The phosphoinositide 3-kinase-AkT-mammalian target of rapamycin (PI3k-AkT-mTOR) pathway is a frequently hyperactivated pathway in cancer and is important for tumor cell growth and survival. The development of targeted therapies against mTOR, a vital substrate along this pathway, led to the approval of allosteric inhibitors, including everolimus and temsirolimus, for the treatment of breast, renal, and pancreatic cancers. However, the suboptimal duration of response in unselected patients remains an unresolved issue. Numerous novel therapies against critical nodes of this pathway are therefore being actively investigated in the clinic in multiple tumour types. In this review, we focus on the progress of these agents in clinical development along with their biological rationale, the need of predictive biomarkers and various combination strategies, which will be useful in counteracting the mechanisms of resistance to this class of drugs.

The use of either lossless snubbers or resonant zero voltage switching (ZVS) and zero current switching (ZCS) techniques can increase efficiency and reduce electromagnetic inter ference (EMI) and noise of industrial power equipment at high switching frequencies. This paper presents an adaptive composite soft switching configuration which combines snubber functions and resonant ZCS circuits for switches in inverters using power bridge leg modules. Simulation and experimental results are included.

In this paper, precise tracking control of a class of high-order hydraulic servo systems is studied, where the dynamics of the electrohydraulic actuator are considered. Different to available results, a new control method is proposed without using the backstepping scheme and any function approximators. The model of the hydraulic servo system is transformed into a Brunovsky form, so that the tracking control of the original system can be carried out by controlling the transformed system. In this case, the widely-used backstepping scheme is avoided. The Levant’s differentiator is then employed to observe the u.known system states of the transformed system. Finally, an u.known dynamics estimator is constructed and then incorporated into the feedback control, which avoids using function approximators（e.g. neural network, fuzzy systems） for addressing u.known nonlinearities. Only the output of the original system needs to be measured. The closed-loop control system stability is rigorously proved. Simulation results based on a realistic plant are provided to illustrate the effectiveness of the proposed control method.

This paper proposes an optimal tracking control of completely u.known affine nonlinear systems using a system augmentation scheme and a modified discounted performance function, where the feedforward and feedback control actions can be obtained simultaneously. We first present an adaptive identifier to estimate the u.known dynamics, where the estimated parameters are updated based on a new adaptive law such that these estimated parameters converge to a small neighborhood around their true values. An augmented system consisting of the tracking error and the generator of the trajectory to be tracked are then constructed, which derives an augmented tracking Hamilton-Jacobi-Bellman（HJB） equation. An online critic neural network（NN） is finally presented to approximate the optimal value function of HJB equation, and then calculate the optimal control action. This leads to an identifier-critic based adaptive dynamic programming（ADP） structure. Simulation results are presented to demonstrate the effectiveness of the proposed method.

The paper considers three common snubber circuits used on gate turn-off thyristor and/or insulated gate bipolar transistor inverters. The three snubbers are passive lossless circuits for power bridge legs, and the improvements and modifications to these snubber circuits are presented. The comparative features and operation of the three improved energy recovery snubbers are discussed and supported by PSPICE simulations and experimental results.

In this article, we provide a review of the consensus control problem for stochastic multi-agent systems(MASs). Recent advances are surveyed according to the method of occurrence of the stochasticity of the MASs. First, the consensus problem is discussed for MASs, wherein individual agents are corrupted by random noises, i.e., the dynamics of agents involve stochasticity in process and/or measurement equations. Both additive noises and multiplicative noises are surveyed in detail and special attention is paid to the MASs whose dynamics are governed by It?o differential equations. Moreover, particular effort is devoted to presenting the latest progress on the consensus problem for a special type of stochastic MAS with Markovian jump parameters. Subsequently,the relevant research is summarized for MASs with noisy communication environments and stochastic ***, we provide a systematic review of the consensus problems for MASs whose communication topology varies randomly in the process of data propagation among agents. Finally, conclusions are drawn and several potential future research directions are outlined.

Gene regulation is inherently a stochastic process due to intrinsic and extrinsic noises which cause the fluctuations and uncertainties of kinetic *** the other hand,time delays are usually inevitable due to different biochemical reactions in the genetic regulatory networks（GRNs）which are also affected by ***,in this paper,we propose a GRN model that is subject to additive and multiplicative noises as well as time-varying *** time-varying delay is assumed to belong to an interval and no restriction on the derivative of the time-varying delay is needed,which allows the delay to be a fast time-varying *** stochastic stability of such GRNs with disturbance attenuation is analyzed by applying the control theory and mathematical *** on the Lyapunov method,new stability conditions are derived in the form of linear matrix inequalities（LMIs）that are dependent on the upper and lower bounds of time *** example is employed to illustrate the applicability and usefulness of the developed theoretical results.

The paper proposes a novel adaptive parameter estimation（APE） framework for nonlinearly parameterized nonlinear systems based on the parameter estimation *** basic idea is to reformulate nonlinearly parameterized systems by using Taylor series expansion,and then tailor a recently proposed APE *** expression of the parameter estimation error is derived by introducing auxiliary filtered variables,which are then used to design the adaptive laws to achieve exponential and even finite-time convergence under the standard persistent excitation（PE） ***,the robustness of the proposed APE against bounded disturbances is also *** simulation examples demonstrate the effectiveness of the proposed estimation algorithms.

This paper is concerned with the finite-horizon H state estimation problem for time-varying complex networks with redundant *** redundant channels are employed in the data transmission to against the fragility of communication networks commonly configured by the single channel subject to data missing and uniform quantization *** aim of the addressed problem is to design an estimator such that the H disturbance attenuation level is guaranteed for the estimation error dynamics over a given finite *** employing the stochastic analysis approach and completing squares method,the desired estimator gains are characterized by solving two coupled backward recursive Riccati Difference Equations(RDEs).Finally,a numerical example is given to illustrate the effectiveness of the results.
