiently while preserving details. Denoising Monte Carlo rendering is different from natural image denoising since inexpensive by-products (feature buffers) can be extracted in the rendering stage. Most of them are noise-free and can provide sufficient details for image reconstruction. However, these feature buffers also contain redundant information. Hence, the main challenge of this topic is how to extract useful information and reconstruct clean images. To address this problem, we propose a novel network structure, dual-encoder network with a feature fusion sub-network, to fuse feature buffers firstly, then encode the fused feature buffers and a noisy image simultaneously, and finally reconstruct a clean image by a decoder network. Compared with the state-of-the-art methods, our model is more robust on a wide range of scenes, and is able to generate satisfactory results in a significantly faster way.

Hypericin is one of the most important phenanthoperylene quinones extracted mainly from plants of the genus Hypericum belonging to the sections Euhypericum and Campylosporus of Keller's classification. Widespread attention to the antiviral and anti-tumor properties of hypericin has spurred investigations of the chemical synthesis and biosynthesis of this unique compound. However, the synthetic strategies are challenging for organic and biological chemists. In this review, specific significant advances in total synthesis, semi-synthesis, and biosynthesis in the past decades are summarized.

Gut microbiota controls the development of chronic pancreatitis:A critical role of short-chain fatty acids-producing Gram-positive bacteria

Chronic pancreatitis(CP)is a progressive and irreversible fibroinflammatory disorder,accompanied by pancreatic exocrine insufficiency and dysregulated gut ***,accumulating evidence has supported a correlation between gut dysbiosis and CP ***,whether gut microbiota dysbiosis contributes to CP pathogenesis remains ***,an experimental CP was induced by repeated high-dose caerulein *** broad-spectrum antibiotics(ABX)and ABX targeting Gram-positive(G+)or Gram-negative bacteria(G-)were applied to explore the specific roles of these *** dysbiosis was observed in both mice and in CP patients,which was accompanied by a sharply reduced abundance for short-chain fatty acids(SCFAs)-producers,especially G+***-spectrum ABX exacerbated the severity of CP,as evidenced by aggravated pancreatic fibrosis and gut dysbiosis,especially the depletion of SCFAs-producing G+***,depletion of SCFAs-producing G+bacteria rather than G-bacteria intensified CP progression independent of TLR4,which was attenuated by supplementation with exogenous ***,SCFAs modulated pancreatic fibrosis through inhibition of macrophage infiltration and M2 phenotype *** study supports a critical role for SCFAs-producing G+bacteria in ***,modulation of dietary-derived SCFAs or G+SCFAs-producing bacteria may be considered a novel interventive approach for the management of CP.
