Preferential infection and depletion of gut-homing a4β7 CD4＋ T cells in the blood are observed in chronic hiv/SIV infection. The dynamic change in gut-homing a4p7 CD4＋ T cells and their functional subsets during the acute stages of hiv-1 infection are less documented. Therefore, we conducted a cohort study to investigate whether acute hiv-1 infection induced abnormalities in gut-homing a4β7 CD4＋ T cells and their functional subsets. We examined the frequency, absolute number, and functionality of gut-homing a4β7 CD4＋ T cells in 26 acute hiv-l-infected patients compared with 20 healthy individuals. We found that circulating gut-homing a4β7 CD4＋ T cells were preferentially depleted during acute hiv-1 infection and were positively correlated with absolute CD4＋ T-cell count in blood. Notably, Th17 and Thl cell subsets of gut-homing CD4＋ T cells were also decreased, which resulted in an imbalance of T helper cells （Th 1）-regulatory T cells （Treg） and *** 17 ratios. Gut-homing Th17 and Thl cells were also positively correlated with the absolute number of total CD4＋ T cells and gut-homing CD4＋ T cells. The gut-homing Treg：Th17 ratio was inversely correlated with the CD4＋ T-cell count. Taken together, the analyses of our acute hiv-1 cohort demonstrate that gut-homing a4β7 CD4＋ T cells and their functional subsets were profoundly depleted during acute hiv-1 infection, which may have resulted in the persistent loss of circulating CD4＋ T cells and an imbalance of Thl-Treg and ***17 ratios and contribute to hiv-1 disease pathogenesis.

HXB2 is primarily used as a template strain in developing hiv vaccines in Europe and the US. However,it is not yet known whether the strain can induce strong hiv-specific CD8+ T cell responses in Chinese hiv/aids patients. In the present study,two groups of subjects were investigated:9 aids patients and 7 long-term nonprogressors (LTNPs). hiv-specific CD8+ T cell responses were examined in all patients through the ELISPOT assay. CD4+ T cell counts,CD8+ T cell counts,viral load and hiv subtype of each patient were also measured. Thailand B virus strain was identified among all the patients. The breadth and magnitude of hiv-specific CD8+ T cell responses in the LTNPs group are greater than those in the aids group (Phiv-specific CD8+ T cell responses and CD4+ T cells,and a negative correlation between hiv-specific CD8+ T cell responses and mean viral load. In summary,the hiv-specific CD8+ T cell responses to the HXB2 Gag and Nef peptide pools are considerable in Chinese hiv/aids patients infected with Thailand B virus strain. hiv-1 vaccines based on HXB2 strain that can induce extensive immunity may be helpful for Chinese.

Here, we report a novel and universal methodology,termed ＂ntarctic thermolabile uracil-DNA-glycosylase （AUDG）-supplemented nucleic acid amplification techniques （NAAs） using a labeled-based nanoparticle lateral flow biosensor （LFB）＂（AUDG-NAAs-LFB）, which merges enzymatic （AUDG） digestion of contaminant amplicons with different nucleic acid amplification techniques （NAAs）, and uses a lateral flow biosensor （LFB） for the rapid and visual confirmation of the presence of a target nucleic acid sequence. AUDG-NNAs-LFB is a one-pot, closedvessel assay, that can effectively eliminate false-positive signals arising from either carryover contaminants or the interaction between labeled primers. A new LFB was devised for detecting three targets （two amplicons generated from amplification of target sequences, and a chromatography control）, without the need for probe- hybridization or additional incubation steps. As a proof of concept, multiple cross displacement amplification （MCDA）, which is a specific, sensitive, and rapid isothermal amplification method, was selected as the model amplification technique to demonstrate the feasibility of AUDG-NAAs-LFB. As a result, we demonstrate the applicability of the AUDG-MCDA-LFB method for simultaneously detecting high-risk human papillomaviruses genotypes 16 and 18, which are the most and second-most prevalent strains of the virus reported in women worldwide. We also confirm the principle behind the AUDG-MCDA- LFB assay and validate its sensitivity, reproducibility, and specificity using serial dilutions of the type-specific plasmids, as well as clinical samples. This proof- of-concept method （AUDG-MCDA-LFB） can be easily reconfigured to detect various nudeic acid sequences by redesigning the specific MCDA primers.

Background: It is controversial whether the apolipoprotein E epsilon 4 allele (APOE ε4) is a risk gene for human immunodeficiency virus (hiv)-related neurocognitive impairment. This meta-analysis aimed to summarize evidence of the associations betweenAPOE ε4 and cognitive impairment in people living with hiv (PLWH).Methods: Our study conducted a systematic literature search of PubMed, Web of Science, Embase, Google Scholar, and ProQuest for studies published before April 11, 2022 that evaluated associations betweenAPOE ε4 and cognitive impairment in adult PLWH (aged ≥18 years). We calculated pooled odds ratios (ORs) of global cognitive impairment and 95% confidence intervals (CIs) and standardized mean differences (SMDs) for specific cognitive domains betweenAPOE ε4 carriers and non-carriers. Subgroup meta-analyses were used to evaluate the result profiles across different categorical ***: Twenty studies met the inclusion criteria, including 19 that evaluated global cognitive *** ε4 was significantly associated with global cognitive impairment in PLWH (OR = 1.36, 95% CI = [1.05, 1.78], number of estimates [k] = 19,P = 0.02, random effects).
