Knock-out of BnHva22c reduces the susceptibility of Brassica napus to infection with the fungal pathogen Verticillium longisporum

Verticillium longisporum(Vl43)is a soilborne hemibiotrophic fungal pathogen causing stem striping on oilseed rape(OSR)and severe yield losses.Breeding for resistant varieties is the most promising approach to control this disease.Here,we report the identification of Hva22c as a novel susceptibility factor and its potential for improving OSR resistance.Hva22c is a member of the Hva22 gene family,originally described for barley(Hordeum vulgare).Several Hva22 members have been located at the endoplasmic reticulum.Hva22c is up-regulated in response to Vl43 in both Arabidopsis and OSR.We demonstrate that knock-out of Hva22c in OSR by CRISPR/Cas9 and its homolog in Arabidopsis by T-DNA insertion reduced plants’susceptibility to Vl43 infection and impaired the development of disease symptoms.To understand the underlying mechanism,we analysed transcriptomic data from infected and non-infected roots of hva22c knock-out and wild type plants.We identified a homozygous mutant with frame-shifts in all four BnHva22c loci displaying a vastly altered transcriptional landscape at 6 dpi.Significantly,a large set of genes was suppressed under mock conditions including genes related to the endomembrane systems.Among the up-regulated genes we found several defense-related and phytohormone-responsive genes when comparing mutant to the wild type.These results demonstrate that Hva22c is functionally required for a fully compatible plant-fungus interaction.Its loss of function reduces plant susceptibility,most likely due to endoplasmatic reticulum and Golgi dysfunction accompanied by additionally activated defense responses.These findings can help improve OSR resistance to V.longisporum infection.

Efficient scar-free knock-ins of several kilobases in plants by engineered CRISPR-Cas endonucleases

In plants and mammals,non-homologous end-joining is the dominant pathway to repair DNA doublestrand breaks,making it challenging to generate knock-in events.In this study,we identified two groups of exonucleases from the herpes virus and the bacteriophage T7 families that conferred an up to 38-fold increase in homology-directed repair frequencies when fused to Cas9/Cas12a in a tobacco mosaic virus-based transient assay in Nicotiana benthamiana.We achieved precise and scar-free insertion of several kilobases of DNA both in transient and stable transformation systems.In Arabidopsis thaliana,fusion of Cas9 to a herpes virus family exonuclease led to 10-fold higher frequencies of knock-ins in the first generation of transformants.In addition,we demonstrated stable and heritable knock-ins in wheat in 1%of the primary transformants.Taken together,our results open perspectives for the routine production of heritable knock-in and gene replacement events in plants.

A New X-Ray Tidal Disruption Event Candidate with Fast Variability

During a close encounter between a star and a supermassive black hole,the star can get disrupted by the black hole’s tidal forces,resulting in a tidal disruption event(TDE).The accretion of the star’s material onto the black hole produces strong emission in different wavelength regimes.Here we report the discovery with the Roentgen Satellite(ROSAT)of an X-ray-selected transient source in an optically non-active galaxy.At the location R.A.:13h31m57s.66 and decl.:-32°43’19".7 a sudden rise in X-ray luminosity by a factor of 8 within 8 days has been observed.Additionally,a very soft X-ray spectrum with a blackbody temperature kT=0.1 keV and a peak luminosity of at least 1×10^(43)erg s^(-1)suggests a TDE interpretation,and the observed properties are very similar to previously identified soft X-ray(ROSAT)TDEs.An optical spectrum taken of the galaxy at the position of RXJ133157.6324319.7 six years after the X-ray outburst does not show any emission lines as would be expected from a persistent active galactic nucleus.The redshift of the galaxy is determined to be 0.051 based on absorption lines.It is therefore likely a member of the galaxy cluster A3560.The rise in X-ray luminosity happens within 8 days and thus appears to be fast for such an event.No X-ray emission was detected 170 days before and 165 days after the event,and none was detected 25 yr later with the Neil Gehrels Swift Observatory.The change in X-ray luminosity is at least a factor of 40.

In the present investigation,we demonstrate the capabilities of the discontinuous Galerkin spectral element method for high order accuracy computation of gas dynamics.The internal flow field of a natural gas injector for bivalent combustion engines is investigated under its operating conditions.The simulations of the flow field and the aeroacoustic noise emissions were in a good agreement with the experimental data.We tested several shockcapturing techniques for the discontinuous Galerkin scheme.Based on the validated framework,we analyzed the development of the supersonic jets during different opening procedures of a compressed natural gas injector.The results suggest that a more gradual injector opening decreases the noise emission.

A machine-learned interatomic potential for silica and its relation to empirical models

Silica(SiO_(2))is an abundant material with a wide range of applications.Despite much progress,the atomistic modelling of the different forms of silica has remained a challenge.Here we show that by combining density-functional theory at the SCAN functional level with machine-learning-based interatomic potential fitting,a range of condensed phases of silica can be accurately described.We present a Gaussian approximation potential model that achieves high accuracy for the thermodynamic properties of the crystalline phases,and we compare its performance(and performance–cost trade-off)with that of multiple empirically fitted interatomic potentials for silica.We also include amorphous phases,assessing the ability of the potentials to describe structures of melt-quenched glassy silica,their energetic stability,and the high-pressure structural transition to a mainly sixfold-coordinated phase.We suggest that rather than standing on their own,machine-learned potentials for silica may be used in conjunction with suitable empirical models,each having a distinct role and complementing the other,by combining the advantages of the long simulation times afforded by empirical potentials and the near-quantum-mechanical accuracy of machine-learned potentials.This way,our work is expected to advance atomistic simulations of this key material and to benefit further computational studies in the field.

Role of conspecifics and personal experience on behavioral avoidance of contaminated flowers by bumblebees

Pollinators use multiple cues whilst foraging in eluding direct cues from flowers and in direct cues from other pollinators. The use of indirect social cues is common in social in sects, such as honeybees and bumblebees, where a social environment facilitates the ability to use such cues. Bumblebees use cues to forage on flowers according to previous foraging experiences. Flowers are an essential food source for pollinators but also pose a high risk of parasite infection through the shared use of flowers leading to parasite spillover. Nevertheless, bumblebees have evolved behavioral defense mechanisms to limit parasite infection by avoiding contaminated flowers. Mechanisms underlying the avoidance of contaminated flowers by bumblebees are poorly understood. Bumblebees were recorded having the choice to forage on non-contaminated flowers and flowers contaminated by a trypan osome gut parasite, Crithidia bombi. The use of different treatments with presence or absence of con specifics on both con taminated and non-contami nated flowers allowed to investigate the role of social visual cues on their pathogen avoidance behavior. Bumblebees are expected to use social visual cues to avoid contaminated flowers. Our study reveals that the presence of a con specific on flowers either con taminated or not does not help bumblebee foragers avoiding contaminated flowers. Nevertheless, bumblebees whereas gaining experie nee tend to avoid their con specific when placed on contami nated flower and copy it whe n on the non-contami nated flower. Our experime nt suggests a detrime ntal impact of floral see nt on disease avoidanee behavior.

Dissecting the Heat Stress Response in Chlamydomonas by Pharmaceutical and RNAi Approaches Reveals Conserved and Novel Aspects

To study how conserved fundamental concepts of the heat stress response （HSR） are in photosynthetic eukaryotes, we applied pharmaceutical and antisense/amiRNA approaches to the unicellular green alga Chlamydomonas reinhardtii. The Chlamydomonas HSR appears to be triggered by the accumulation of unfolded proteins, as it was induced at ambient temperatures by feeding cells with the arginine analog canavanine. The protein kinase inhibitor staurosporine strongly retarded the HSR, demonstrating the importance of phosphorylation during activation of the HSR also in Chlamydomonas. While the removal of extracellular calcium by the application of EGTA and BAPTA inhibited the HSR in moss and higher plants, only the addition of BAPTA, but not of EGTA, retarded the HSR and impaired thermotoler- ance in Chlamydomonas. The addition of cycloheximide, an inhibitor of cytosolic protein synthesis, abolished the attenu- ation of the HSR, indicating that protein synthesis is necessary to restore proteostasis. HSP90 inhibitors induced a stress response when added at ambient conditions and retarded attenuation of the HSR at elevated temperatures. In addition, we detected a direct physical interaction between cytosolic HSP90A/HSP70A and heat shock factor 1, but surprisingly this interaction persisted after the onset of stress. Finally, the expression of antisense constructs targeting chloroplast HSP70B resulted in a delay of the cell＇s entire HSR, thus suggesting the existence of a retrograde stress signaling cascade that is desensitized in HSP7OB-antisense strains.

Uncoupling protein 2 deficiency reduces proliferative capacity of murine pancreatic stellate cells

BACKGROund： Uncoupling protein 2 （UCP2） has been suggested to inhibit mitochondrial production of reactive oxygen species （ROS） by decreasing the mitochondrial membrane potential. Experimental acute pancreatitis is associated with increased UCP2 expression, whereas UCP2 deficiency retards regeneration of aged mice from acute pancreatitis. Here, we have addressed biological and molecular functions of UCP2 in pancreatic stellate cells （PSCs）, which are involved in pancreatic wound repair and fibrogenesis. METHODS： PSCs were isolated from 12 months old （aged） UCP2^-/- mice and animals of the wild-type （WT） strain C57BL/6. Proliferation and cell death were assessed by em- ploying trypan blue staining and a 5-bromo-2＇-deoxyuridine incorporation assay. Intracellular fat droplets were visualized by oil red O staining. Levels of mRNA were determined by RT-PCR, while protein expression was analyzed by immunoblotting and immunofluorescence analysis. Intracellular ROS levels were measured with 2＇,7＇-dichlorofluorescin diacetate. Expression of senescence-associated β-galactosidase （SA β-Gal） was used as a surrogate marker of cellular senescence. RESULTS： PSCs derived from UCP2^-/- mice proliferated at a lower rate than cells from WT mice. In agreement with this observation, the UCP2 inhibitor genipin displayed dose- dependent inhibitory effects on WT PSC growth. Interestingly, ROS levels in PSCs did not differ between the two strains, and PSCs derived from UCP2^-/- mice did not senesce faster than those from corresponding WT cells. PSCs from UCP2^-/- mice and WT animals were also indistinguishable with respect to the activation-dependent loss of intracellular fat droplets, expression of the activation marker α-smooth muscle actin, type I collagen and the autocrine/paracrine mediators interleukin-6 and transforming growth factor-I~ 1. CONCLUSIONS： A reduced proliferative capacity of PSC from aged UCP2^-/- mice may contribute to the retarded regeneration after acute pancreatitis. Apart from their slower growth, PSC of UCP2^-/-

Light Induced Inhibition of Protein Phosphatase Calcineurin

Light-induced switching of drug activity can be used to realize spatio-temporal control of biological activity in cells and in vivo.In our study of light-switchable derivatives of immunosuppressive drug cyclosporin A(CsA)in their inhibition of protein phosphatase calcineurin(CaN),we have discovered that 7-methyl-6-thioguanosine(MESG)inhibits CaN under light irradiation through a photo-crosslinking mechanism.The inhibition has shown selectivity over other enzymes such as phosphatase PP2A,protease chymotrypsin,and cyclophilin 18,the receptor protein for CsA.The discovery could pave the way to discover new covalent inhibitors for protein phosphtase,to design immunophilin-independent CaN inhibitors,and to design new light controlled inhibitors to modulate signal transduction.
