Ferroptosis is an iron-dependent regulatory cell necrosis induced by iron overload and lipid *** occurs when multiple redoxactive enzymes are ectopically expressed or show abnormal ***,the precise regulation of ferroptosis-related molecules is mediated across multiple levels,including transcriptional,posttranscriptional,translational,and epigenetic levels.N^(6)-methyladenosine(m^(6)A)is a highly evolutionarily conserved epigenetic modification in *** m^(6)A modification is commonly linked to tumor proliferation,progression,and therapy resistance because it is involved in RNA metabolic ***,accumulating evidence suggests that dysregulated ferroptosis caused by the m^(6)A modification drives tumor *** this review,we summarized the roles of m^(6)A regulators in ferroptosis-mediated malignant tumor progression and outlined the m^(6)A regulatory mechanism involved in ferroptosis *** also analyzed the potential value and application strategies of targeting m^(6)A/ferroptosis pathway in the clinical diagnosis and therapy of tumors.

Gear walk instability caused by brake-disc friction characteristics

A multi-body dynamic rigid-flexible coupling model of landing gear is established to study the gear walk instability caused by the friction characteristics of the brake *** validating the model with the experimental results,the influence of the landing gear structure and braking system parameters on gear walk is further *** the above factors,the slope of the graph for the friction coefficient of the brake disc and the relative velocity of brake stators and rotors is the most influential factor on gear walk *** trajectory analysis verifies that gear walk occurs when the coupling of multiple factors causes the system to exhibit an equivalent negative damping *** consider a more realistic braking case,a back propagation neural network method is employed to describe the nonlinear behavior of the friction coefficient of the brake *** the realistic nonlinear model of the friction coefficient,the maximum error in predicting the braking torque is less than 10%and the effect of the brake disc temperature on gear walk is *** results reveal that a more negative friction slope may contribute to a more severe unstable gear walk,and reducing the braking pressure is an effective approach to avoid gear walk,which provides help for future braking system design.

Transformation and mechanisms of climate wet/dry change on the northern Tibetan Plateau under global warming:A perspective from paleoclimatology

The northern Tibetan Plateau is a climatically sensitive zone influenced by monsoon and westerly *** summer,water vapor transport can reach Qinghai Lake and the eastern section of the Qilian Mountains;in winter,westerly winds mainly control the *** article compares the wet/dry changes in the region during the mid-Holocene(MH)warm period,the medieval climate anomaly(MCA),the current warm period(CWP),and the future warm period from the perspective of *** found that the MH warm period was mainly affected by the orbit-controlled East Asian summer monsoon,and the region showed warm and humid climate *** MCA was mainly controlled by solar radiation,and there was a warm and dry *** CWP and the future warm period are mainly controlled by the rise in temperature caused by the increase in greenhouse gases,and the climate is becoming more *** wet/dry patterns in the CWP and the future warm period in the next century on the northern Tibetan Plateau are similar to those in the *** warming will lead to the expansion of the westerly belt and a gradually humid *** future wet/dry changes will be more similar to the MH warm period.

Catalytic four-component radical carbonylation of unactivated alkenes has recently been recognized as a robust protocol for rapid construction of various structurally diverse carbonyl *** the significance of fluorine-containing groups,this reaction class has been extensively applied to assembly of a variety of perfluoroalkyl carboxylic acid derivatives by transition metal ***,we report a visible-light-driven radical relay 1,2-perfluoroalkylation aminocarbonylation of unactivated alkenes using CO gas as carbonyl source and 4CzIPN as organic photocatalyst.A wide range of alkenes and amines were well tolerated,providing the valuableβ-perfluoroalkylated amides with generally good yields and high chemoselectivity.

The VAMP-associated protein VAP27-1 plays a crucial role in plant resistance to ER stress by modulating ER–PM contact architecture in Arabidopsis

The endoplasmic reticulum(ER)and the plasma membrane(PM)form ER–PM contact sites(EPCSs)that allow the ER and PM to exchange materials and ***-induced disruption of protein folding triggers ER stress,and the cell initiates the unfolded protein response(UPR)to resist the ***,whether EPCSs play a role in ER stress in plants remains ***-ASSOCIATED MEMBRANE PROTEIN(VAMP)-ASSOCIATED PROTEIN 27-1(VAP27-1)functions in EPCS tethering and is encoded by a family of 10 genes(VAP27-1–10)in Arabidopsis ***,we used CRISPR-Cas9-mediated genome editing to obtain a homozygous vap27-1 vap27-3 vap27-4(vap27-1/3/4)triple mutant lacking three of the key VAP27 family members in *** vap27-1/3/4 mutant exhibits defects in ER–PM connectivity and EPCS architecture,as well as excessive UPR *** further showed that relocation of VAP27-1 to the PM mediates specific VAP27-1-related EPCS remodeling and expansion under ER ***,the spatiotemporal dynamics of VAP27-1 at the PM increase ER–PM connectivity and enhance Arabidopsis resistance to ER *** addition,we revealed an important role for intracellular calcium homeostasis in the regulation of UPR *** together,these results broaden our understanding of the molecular and cellular mechanisms of ER stress and UPR signaling in plants,providing additional clues for improving plant broad-spectrum resistance to different stresses.

Discovery and structure-activity relationship studies of novel tetrahydro-β-carboline derivatives as apoptosis initiators for treating bacterial infections

Developing and excavating new agrochemicals with highly active and safe is an important tactic for protecting crop health and food *** this paper,to discover the new bactericide candidates,we designed,prepared a new type of1,2,3,4-tetrahydro-β-carboline(THC)derivatives and evaluated the in vitro and in vivo bioactivities against the Xanthomonas oryzae ***(Xoo),Xanthomonas axonopodis ***(Xac),and Pseudomonas syringae ***(Psa).The in vitro bioassay results exhibited that most title molecules possessed good activity toward the three plant pathogenic bacteria,the compound A17 showed the most active against Xoo and Xac with EC50 values of 7.27 and 4.89 mg mL^(-1)respectively,and compound A8 exhibited the best inhibitory activity against Psa with EC50value of 4.87 mg mL^(-1).Pot experiments showed that compound A17 exhibited excellent in vivo antibacterial activities to manage rice bacterial leaf blight and citrus bacterial canker,with protective efficiencies of 52.67 and 79.79%at 200 mgmL^(-1),***,compound A8 showed good control efficiency(84.31%)against kiwifruit bacterial canker at 200 mg mL^(-1).Antibacterial mechanism suggested that these compounds could interfere with the balance of the redox system,damage the cell membrane,and induce the apoptosis of Xoo *** together,our study revealed that tetrahydro-β-carboline derivatives could be a promising candidate model for novel broadspectrum bactericides.

Although CRISPR/Cas9 has been widely used in insect gene editing,the need for the microinjection of preblastoderm embryos can preclude the technique being used in insect species with eggs that are small,have hard shells,and/or are difficult to collect and maintain outside of their normal *** is the case with Sogatella furcifera,the white-backed planthopper(WBPH),a significant pest of Oryza sativa(rice)that oviposits inside rice *** extraction from the stem runs the risk of mechanical damage and hatching is heavily influenced by the micro-environment of the rice *** bypass these issues,we targeted embryos prior to oviposition via direct parental(DIPA)-CRISPR,in which Cas9 and single-guide RNAs(sgRNAs)for the WBPH eye pigment gene tryptophan 2,3-dioxygenase were injected into the hemocoel of adult *** at varying numbers of days posteclosion were evaluated to determine at what stage their oocyte might be most capable of taking up the gene-editing *** evaluation of the offspring indicated that the highest G0 gene-edited efficacy(56.7%)occurred in females injected 2 d posteclosion,and that those mutations were heritably transmitted to the G1 *** study demonstrates the potential utility of DIPA-CRISPR for future gene-editing studies in non-model insect species and can facilitate the development of novel pest management applications.

Two-dimensional porous carbon derived from pollen with enlarged interlayer distance enhanced electrocatalytic oxidation of n-valeraldehyde to octane

Electrocatalytic n-valeraldehyde oxidation reaction was an inexpensive and eco-friendly method to control n-valeraldehyde contamination and produce high value-added ***,low-cost and readily available electrocatalysts with high current efficiency were urgently ***,two-dimensional porous carbon derived from pollen with enlarged interlayer distance was built by alkali activation method,applying in electrocatalytic n-valeraldehyde oxidation *** enlarged interlayer distance was verified by X-ray diffraction(XRD)and high-angle annular dark-field scanning transmission electron microscope(HAADF-STEM).Electrocatalytic experiments consequences showed activated biomass derived carbon possessed a higher electrocatalytic activity and octane selectivity than unactivated *** tests and in situ infrared experiments demonstrated that enlarged interlayer distance was positively correlated with specific surface area of catalysts,large specific surface area provided abundant absorption sites,facilitated the adsorption for n-valeraldehyde,and further promoted the transformation of n-valeraldehyde to *** work also provides a new avenue for creating high-performance electrocatalysts in terms of lattice engineering.

Organic conjugated polymers have received extensive attention due to their unique electronic ***,there have been relatively few reports on the dark photocatalytic reactions utilizing organic conjugated ***,we report the successful synthesis of an organic conjugated polymer based on poly(heptazine imide)nanocrystals(CNNCs)for H_(2)O_(2)evolution and biomedical applications using a simple salt molten method and sonication-centrifugation *** results show that these colloid CNNCs have the characteristics of photogenerated electrons accumulation and realize dark photocatalysis with high reducibility under visible light ***,these accumulating photogenerated electrons can reduce O_(2)in darkness to produce H_(2)O_(2).In addition,cytotoxicity tests were conducted and it was found that H_(2)O_(2)produced under dark conditions could oxidize L-arginine(L-Arg)to NO,which effectively killed tumors in the *** work provides an important strategy to construct organic conjugated semiconductor nanocrystals and applying them to future energy and biomedical fields.
