Photo-manipulated polyunsaturated fatty acid-doped liposomal hydrogel for flexible photoimmunotherapy

Despite the synergy of immune checkpoint blockade(ICB) therapy and photodynamic therapy(PDT) holds great promise as countermeasures against breast cancer, exploring long-term or flexible short-time therapeutic strategies in “cold” tumors remains a great challenge. Here, we present a polyunsaturated fatty acid-doped liposomal hydrogel Lp(DHA)@CP Gel loaded with photosensitizer chlorin e6(Ce6) and programmed death-ligand 1 antibody(αPD-L1) for flexible local photoimmunotherapy with merely singledosed administration. The presence of polyunsaturated fatty acid(docosahexaenoic acid, DHA) doped in particle membrane endows liposomes with flexibly reactive oxygen species(ROS)-responsive release capability, which was attributed to the presence of abundant unsaturated groups. The αPD-L1 was repeatedly induced to in situ release in response to the PDT under photo-exposure. The immunogenic cell death(ICD) effect of PDT evoked “cold” breast tumor to “hot” one, and then assisted the cascade releasedαPD-L1 to synergistically boost the immunotherapy. After a single dose of peritumoral administration of Lp(DHA)@CP Gel, the on-demand treatment can maximize patient compliance and safety by adjusting therapeutic behaviors via a photo on-off switch. This work presents a flexible medication platform,showing promise in improving the objective response rate of ICB therapy and minimizing its systemic toxicity.

Development of a droplet digital polymerase chain reaction assay for the sensitive detection of total and integrated HIV-1 DNA

Background:Total human immunodeficiency virus(HIV)DNA and integrated HIV DNA are widely used markers of HIV *** digital polymerase chain reaction(ddPCR)can be used for absolute quantification without needing a standard ***,we developed duplex ddPCR assays to detect and quantify total HIV DNA and integrated HIV ***:The limit of detection,dynamic ranges,sensitivity,and reproducibility were evaluated by plasmid constructs containing both the HIV long terminal repeat(LTR)and human CD3 gene(for total HIV DNA)and ACH-2 cells(for integrated HIV DNA).Forty-two cases on stable suppressive antiretroviral therapy(ART)were assayed in total HIV DNA and integrated HIV *** coefficient analysis was performed on the data related to DNA copies and cluster of differentiation 4 positive(CD4^(+))T-cell counts,CD8^(+)T-cell counts and CD4/CD8 T-cell ratio,*** assay linear dynamic range and lower limit of detection(LLOD)were also ***:The assay could detect the presence of HIV-1 copies 100%at concentrations of 6.3 copies/reaction,and the estimated LLOD of the ddPCR assay was 4.4 HIV DNA copies/reaction(95%confidence intervals[CI]:3.6-6.5 copies/reaction)with linearity over a 5-log_(10)-unit range in total HIV DNA *** the integrated HIV DNA assay,the LLOD was 8.0 copies/reaction(95%CI:5.8-16.6 copies/reaction)with linearity over a 3-log 10-unit *** HIV DNA in CD4^(+)T cells was positively associated with integrated HIV DNA(r=0.76,P<0.0001).Meanwhile,both total HIV DNA and integrated HIV DNA in CD4^(+)T cells were inversely correlated with the ratio of CD4/CD8 but positively correlated with the CD8^(+)T-cell ***:This ddPCR assay can quantify total HIV DNA and integrated HIV DNA efficiently with robustness and *** can be readily adapted for measuring HIV DNA with non-B clades,and it could be beneficial for testing in clinical trials.

Undifferentiated embryonal sarcoma of the liver (UESL) very rarely presents *** 1% to 4% of all solid tumors in children are malignant liver tumors,and UESL accounts for 9% to 15% of the liver *** onset of UESL usually occurs at 6 to 10 years of age and there is no obvious difference based on sex.[1] The first symptoms of UESL are abdominal distention and abdominal pain without jaundice,while a physical examination commonly reveals no positive vital signs except the palpable liver tumor.[1] The serum alphafetoprotein (AFP) test is not sensitive for UESL.[2] Preoperative diagnosis of UESL is very difficult.

Comparison of Kernel Entropy Component Analysis with Several Dimensionality Reduction Methods

Dimensionality reduction techniques play an important role in data mining. Kernel entropy component analysis( KECA) is a newly developed method for data transformation and dimensionality reduction. This paper conducted a comparative study of KECA with other five dimensionality reduction methods,principal component analysis( PCA),kernel PCA( KPCA),locally linear embedding( LLE),laplacian eigenmaps( LAE) and diffusion maps( DM). Three quality assessment criteria, local continuity meta-criterion( LCMC),trustworthiness and continuity measure(T&C),and mean relative rank error( MRRE) are applied as direct performance indexes to assess those dimensionality reduction methods. Moreover,the clustering accuracy is used as an indirect performance index to evaluate the quality of the representative data gotten by those methods. The comparisons are performed on six datasets and the results are analyzed by Friedman test with the corresponding post-hoc tests. The results indicate that KECA shows an excellent performance in both quality assessment criteria and clustering accuracy assessing.

Economic Dispatch of Electrical Power System Based on the Multi-objective Co-evolutionary Algorithm

It is important to distribute the load efficiently to minimize the cost of the economic dispatch of electrical power system. The uncertainty and volatility of wind energy make the economic dispatch much more complex when the general power systems are combined with wind farms. The short term wind power prediction method was discussed in this paper. The method was based on the empirical mode decomposition (EMD) and ensemble empirical mode decomposition (EEMD). Furthermore,the effect of wind farms on the traditional economic dispatch of electrical power system was analyzed. The mathematical model of the economic dispatch was established considering the environmental factors and extra spinning reserve cost. The multi-objective co-evolutionary algorithm was used to figure out the model. And the results were compared with the NSGA-Ⅱ(non-dominated sorting genetic algorithm-Ⅱ) to verify its feasibility.

Association of apolipoprotein E epsilon 4 and cognitive impairment in adults living with human immunodeficiency virus: a meta-analysis

Background: It is controversial whether the apolipoprotein E epsilon 4 allele (APOE ε4) is a risk gene for human immunodeficiency virus (HIV)-related neurocognitive impairment. This meta-analysis aimed to summarize evidence of the associations betweenAPOE ε4 and cognitive impairment in people living with HIV (PLWH).Methods: Our study conducted a systematic literature search of PubMed, Web of Science, Embase, Google Scholar, and ProQuest for studies published before April 11, 2022 that evaluated associations betweenAPOE ε4 and cognitive impairment in adult PLWH (aged ≥18 years). We calculated pooled odds ratios (ORs) of global cognitive impairment and 95% confidence intervals (CIs) and standardized mean differences (SMDs) for specific cognitive domains betweenAPOE ε4 carriers and non-carriers. Subgroup meta-analyses were used to evaluate the result profiles across different categorical ***: Twenty studies met the inclusion criteria, including 19 that evaluated global cognitive *** ε4 was significantly associated with global cognitive impairment in PLWH (OR = 1.36, 95% CI = [1.05, 1.78], number of estimates [k] = 19,P = 0.02, random effects).
