ZNF554 Inhibits Endometrial Cancer Progression via Regulating RBM5 and Inactivating WNT/β-Catenin Signaling Pathway

Lipid is a key component of plasma membrane,which plays an important role in the regulation of various cell biological behaviors,including cell proliferation,growth,differentiation and intracellular signal *** have shown that abnormal lipid metabolism is involved in many malignant processes,including colorectal cancer(CRC).Lipid metabolism in CRC cells can be regulated not only by intracellular signals,but also by various components in the tumor microenvironment,including various cells,cytokines,DNA,RNA,and nutrients including *** contrast,abnormal lipid metabolism provides energy and nutrition support for abnormal malignant growth and distal metastasis of CRC *** this review,we highlight the remodeling roles of lipid metabolism crosstalk between the CRC cells and the components of tumor microenvironment.

Exome sequencing analysis identifies novel homozygous mutation in ABCA4 in a Chinese family with Stargardt disease

AIM: To identify the disease-associated mutations in a Chinese Stargardt disease(STGD) family, extend the existing spectrum of disease-causing mutations and further define the genotype-phenotype ***: A Chinese STGD family and 200 normal controls were collected. Whole exome sequencing(WES) and bioinformatics analysis were performed to find the pathogenic gene mutation. Physico-chemical parameters of mutant and wildtype proteins were computed by Prot Param tool. Domains analysis was performed by SMART online software. HOPE online software was used to analyze the structural effects of mutation. Immunofluorescence, quantitative real-time polymerase chain reaction and Western blotting were used for expression ***: Using WES, a novel homozygous mutation(NM_000350: c.G3190 C, p.G1064 R) in ABCA4 gene was identified. This mutation showed co-segregation with phenotype in this family. It was not found in the 200 unrelated health controls and absent from any databases. It was considered "Deleterious" as predicted by five function prediction softwares, and was highly conserved during evolution. ABCA4 was expressed highly in the human eye and mouse retina. The p.G1064 R was located in AAA domain, may force the local backbone into an incorrect conformation, disturb the local structure, and reduce the activity of ATPase resulting in the disease pathology. CONCLUSION: We define a novel pathogenic mutation(c.G3190 C of ABCA4) of STGD. This extends the existing spectrum of disease-causing mutations and further defines the genotype-phenotype correlations.

Characterization of a Triticum aestivum-Aegilops germplasm line presenting reduced plant height and early maturation

Plant height plays an important role in the potential and stability of crop yields and represents one of the most important agronomic traits of wheat. Although more than 30 dwarfing genes have been identified in wheat, only a few are used in wheat breeding, which has narrowed the genetic basis of newly developed varieties. Therefore, continually identifying new dwarfing genes is required to produce improved wheat cultivars. TA001 is a new germplasm line marked by reduced plant height and early maturation, and it was derived from a hybridization between the common wheat Yannong 15 and the Aegilops ventricosa × Aegilops cylindrica amphiploid SDAU18. In this study, cytological observations, agronomic trait examinations, genomic in situ hybridization(GISH), multicolor genomic in situ hybridization(mc-GISH), multicolor fluorescence in situ hybridization(mc-FISH), SSR analysis and seed storage protein electrophoresis were combined to determine the cytological stability, main agronomic traits, chromosomal constituents and seed storage protein subunits of TA001. Twenty-one bivalents were observed in most of the pollen mother cells at metaphase I(PMCs MI) in TA001, which housed 42 chromosomes, and the chromosomes in most pollen mother cells at anaphase I(PMCs AI)displayed 21/21 segregation. Twenty bivalents plus two univalents were simultaneously observed in most of the PMCs MI of the hybrid F_1 between TA001 and Yannong 15. TA001 possessed all chromosomes from genomes A, B and D except for chromosome 7B, which was replaced by one pair of N-genome chromosomes from Aegilops ventricosa. Several pairs of chromosomes in TA001 exhibited different FISH patterns from the equivalent chromosomes in Yannong 15. TA001 housed alien genetic materials from Aegilops ventricosa and Aegilops cylindrica and possessed new glutenin and gliadin subunits specific to SDAU18, as revealed by molecular marker analysis and protein electrophoresis respectively.

目的观察依帕司他(EPS)对血小板衍生生长因子(PDGF)诱导的大鼠肺动脉平滑肌细胞(PASMC)增殖的影响,并探讨与醛糖还原酶(AR)表达的关系。方法组织块贴壁法制备PASMC,PDGF30 mg·L^(-1)诱导细胞增殖,用Lipofectamin 3000将AR小干扰RNA(ARsi RNA)干扰片段转入PASMC细胞,24 h后,检测AR的mRNA和蛋白水平。另取细胞分为6组,分别为正常对照组,PDGF 30 mg·L^(-1)组,PDGF+EPS 1,10和100μmol·L^(-1)组,EPS 100μmol·L^(-1)组,定量PCR法和Western蛋白印迹法检测增殖细胞核抗原(PCNA)、胶原蛋白Ⅰ和AR的mRNA和蛋白表达水平。Brd U法和流式细胞术检测细胞增殖,检测PCNA、胶原蛋白Ⅰ和AR的表达。结果与正常对照相比,PDGF 30μg·L^(-1)处理48 h后,PASMC增殖显著增加(P<0.01),PCNA和胶原蛋白Ⅰ的表达明显升高(P<0.01);与PDGF组相比,ARsi RNA干扰组细胞增殖显著减少(P<0.01),PCNA和胶原蛋白Ⅰ的表达明显降低(P<0.01);与PDGF组相比,EPS 1,10和100μmol·L^(-1)组增殖显著减少(P<0.05,P<0.01),PCNA和胶原蛋白Ⅰ的表达也明显降低(P<0.05,P<0.01)。提示PDGF诱导细胞增殖与其上调AR的表达有关,提示EPS抑制细胞增殖与其下调AR的表达有关。结论 EPS对PDGF诱导的PASMC增殖具有一定的抑制作用,其机制可能与其抑制PDGF诱导AR的表达有关。

The present study was designed to evaluate the possible in vivo protective effects of sesamin on hypertension,arterial dysfunction and remodeling in spontaneously hypertensive rats (SHR).Sesamin was orally administered for 16 weeks in *** blood pressure(SBP) was measured by tail cuff every 2 *** 16 weeks,vasorelaxation response to acetylcholine and nitroprusside,and functional assessment of endothelial nitric oxide synthase(eNOS) activity were determined in aortic *** HultrastructureH of endothelium and vascular smooth muscle cells (VSMCs) of aortic rings were examined by electron *** medial thickness(MT), luminal diameter(LD),the ratio of MT to LD(MT/LD) and the volume fraction of collagen(VFC) of aortas were observed by Masson’s *** protein expression of transforming growth factorβ1(TGF-β1),eNOS,nitrotyrosine,nicotinamide adenine dinucleotide phosphate(NADPH) oxidase subunit p22phox and p47phox in aortas were detected by Western *** with sesamin in SHR reduced SBP,improved aortic remodeling and vasodilatation induced by acetylcholine,increased LD,reduced VFC,MT,MT/LD,and the ultrastructural damage of endothelium and VSMCs in *** changes were related to increased eNOS activity/expression,and reduced TGF-β1,nitrotyrosine,p22phox,p47phox protein expression. The results demonstrated that long-term treatment with sesamin reduces hypertension,and improves arterial dysfunction and remodeling in SHR through upregulation of eNOS expression/activity and downregulation of TGF-β1 expression associated with reduced p22phox and p47phox expression.
